The orphan G protein-coupled receptor (GPCR) GPR124/tumor endothelial marker 5 is highly expressed in central nervous system (CNS) endothelium. Here, we show that complete null or endothelial-specific GPR124 deletion resulted in embryonic lethality from CNS-specific angiogenesis arrest in forebrain and neural tube. Conversely, GPR124 overexpression throughout all adult vascular beds produced CNS-specific hyperproliferative vascular malformations. In vivo, GPR124 functioned cell-autonomously in endothelium to regulate sprouting, migration, and developmental expression of the blood-brain barrier marker Glut1, whereas in vitro, GPR124 mediated Cdc42-dependent directional migration to forebrain-derived, vascular endothelial growth factor-independent cues. Our results demonstrate CNS-specific angiogenesis regulation by an endothelial receptor and illuminate functions of the poorly understood adhesion GPCR subfamily. Mixtures of Eucalyptus globulus Labill. and Acacia mearnsii de Wildeman are twice as productive as E. globulus monocultures growing on the same site in East Gippsland, Victoria, Australia, possibly because of increased nitrogen (N) availability owing to N(2) fixation by A. mearnsii. To investigate whether N(2) fixation by A. mearnsii could account for the mixed-species growth responses, we assessed N(2) fixation by the accretion method and the (15)N natural abundance method. Nitrogen gained by E. globulus and A. mearnsii mixtures and monocultures was calculated by the accretion method with plant and soil samples collected 10 years after plantation establishment. Nitrogen in biomass and soil confirmed that A. mearnsii influenced N dynamics. Assuming that the differences in soil, forest floor litter and biomass N of plots containing A. mearnsii compared with E. globulus monocultures were due to N(2) fixation, the 10-year annual mean rates of N(2) fixation were 38 and 86 kg ha(-1) year(-1) in 1:1 mixtures and A. mearnsii monocultures, respectively. Nitrogen fixation by A. mearnsii could not be quantified on the basis of the natural abundance of (15)N because such factors as mycorrhization type and fractionation of N isotopes during N cycling within the plant confounded the effect of the N source on the N isotopic signature of plants. This study shows that A. mearnsii fixed significant quantities of N(2) when mixed with E. globulus. A decline in delta(15)N values of E. globulus and A. mearnsii with time, from 2 to 10 years, is further evidence that N(2) was fixed and cycled through the stands. The increased aboveground biomass production of E. globulus trees in mixtures when compared with monocultures can be attributed to increases in N availability. 相似文献

The role of European forests and forest management in the carbon balance has received much attention in research recently.
This was particularly motivated by the recognition of forest management as one possible measure countries may adopt in the
framework of the Kyoto Protocol to reduce the concentration of greenhouse gases in the earth’s atmosphere. The main method
to assess carbon budget in forests is based on traditional forest inventories. This method requires the conversion of measured
stem volume to carbon pools. This conversion has been identified as a large source of uncertainty in past assessments. Over
the last 5 years, intensive research efforts have resulted in significant advances in the reliability of forest inventory
based carbon budgets. In parallel, the impact of forest management on the carbon balance of forest ecosystems has been investigated
and the carbon mitigation potential of these activities has been analysed. This paper reviews the progress that was made in
these two fields of research with a particular focus on European forests.

Forest management activities may help reduce global net CO2 concentrations by capturing and storing atmospheric CO2. Research related to carbon sequestration potential of plantations in North America has focused predominantly on conifers, with relatively little emphasis thus far on temperate deciduous forest tree species. American chestnut (Castanea dentata (Marsh.) Borkh.), a former dominant tree species in eastern North America until its demise associated with the introduced chestnut blight (Cryphonectria parasitica (Murr.) Barr.), is a temperate deciduous species that holds promise for future carbon sequestration programs with expected availability of blight-resistant backcross hybrids. We quantified aboveground biomass and bole carbon of American chestnut interplanted with black walnut (Juglans nirga L.) and northern red oak (Quercus rubra L.) across four blight-free experimental sites varying in site quality and/or age (8, 8, 12, and 19 years) isolated from the native American chestnut range in the Coulee Region of southwestern Wisconsin, USA. American chestnut exhibited more rapid growth and greater aboveground biomass and bole carbon than either of the other interplanted species. Aboveground biomass ranged from 46.9, 60.7, 55.0, and 179.9 Mg ha−1 for the 8-, 8-, 12-, and 19-year-old sites, respectively, while bole carbon content ranged from 13.6, 18.6, 14.1, and 60.1 Mg ha−1 for the 8-, 8-, 12-, and 19-year-old sites, respectively. Cross-referencing our data to studies conducted within this same physiographic region using other important forestry species (i.e., Populus tremuloides Michx., Pinus resinosa Ait., and Pinus strobus L.) showed that American chestnut compared favorably in growth and carbon uptake. Incorporating American chestnut into carbon sequestration plantations provides additional ecological and economic benefits associated with consistent production of quality nuts for wildlife, valuable timber, and contribution toward species restoration. Our data lend support to building evidence demonstrating rapid and sustained growth of American chestnut and the potential role of plantation-grown American chestnut in helping to mitigate climate change through carbon sequestration. 相似文献

Objective To study the analgesic potency of the α2‐agonist romifidine in the horse using both an electrical current and a mechanical pressure model for nociceptive threshold testing. In addition, a comparison was made with doses of detomidine and xylazine that produce equivalent degrees of sedation. Study design Randomized, placebo‐controlled, blinded cross‐over study. Animals Six adult Swiss warmblood horses, one mare and five geldings, weighing from 530 to 650 kg and aged 6–15 years. Methods Nociceptive thresholds were measured using an electrical stimulus applied to the coronary band and using a pneumatically operated pin pressing on the cannon bone. Measurements were made immediately before and every 15 minutes for 2 hours after IV injection of the test substances. Lifting of the foot indicated the test end point. Results The three α2‐agonists caused a temporary increase in nociceptive thresholds with a maximal effect within 15 minutes and a return to baseline levels within 1 hour. Using electrical current testing nociceptive thresholds were significantly different from placebo (mean ± SD) for detomidine at 15 minutes (from control 5.8 ± 0.9 to 23.3 ± 3.9 mA, p = 0.0066) and 30 minutes (from control 6.6 ± 1.1 to 18.8 ± 3.3 mA, p = 0.0091). The difference was significant for romifidine at 15 minutes only (from control 5.8 ± 0.9 to 18.7 ± 3.8 mA, p = 0.0066). With mechanical pressure testing nociceptive thresholds were significantly different from control for detomidine at 15 minutes (from 3.2 ± 0.2 to 6.2 ± 0.5 N, p = 0.00076) and 30 minutes (from 3.2 ± 0.7 to 5.7 ± 0.8 N, p = 0.0167). The difference was significant for xylazine at 15 minutes (from control 3.2 ± 0.2 to 5.6 ± 0.7 N, p = 0.0079). At 15 minutes the order of magnitude of the measured antinociceptive effect was significantly different between the two pain tests for both romifidine and detomidine, but not for xylazine. For romifidine, the increase of mean thresholds compared to placebo was 4.0 ± 1.3 times placebo levels with the electrical current test compared to 1.3 ± 0.3 times for the mechanical pressure test (p = 0.037). For detomidine, the increase of mean thresholds compared to placebo was 5.4 ± 1.7 times control levels with the electrical current test compared to 2.0 ± 0.2 times for the mechanical pressure test (p = 0.040). This represents a 2.7 (romifidine) and 3.4 times (detomidine) greater increase in thresholds using electrical current testing compared to the use of mechanical pressure testing. Conclusion and clinical relevance This study demonstrates the analgesic potential of α2‐agonists in the horse for somatic pain and that they can have quantitatively different antinociceptive effects according to the antinociceptive test used. 相似文献

Twenty-eight research dogs were enrolled to determine the prevalence of salmonellae shedding after consumption of 1 Salmonella-contaminated commercial raw food diet meal. Sixteen dogs were exposed to Salmonella-contaminated commercial raw food diets and 12 to Salmonella-free commercial raw food diets. Seven of the exposed dogs shed salmonellae 1-7 days after consumption of Salmonella-contaminated raw food diets. None of the dogs fed Salmonella-free diets shed salmonellae. No clinical signs were observed in either group. Five of the 7 dogs shed the same serotypes as those recovered from food samples used for feeding. Results showed the same serotypes and antimicrobial resistance pattern in 2 of the 7 shedders. Dogs fed Salmonella-contaminated raw food diets can shed salmonellae and may, therefore, be a source of environmental contamination potentially leading to human or animal illness. 相似文献
