The commercialization of animal feeds infected by prions proved to be the main cause of transmission of bovine spongiform encephalopathy (BSE). Therefore, feed bans were enforced, initially for ruminant feeds, and later for all feeds for farmed animals. The development and validation of analytical methods for the species-specific detection of animal proteins in animal feed has been indicated in the TSE (Transmissible Spongiform Encephalopathies) Roadmap (European Commission. Methods based on polymerase chain reaction (PCR) seem to be a promising solution for this aim. The main objective of this study was to determine the applicability of four different real-time PCR methods, developed by three National expert laboratories from the European Union (EU), for the detection and identification of cattle or ruminant species in typical compound feeds, fortified with meat and bone meals (MBM) from different animal species at different concentration levels. The MBM samples utilized in this study have been treated using the sterilization condition mandatory within the European Union (steam pressure sterilization at 133 degrees C, 3 bar, and 20 min), which is an additional challenge to the PCR methods evaluated in this study. The results indicate that the three labs applying their PCR methods were able to detect 0.1% of cattle MBM, either alone or in mixtures with different materials such as fishmeal, which demonstrates the improvement made by this technique, especially when compared with results from former interlaboratory studies.  相似文献

Summary One of the most promising control options against the parasitic weed Striga hermonthica is the use of crop varieties that combine resistance with high levels of tolerance. The aim of this study was to clarify the relation between Striga infestation level, Striga infection level and relative yield loss of sorghum and to use this insight for exploring the options for a proper screening procedure for tolerance. In three pot experiments, conducted in Mali (2003) and The Netherlands (2003, 2004), four sorghum genotypes were exposed to a range of Striga infestation levels, ranging from 0.0625 to 16 seeds cm−3. Observations included regular Striga emergence counts and sorghum grain yield at maturity. There were significant genotype, infestation and genotype × infestation effects on sorghum yield. The relation between infestation level and infection level was density dependent. Furthermore, the relation between Striga infection level and relative yield loss was non-linear, though for the most resistant genotype Framida only the linear part of the relation was obtained, as even at high infestation levels only moderate infection levels were achieved. The results suggest that for resistant genotypes, tolerance can best be quantified as a reduced relative yield loss per aboveground Striga plant, whereas for less resistant genotypes the maximum relative yield loss can best be used. Whether both expressions of tolerance are interrelated could not be resolved. Complications of screening for tolerance under field conditions are discussed.  相似文献

In this study the anti-inflammatory properties of zilpaterol, a beta2-adrenergic receptor (AR) agonist specifically developed as a growth promoter in cattle were investigated. Although zilpaterol has a different structure compared with the beta2-AR agonists known to date, it was noted that it was able to bind to both the beta2-AR (Ki = 1.1 x 10(-6)) and the beta1-AR (Ki = 1.0 x 10(-5)). Using lipopolysaccharide (LPS)-exposed U937 macrophages, the production of cyclic adenosine-3',5'-cyclic monophosphate (cAMP) and tumor necrosis factor alpha (TNF-alpha) were investigated. Zilpaterol inhibited TNF-alpha release and induced intracellular cAMP levels in a dose-dependent manner. The inhibition of TNF-alpha release and induction of cAMP production was mainly mediated via the beta2-AR, as indicated by addition of beta1- and beta2-specific antagonists. The effects of zilpaterol were investigated in LPS-treated male Wistar rats after pretreatment with zilpaterol. Zilpaterol dosed at 500 microg/kg body weight reduced the TNF-alpha plasma levels. In conclusion, zilpaterol is a beta2-adrenergic agonist and an inhibitor of TNF-alpha production induced by LPS both in vivo and in vitro.  相似文献

Root drench application of Trichoderma atroviride isolates R32, R33, R40 and R84 promoted the growth of potted radiata pine seedlings. After 6 weeks, seedlings treated with R33 and R84 had thicker stems and greater stem and root biomass (p < 0.05) than untreated controls. Treatment with R32 increased seedling root biomass whilst R40 increased stem diameter. None of the isolates affected seedling height. One isolate, R33, induced systemic resistance to stem inoculation with Diplodia pinea and reduced dieback incidence by 20% compared with untreated controls. To our knowledge, this is the first report of systemic induced resistance by Trichoderma in a pine species. Furthermore, seedlings that were treated with R33 (root drench) plus foliar application of methyl jasmonate (MeJA) expressed elevated peroxidase activity in their stems 2 weeks later, compared with seedlings treated only with MeJA. Because R33 itself did not affect peroxidase activity, this may be indicative of treatment synergy or defence potentiation by R33. Curiously, R33 + MeJA induced terpenoids but suppressed phenylalanine ammonia‐lyase activity suggesting possible trade‐offs between phenolic and terpenoid defence pathways in the treated seedlings.  相似文献

Weeds are a major constraint to rice (Oryza spp.) production in West Africa. Superior weed competitive rice genotypes may reduce weed pressure and improve rice productivity. Two upland and two lowland experiments were conducted in southern Benin to examine genotypic variations in weed-suppressive ability and grain yield under weedy conditions, and to identify plant characteristics that could be used as selection criteria for improved weed competitiveness. A total of 19 genotypes, including Oryza sativa and Oryza glaberrima genotypes and interspecific hybrids developed from crossing O. sativa and O. glaberrima, were grown under weed-free and weedy conditions in an upland with supplemental irrigation and in a flooded lowland. In weedy plots, hand weeding was done once or not at all. Mean relative yield loss across all genotypes due to weed competition ranged from almost 0% to 61%. Large genotypic variations in weed biomass and grain yield under weedy conditions were found. Visual growth vigor at 42 and 63 days after sowing (DAS) under weed-free conditions significantly correlated with weed biomass at maturity in both upland and lowland experiments (R2 = 0.26–0.48). Where weed pressure was low to moderate, with mean relative yield loss less than 23%, the multiple regression models using grain yield and plant height at maturity or only grain yield measured under weed-free conditions as independent variables could explain 66–88% of the genotypic variation in grain yield under weedy conditions. At higher weed pressure (mean relative yield loss: 61%), as observed in one of the upland experiments, biomass accumulation of rice at 42 days after sowing was associated with higher grain yield under weedy conditions. Biomass accumulation also significantly correlated with visual growth vigor at the same sampling dates. Therefore, we conclude that grain yield, plant height at maturity and visual growth vigor at 42–63 DAS under weed-free conditions appear to be useful selection criteria for developing superior weed competitive rice genotypes.  相似文献

Weeds and granivorous birds are the two major biotic constraints to rice production in sub‐Saharan Africa. Anecdotal evidence advances the hypothesis that weedy fields attract granivorous birds to the extent that the joint weed × bird impact on crops is synergistic. We develop a theoretical model, based on experimental data, of the interaction between weed and bird damage to formally test this hypothesis. We observe that the response of bird‐inflicted yield loss to weeds is unimodal and inverse‐U‐shaped. Bird‐inflicted yield loss increases from 44% in weed‐free conditions to a maximum of 55% at a critical weed infestation level of 20 g dry weight m?2. We conclude that the 11% increase in yield loss can be attributed to the synergistic interaction between weed and bird damage in rice. Our theory provides new insights into the analysis of the interaction between weeds and pests in general and supports a revision of currently accepted recommendations for weed management in rice. In areas where birds constitute an important production risk, it would be beneficial for rice farmers to remove those weeds that are tall enough to attract birds, even beyond the critical weed period.  相似文献

Striga hermonthica (Del.) Benth. is a parasitic weed on tropical cereals causing serious yield losses in Africa. The use of host crop varieties with improved resistance and tolerance against this parasite is a key component of an integrated control strategy. Breeding for tolerance is however seriously hampered by the absence of reliable and yet practical selection measures. The observation that the photosynthetic rate of tolerant genotypes is less sensitive to Striga infection was used as a starting point to search for suitable selection measures. In a greenhouse pot experiment the effect of Striga infection on the photosynthesis of four sorghum (Sorghum bicolor [L.] Moench) genotypes, differing in Striga tolerance level, was measured at three moments in time (26, 48 and 75 days after sowing). Genotypes were CK60-B, E36-1, Framida and Tiémarifing. Measurements involved CO2-assimilation (A) and three chlorophyll fluorescence characteristics (electron transport rate through photosystem II [ETR], photochemical [Pq] and non-photochemical quenching [NPq]). Striga infection negatively affected A, ETR and Pq. Based on A and Pq, genotypes with superior levels of tolerance (Tiémarifing) could be discriminated from genotypes with superior level of resistance (Framida). Both A and Pq showed high heritabilities and consequently clear and predictable differences between genotypes. Using discriminative ability, heritability and cost effectiveness as main criteria, photochemical quenching (Pq) was concluded to possess the highest potential to serve as indirect selection measure for host plant tolerance to Striga. Screening should preferably be conducted at relatively high Striga infestation levels, between Striga emergence and host plant flowering.  相似文献

Rhamphicarpa fistulosa is a facultative hemi‐parasitic plant of the Orobanchaceae family, adapted to wet soils. Apart from tropical Australia, it is only found in sub‐Saharan Africa, where it is considered a minor weed in cereal crops such as rice. Due to this status, the species has received only sporadic attention. Recent field observations and encounters with rice farmers in several African countries showed that R. fistulosa is, however, a more serious and increasing production constraint than previously thought. Results from a systematic literature review and a global herbarium study support this. The species has a broad distribution over Africa (at least 35 countries from Madagascar to Senegal and from Sudan to South Africa) and a wide range in altitude (0–2150 m a.s.l.) and environment (waterlogged swamps to moist free‐draining uplands). Rhamphicarpa fistulosa is relatively independent and persistent because of the presumably wide host range, the facultative nature of its parasitism and its prolific seed (estimated 100 000 seeds m?2 under moderate infestation levels). Finally, R. fistulosa causes severe yield losses (average 60%) and high regional annual economic losses (estimated US $175 million), while effective control options are scant and awareness of the species among important R&D stakeholders is almost absent. An integrated approach is advocated to assist the rice sector to reduce current R. fistulosa‐inflicted losses and to prevent further spread of the species into new areas.  相似文献
