Wildlife, domesticated animals and humans are exposed daily to myriad chemicals present in our environment. The risk posed by these chemicals to one species is often determined by extrapolation from data gathered from another species. Several extensive studies have examined the capability of the liver to biotransform xenobiotics in animals commonly used in toxicity testing and in livestock. The present paper is a compilation of these data into a single source to permit comprehensive examination of inter-species variation in rates of hepatic biotransformation. Several substrates were studied for each enzyme system, including cytochrome P-450-dependent monooxygenases, epoxide hydrolases, UDP-glucuronosyltransferases, N-acetyltransferases, glutathione S-transferases and sulfotransferases. The numerous differences in substrate specificity for an individual enzymatic pathway reflect the apparent multiplicity of these enzymes in all 11 species studied. Several hundred- to several thousand-fold differences between species in enzymatic activities for certain substrates under well-defined conditions emphasize the need for caution and the risk of error in extrapolation of xenobiotic metabolism from one species to another. In spite of these uncertainties, knowledge of the rate of biotransformation may help us predict the fate of new chemicals in various species.  相似文献

Lethargic Crab Disease (LCD) caused extensive epizootic mortality of the mangrove land crab Ucides cordatus (Brachyura: Ocypodidae) along the Brazilian coast, mainly in the Northeastern region. The disease was named after the symptoms of slow movement of infected crabs. Causative agents were suspected to be two black yeast-like fungi of the family Herpotrichiellaceae (ascomycete order Chaetothyriales), judged by infected tissue biopsies from moribund U. cordatus. The aim of the present study is to prove that two species are involved in the disease: the recently described black yeast Exophiala cancerae, but also a less virulent, hitherto undescribed fonsecaea-like species, introduced here as the novel species Fonsecaea brasiliensis. Strains were identified by ITS rDNA sequencing, and species borderlines were established by multilocus sequencing and AFLP analysis. Fonsecaea brasiliensis proved to be closely related to the pathogenic species Cladophialophora devriesii which originally was isolated from a systemic infection in a human patient. The virulence of F. brasiliensis is lower than that of E. cancerae, as established by artificial inoculation of mangrove crabs.  相似文献

Species-specific agonist binding regions of nicotinic acetylcholine receptors (nAChR) were examined. Imidacloprid and physostigmine (Phy) selectively activated insect nAChR composed of Drosophila second alpha-like subunit (SAD) and chick β2, in contrast to rat α7 nAChR. The Phy-activated currents were α-bungarotoxin (α-BGT) sensitive, suggesting activation at the agonist binding loop C. Several SAD-α7 chimeras were constructed, by switching agonist binding regions, and expressed in oocytes. Though none of the chimeras was activated by a range of nicotinic agonists, [125I]α-BGT binding revealed homomeric assembly of all chimeric cDNAs. Phy differentially displaced [125I]α-BGT from the nAChR chimeras, suggesting that the β subunit is not involved in Phy binding, and that Phy targets the insect agonist binding loop C.  相似文献

An interactive relationship between vitiviruses and grapevine leafroll viruses was characterized in grapevine. Grapevine viruses A and B (GVA and GVB) were found more frequently in the presence of co-infecting Grapevine leafroll associated viruses (GLRaV-1, ?2 or ?3) than in their absence. The titers of the vitiviruses in co-infection with leafroll viruses were found to be higher than were their titers in the absence of leafroll virus infection. The occurrence of vitivirus-associated stem-pitting symptoms was correlated with leafroll virus co-infection. Specific pairing associations on the species level were found between different viti- and leafroll virus species: GVB was associated preferentially with GLRaV-2; GVA was associated preferentially with GLRaV-1 and GLRaV-3. In contrast to the increase in vitivirus titer seen with leafroll virus co-infection, the incidence and titer of grapevine leafroll virus appeared to be unaltered by vitivirus co-infection. The potential for a synergistic enhancement of grapevine disease in co-infected vines is discussed.  相似文献

Network analysis of correlations between abundances of metabolites across tomato genotypes can help in unraveling the biological basis of organoleptic variation in tomato. We illustrate how to construct and interpret simple correlations networks using metabolic data collected on a diverse set of tomato genotypes. Various types of correlations are calculated and displayed in the form of networks. Interpretations on the basis of network analyses are compared to interpretations following principal components analysis.  相似文献

The effects of selenium supplementation on mastitis parameters in milk and on glutathione peroxidase (GPx) levels in blood were evaluated. Fifty-five Estonian dairy cows were allocated to selenium-supplemented (n=39) and nonsupplemented (n=16) groups. The supplemented group received 0.2 ppm organic selenium in the form of selenium yeast in their diet daily for 8 weeks. The nonsupplemented cows received their standard diet with no selenium supplementation. Mastitis parameters (i.e., bacteriologic findings and somatic cell count, N-acetyl-beta-D-glucosaminidase, and bovine serum albumin concentration) and GPx levels were monitored. The increase in the activity of GPx was significantly (P<.001) greater in selenium-supplemented cows than in nonsupplemented ones. Milk samples from each quarter were examined before and 8 weeks after initiation of the study. The proportion of quarters still pathogen-free after 8 weeks was significantly (P<.01; odds ratio [OR]=9.7) higher in selenium-supplemented cows than in nonsupplemented cows. However, when quarters milk-tested as pathogen-infected at the start of the study were reexamined after 8 weeks, there was no significant (P=.14; OR 3.6) difference in the proportion of pathogen-free quarters between selenium-supplemented cows and nonsupplemented cows. Differential positive rate (Youden's index) revealed that individual quarters were more prone to be infected by pathogens when the blood GPx activities in cows were below the cutoff value of 3.3 microkat/g hemoglobin than when GPx activity was above this value. It was concluded that selenium supplementation in cows with low GPx activity seems to support udder defense mechanisms that favor reduction of the incidence of new mastitis cases.  相似文献

Bovine udders, together with the attached section of the ventral abdominal wall, as well as the bony pelvic floor were studied. The parts of the suspensory apparatus and attachment to the body were investigated. The location of the medial and lateral laminae served as the basis for classifying them into abdominal and pelvic parts. The udder base is attached to the ventral abdominal wall by means of the dorsal connective tissue plate. The abdominal part of the medial lamina and the pelvic part of the lateral lamina are the functionally more important bands. Sixty per cent of the udder base is attached to the ventral abdominal wall and 40% to the pelvic floor by the symphysial tendon. The symphysial tendon is attached to the ventral pubic tubercle and the symphysial eminence as well as to the symphysial crest, which develops by the fourth year. In order to study the osseous changes, bovine pelvises from the Middle Ages, the beginning of the 20th century, and the present time were compared. Increased udder mass has brought about changes in the structures of the suspensory apparatus of the udder and in the structure of the pelvis.  相似文献
