The effects of crude extracts and an isolated compound from the leaves of milkweed, Pergularia daemia (Forssk) Choiv., on the antifeedant activity against two important lepidopteran pests, Helicoverpa armigera (Hub.) and Spodoptera litura (F.), were studied. Maximum antifeedant activity was recorded in ethyl acetate crude extract against H. armigera (70.3%) and S. litura (71.82%) at 1% concentration. Ethyl acetate crude extract was further subjected to column chromatography, which was performed using hexane as initial solvent and then by increasing the polar strength using ethyl acetate. Fractions collected at hexane and ethyl acetate (80:20) yielded 6-(4,7-hydroxy-heptyl) quinone, a novel compound which showed significant antifeedant activity against H. armigera (80.22% at 2000 ppm) and S. litura (68.31% at 2000 ppm).  相似文献

1,3-disubstituted aliphatic and aromatic symmetrical and unsymmetrical thioureas were synthesised by novel routes and the antifungal activities of these compounds against two plant pathogens, Pyricularia oryzae and Drechslera oryzae were studied in Czapek-Dox medium at four different concentrations using acetone as the control and the structure-activity relationship among the substituted thioureas is reported. © 1998 SCI.  相似文献

Pulses, the most important source of high-quality dietary proteins are prone to serious damage due to insect pests, of which 20–25% damage was inflicted by stored product bruchid pest, Callosobruchus maculatus (F.). An alternative approach to control these insect pests apart from fumigation is the use of primary metabolites involved in plant defense mechanism. In the present study, one of such molecule, an insect anti-metabolic protein, arcelin was screened from nine different wild and cultivated varieties of pulses. Insect feeding bioassay with C. maculatus revealed that seven of these seed varieties were not infested by the bruchid insect pest. Additionally, the presence of arcelin polypeptide subunits (27–42 kDa) was demonstrated in the seeds of Phaseolus limensis L. and Phaseolus lunatus L. whereas, an insignificant amount was observed in Lablab purpureus L. Besides, seed kernel extract of P. lunatus registered a highest HA activity against native human-A erythrocytes and this activity was increased to several fold when the erythrocytes were treated with proteolytic enzymes revealing the characteristic feature of arcelin. Similarly, HA activity was observed when rat and mouse erythrocyte was treated with pronase, whereas, no titer value was observed upon treatment with its native erythrocytes. Of the 24 carbohydrates tested, only acetylated amino sugar, N-acetyl-d-galactosamine inhibited the agglutinating activity of crude seed extract against human-A RBC. Among the three glycoproteins tested, none were found potential to competitively inhibit the agglutination activity against human-A erythrocytes. Overall, this study reveals the presence of arcelin, an anti-metabolic insecticidal protein in seeds of P. lunatus.  相似文献

Utilization of shrimp head waste from the processing industry offers a solution to environmental pollution and also produces valuable bioactive peptides. The present study was aimed to optimize the alcalase assisted extraction conditions by response surface methodology (RSM) to obtain protein hydrolysates from shrimp (Metapenaeus dobsoni) head waste with maximum antioxidant activity. Based on the models derived by RSM, the optimized conditions for maximum degree of hydrolysis (DH) were pH 8.2, temperature of 45.4°C, and enzyme/substrate (E/S) of 1.8%. Meanwhile, the optimized conditions for maximum antioxidant activity of protein hydrolysates as measured by 2, 2-diphenyl-1-picrylhydrozyl (DPPH) and ferric reducing antioxidant power (FRAP) assays were pH 8.2 and 8.1, temperatures of 49.7 and 59.4°C, and E/S of 2.1 and 2.7%, respectively. The maximum predicted values for DH, DPPH, and FRAP were 42.44%, 39.64% inhibition, and 8.3 μM Fe (II)/g wet wt. of sample, respectively. The extraction of protein hydrolysates at optimal conditions derived by these models resulted in DH, DPPH, and FRAPs of 40.31%, 38.93%, and 8.21 μM Fe (II)/g of sample, respectively, demonstrating the fitness of the models. The present study demonstrates the potential of Metapenaeus dobsoni head waste as a source of protein hydrolysates with prospective antioxidant activity.  相似文献

Muskmelons, both cantaloupe (Cucumis melo Reticulatus Group) and orange-fleshed honeydew (C. melo Inodorus Group), a cross between orange-fleshed cantaloupe and green-fleshed honeydew, are excellent sources of β-carotene. Although β-carotene from melon is an important dietary antioxidant and precursor of vitamin A, its bioaccessibility/bioavailability is unknown. We compared β-carotene concentrations from previously frozen orange-fleshed honeydew and cantaloupe melons grown under the same glasshouse conditions, and from freshly harvested field-grown, orange-fleshed honeydew melon to determine β-carotene bioaccessibility/bioavailability, concentrations of novel β-apocarotenals, and chromoplast structure of orange-fleshed honeydew melon. β-Carotene and β-apocarotenal concentrations were determined by HPLC and/or HPLC-MS, β-carotene bioaccessibility/bioavailability was determined by in vitro digestion and Caco-2 cell uptake, and chromoplast structure was determined by electron microscopy. The average β-carotene concentrations (μg/g dry weight) for the orange-fleshed honeydew and cantaloupe were 242.8 and 176.3 respectively. The average dry weights per gram of wet weight of orange-fleshed honeydew and cantaloupe were 0.094 g and 0.071 g, respectively. The bioaccessibility of field-grown orange-fleshed honeydew melons was determined to be 3.2 ± 0.3%, bioavailability in Caco-2 cells was about 11%, and chromoplast structure from orange-fleshed honeydew melons was globular (as opposed to crystalline) in nature. We detected β-apo-8'-, β-apo-10', β-apo-12'-, and β-apo-14'-carotenals and β-apo-13-carotenone in orange-fleshed melons (at a level of 1-2% of total β-carotene). Orange-fleshed honeydew melon fruit had higher amounts of β-carotene than cantaloupe. The bioaccessibility/bioavailability of β-carotene from orange-fleshed melons was comparable to that from carrot (Daucus carota).  相似文献

In this study, Enterobacter cloacae bacterium was isolated from curd and its antibacterial potential against the pathogen Plesiomonas shigelloides was evaluated using the freshwater ornamental fish Kenyi cichlid (Maylandia lombardoi). Among the bacterial isolates, E. cloacae exhibited tolerance to acidic pH 2 and demonstrated the highest antibacterial activity against P. shigelloides in various in vitro assays. Dietary supplementation of E. cloacae with prebiotic supplement 2% mannan oligosaccharide improved the growth performance and reduced the toxic metabolites such as nitrite in culture tank water of Kenyi cichlid. In vivo application of E. cloacae with mannan oligosaccharide significantly elevated (p?<?0.05) white blood cell counts (88.47?±?2.15 103 mm?3) and respiratory burst activity (0.243?±?0.007) of Kenyi cichlid when challenged with P. shigelloides. Intestinal morphology of Kenyi cichlid fish treated with probiotic and prebiotic combinations showed improved intestinal architectures. The present findings confirm that the isolated bacterium E. cloacae is a potential probiotic and can be used effectively to prevent infection of P. shigelloides in freshwater ornamental fish culture.  相似文献

The calli induction from different Sunflower necrosis virus (SNV)-infected explants of sunflower was initiated on Murashige & Skoog medium amended with benzyladenine at 1 mg l -1 and indole acetic acid at 0.5 mg l -1. The virus was purified from the induced calli by sucrose density gradient centrifugation and the viral particles were confirmed by transmission electron microscopy. The SNV infection in uninfected calli was established by a soak prick method and the viral development was confirmed by a plant infectivity test. Serological analyses such as Ouchterlony double immnuodiffusion test, enzyme-linked immunosorbent assay and Western blot using anti-SNV antibodies yielded positive reactions with the purified SNV. These investigations have demonstrated that the SNV could be cultured in calli and maintained in vitro in pure form for a long period of time.  相似文献

The diversity among 269 rhizobia isolated from naturally occurring root nodules of soybean collected from two different agro-ecological regions of India, based on RFLP and sequences of the intergenic spacer (IGS) between the 16S and 23S rRNA genes, growth rate, and indole acetic acid production, revealed their significant, site-dependent genomic diversity. Among these bacteria, nine IGS genotypes were identified with two endonucleases. They were distributed into five divergent lineages by sequence analysis of each IGS representative strain, i.e., (1) comprising IGS genotypes I, II, III, and reference Bradyrhizobium yuanmingense; (2) with genotype IV and strains of unclassified bradyrhizobia genomic species; (3) including genotypes V, VI, and Bradyrhizobium liaoningense; (4) with IGS genotype VII and Bradyrhizobium elkanii strains; and (5) comprising IGS genotypes VIII, IX, and different Ensifer genus bacteria. Host-specificity test revealed that all rhizobia-nodulated soybean and cowpea and only part of them formed nodules on Arachis hypogeae and Cajanus cajan. The great diversity of soybean nodulators observed in this study emphasises that Indian soil is an important reservoir of nitrogen-fixing rhizobia.  相似文献

A partially purified protein fraction was isolated from seed flour of the Indian wild bean, Lablab purpureus, by ion exchange and size-exclusion chromatographies. Partially purified L. purpureus proteins had hemagglutination and glycoslyation properties similar to those of lectins or lectin-like proteins from other pulses. Data obtained from two-dimensional gel electrophoresis, MALDI-TOF, and MALDI-TOF/TOF and N-terminal protein sequencing of the isolated polypeptides from L. purpureus demonstrated that the extract contained proteins similar to isoforms of arcelins 3 and 4 and pathogenesis-related protein 1 (PvPR1) of Phaseolus vulgaris. L. purpureus proteins were resistant to degradation by the commercial enzymes trypsin and chymotrypsin and were moderately resistant to pepsin, but were readily hydrolyzed to smaller peptides by papain. Insect feeding bioassays of the extract with the storage pests Rhyzopertha dominica and Oryzaephilus surinamensis, internal and external feeders of grain, respectively, demonstrated that L. purpureus proteins at 2% in the diet resulted in retarded development. However, a 5% dose of the L. purpureus fraction resulted in complete mortality of all larvae in both species. This study has demonstrated that proteins in the partially purified L. purpureus extract have the potential to control storage pests in cereals transformed with L. purpureus defense-related genes, but the need for more studies regarding efficacy and safety is discussed.  相似文献
