This paper describes the practical use of a global positioning system receiver (hand-held GPS) as a means of measuring and describing pasture areas invaded by weeds. The accuracy of two GPS units, a hand-held GPS with an external antenna (GPS with an antenna) and the differential global positioning system receiver (DGPS), were examined in Morioka, northern Japan. In addition, an area of weed patches and a pasture, determined using the GPS with an antenna, were compared to the measurements made with a conventional tape and a weed map was created based on the coordinate data of latitude and longitude measurements. The accuracy of the GPS with an antenna was poor (8.3 m); however, the precision of the unit was reasonable in measuring area. An area estimation error by the GPS with an antenna was 7% when practically measuring weed patches of 141 m2 and 1% in a paddock of 12 566 m2 . From these results, it appears that the GPS with an antenna might have an acceptable error in measuring areas for weed control in a pasture. A weed map produced from the coordinate data surveyed using the GPS with an antenna enables the state of weed growth and its domination in an area of pasture to be visually understood. We investigated the in
vitro differentiation of canine bone marrow stromal cells (BMSCs) into voltage-
and glutamate-responsive neuron-like cells. BMSCs were obtained from the bone marrow of
healthy beagle dogs. Canine BMSCs were incubated with the basal medium for neurons
containing recombinant human basic fibroblast growth factor (bFGF; 100
ng/ml). The viability of the bFGF-treated cells was
assessed by a trypan blue exclusion assay, and the morphology was monitored. Real-time
RT-PCR was performed to evaluate mRNA expression of neuronal, neural stem cell and glial
markers. Western blotting and immunocytochemical analysis for the neuronal markers were
performed to evaluate the protein expression and localization. The Ca2+
mobilization of the cells was evaluated using the Ca2+ indicator Fluo3 to
monitor Ca2+ influx. To investigate the mechanism of bFGF-induced neuronal
differentiation, the fibroblast growth factor receptor inhibitor, the phosphoinositide
3-kinase inhibitor or the Akt inhibitor was tested. The bFGF treatment resulted in the
maintenance of the viability of canine BMSCs for 10 days, in the expression of neuronal
marker mRNAs and proteins and in the manifestation of neuron-like morphology. Furthermore,
in the bFGF-treated BMSCs, a high concentration of KCl and L-glutamate induced an increase
in intracellular Ca2+ levels. Each inhibitor significantly attenuated the
bFGF-induced increase in neuronal marker mRNA expression. These results suggest that bFGF
contributes to the differentiation of canine BMSCs into voltage- and glutamate-responsive
neuron-like cells and may lead to the development of new cell-based treatments for

Chondrostereum purpureum, a phytopathogenic fungus, produces endopolygalacturonase (endoPG) which has been suggested to have a causal role in the silver-leaf symptom of apple trees. In this paper, we detected C. purpureurn-derived endoPG at the infection sites using ELISA with a polyclonal antibody against endoPG I. A gene encoding endoPG I and its homolog were also isolated from the C. purpureum genome. The endoPG I gene was designated as cppg1. The cppg1 gene is the first fungal endoPG gene reported in the Basidiomycetes.

ABSTRACT: The aim of this study was to develop a practical preventive method for color deterioration of sliced or filleted yellowtail muscle, especially of dark muscle, during frozen storage and post thawing. When the sliced meats were packaged in a vacuum with low oxygen permeable flexible films and then stored frozen below −40°C, no significant discoloration or browning of dark muscle was observed for 9 months or more. For higher temperature storage at −20°C or −30°C, nitrogen gas substituted packaging was a useful practical method for storing sliced meats for 6 weeks. In order to prevent color deterioration of sliced meats after thawing and subsequent storage at 0°C, the efficacy of materials of packaging was investigated. The most desirable result was obtained by using a film with oxygen permeability of 50–90 cm3 /m2 per 24 h. 相似文献

ABSTRACT: We carried out mail and access point surveys to estimate the catch numbers, angling effort of recreational angling and the population of sockeye salmon Oncorhynchus nerka in Lake Toya in 1998 and 1999. The mail survey questions in questionnaires distributed to anglers with a recreational angling license included date of angling, caught fish species and size, and catch number. In the access point survey we also asked anglers if they had a license. We measured the fork length and weight of caught fish. The catch rate (catch number in a day by each angler) from both surveys showed no significant difference, suggesting no bias for the catch rate between surveys. The estimated total angling effort was 1760 people (1998), and 1516 people (1999). The estimated recreational catch was 28 889 (1998) and 5455 (1999), that is, two or three times larger than the commercial catch. The fish population was estimated by using the DeLury method as 60 262 (1998) and 10 806 (1999). The total exploitation rate was 62% (1998) and 78% (1999). The size of caught fish was much larger in 1999 than 1998 because of the difference in age composition. 相似文献

Estrogen action is mediated through several types of receptors (ERs), such as ERα, ERβ
and putative membrane ERs. Oxytocin receptor (OTR) and ER expression levels in the rat
uterus are regulated by estrogen; however, which types of ERs are involved has not been
elucidated. This study examined OTR, ERα and ERβ levels in ovariectomized rats treated
with 17β-estradiol (E2), an ERα agonist (PPT), an ERβ agonist (DPN) or estren (Es). E2 and
PPT increased OTR mRNA levels and decreased ERα and ERβ mRNA levels 3 and 6 h
posttreatment. DPN decreased ERα and ERβ mRNA levels at 3 and 6 h, while OTR mRNA levels
increased at 3 h and decreased at 6 h. OTR mRNA levels increased 3 h after the Es
treatment and then declined until 6 h. ERα and ERβ mRNA levels decreased by 3 h and
remained low until 6 h posttreatment with Es. The ER antagonist ICI182,780 (ICI)
suppressed the increases in OTR mRNA levels induced 3 h after the Es treatment. However,
ICI and tamoxifen (Tam) had no significant effect on ERα and ERβ mRNA levels in the
Es-treated or vehicle-treated group. In intact rats, proestrus-associated increases in OTR
mRNA levels were antagonized by both ICI and Tam. However, decreases in ERα and ERβ mRNA
levels were not antagonized by Tam and ICI, respectively. Therefore, uterine OTR gene
expression is upregulated by estrogen through the classical nuclear (or non-nuclear) ERs,
ERα and ERβ, while the levels of these ERs are downregulated by estrogen through multiple
pathways including Es-sensitive nonclassical ERs. 相似文献

The objective of this grazing study was to determine the herbage mass, nutritive value and palatability of five grass weed species ( Eleusine indica (L.) Gaertn, Paspalum thunbergii Kunth, Digitaria ciliaris (Retz.) Koeler, Setaria glauca (L.) Beauv. and Echinochloa crus-galli (L.) Beauv. var. crus-galli ) as feeds compared with six forage cultivars ( Lolium perenne L., two Lolium multiflorum cultivars and three Echinochloa crus-galli (L.) Beauv. cultivars). The herbage mass of weed species, with the exception of S. glauca , was most prevalent in August. The total amounts of herbage mass of weed species (except E. indica ) were close to those of the Lolium cultivars, but lower than those of the E. crus-galli cultivars. During the summer, weed species (except E. crus-galli var. crus-galli ) contained favorable levels of total digestible nutrients (TDN), crude protein (CP), acid detergent fiber (ADF), neutral detergent fiber (NDF) and mineral contents, and exhibited adequate calcium (Ca):phosphorus (P) ratios of 1:2. All weed species had the recommended ratios of potassium (K):(Ca + [magnesium] Mg) < 2.2:1. By autumn, the summertime levels of nutritive values (TDN, CP, ADF and NDF) in the weed species had severely deteriorated, in contrast with those of the forage cultivars. However, they maintained satisfactory mineral content levels and Ca:P and K:(Ca + Mg) ratios (with the exception of E. indica with poor Ca:P ratios and E. crus-galli var. crus-galli with poor P content and Ca:P ratios).
