The molecular interactions between plants and sedentary nematodes are undergoing intense study, not only for reasons of fundamental research but also for the potential benefits to agriculture. The present technology allows the transformation of an increasing number of crop plants, providing new ways to introduce resistance against plant-parasitic nematodes. The ability of sedentary nematodes to induce specialized feeding sites in plant roots is one of the most fascinating aspects of this host–parasite interaction. Molecular approaches have been initiated to identify and characterize plant genes altered in expression after infection by sedentary nematodes. The results obtained indicate that many genes indeed become up-regulated upon nematode infection. Surprisingly, several so-called constitutive promoters that are normally used to achieve high expression in plant cells are completely ‘silenced’ in the feeding sites within days after nematode infection. Generally, there are two options available for the genetic engineering of nematode resistance: the synthesis of anti-nematode proteins or the localized production of a cytotoxic protein that interferes with the development of feeding cells. Nematode-induced promoters are very useful for the production by plants of sufficiently high levels of anti-nematode proteins at feeding sites. Alternatively, interfering with feeding-cell development is somewhat similar to the hypersensitive response evoked by nematodes in a naturally resistant plant. Here, destruction of specific plant cells can be achieved by the localized expression of a cytotoxin such as barnase, a potent ribonuclease. This approach, however, calls for a highly specific ‘non-leaky’ promoter, which is active only in the feeding cells. Another possibility is to use a two-component system, where the leakiness of the promoter in other tissues is counterbalanced by the constitutive expression of a neutralizing gene.  相似文献

This study focused on clarifying phylogenetic relationships among Citrus accessions from Vietnam. Our phylogenetic analysis based on nucleotide sequences from the ITS of the ribosomal DNA included 69 accessions belonging to Citrus and related (sub)genera. Maximum parsimony and Bayesian analysis confirmed a clear separation of the three ‘true’ Citrus species (C. medica, C. maxima and C. reticulata). Confirming recent taxonomic revisions, Fortunella, Poncirus trifoliata and Citrus hystrix are clustered among the accessions of subgenus Citrus. C. × sinensis accessions revealed a close evolutionary relationship to either C. maxima or C. reticulata, thereby confirming their involvement in its hybrid origin. Also, some other hybrid taxa and their proposed parental species were investigated and their origin could in some cases be confirmed using the ITS sequence data.  相似文献

The mode of inheritance of chloroplast and mitochondrial DNA has been determined in intergeneric hybrids between C. papaya and four different Vasconcellea species by employing a restriction fragment length polymorphism analysis of a PCR-amplified chloroplast and mitochondrial DNA region. Artificial F1 hybrids were produced between a female specimen of C. papaya and male specimens of either V. parviflora, V. goudotiana, V. cundinamarcensis or V. quercifolia. The hybridization patterns of all hybrids correspond in all cases with that of the C. papaya mother, and are different from that of the paternal Vasconcellea species, thus indicating the maternal inheritance of cpDNA and mtDNA in intergeneric hybrids between C. papaya and wild relatives of the Vasconcellea genus.  相似文献

Bacterial blight (BB) incited by Xanthomonas citri subsp. malvacearum (Xcm) is an important bacterial disease occurring in all cotton growing areas throughout the world, including parts of Western Kenya, that are characterized by a hot and humid climate. The disease causes seedling blight, angular leaf spot, boll rot and black arm on petioles and branches leading to a loss of fruiting branches with yield losses of up to 35 %. Fifty- one Kenyan cotton genotypes (Gossypium hirsutum L.) were established in the two counties of Siaya and Busia famous for their cotton cultivation in the western region. The BB symptoms caused by natural Xcm infection in the field were scored for each cotton accession. In addition artificial inoculation with the same strains was done to confirm the status under controlled greenhouse conditions. The results of BB disease scoring revealed that some accessions (e.g. T 24A and T 24B) possess a reasonable level of resistance. However, most of the Kenyan genotypes surveyed showed medium to severe symptoms of BB, including KSA 81 M, the only commercially grown cultivar in the region of Western Kenya. Overall, 71 % of the genotypes showed susceptibility and 29 % were classified as either resistant or moderately resistant. There is therefore need to improve the local commercial genotypes by introducing new genetic resources with a more durable BB resistance to ensure a successful revitalization of the Kenyan cotton sector.  相似文献

Baobab (Adansonia digitata L.) is one of the predominant tree species in West African agroforestry systems. A local morphological classification system is used by farmers, identifying trees with desired or undesired combinations of traits. This study evaluates the genetic significance of these morphotypes by comparing local identification with AFLP marker information. Eight morphotypes were recognized by seven ethnic groups from Benin, Ghana and Senegal, among 182 sampled baobab trees. Five primer pairs were used for DNA fingerprinting, resulting in a total of 254 scored bands, of which between 94.1% and 100% was polymorphic within morphotypes. Generally, genetic fingerprinting did not correlate with the traditional morphological identification of Adansonia digitata. Probably, AFLP markers are not directly linked to the differences in phenotype or the traits used for the traditional classification are largely dependent on environmental factors. Since no genetic differentiation is found between the morphotypes, a morphotype-based approach in the collection of genetic variation for conservation programs is not advisable. A. E. ? Context

Over the past few decades, the impact of large herbivorous ungulates on forest vegetation has been clearly highlighted. Among those impacts, bark stripping of coniferous trees is one of the most damaging. Bark stripping leads to rot development, inducing serious loss of timber value.

? Aims

The present study aimed firstly at evidencing the factors explaining the variations observed in fresh bark peeling rate for spruce and Douglas-fir in southern Belgium and secondly at identifying the key factors to consider when setting up a deer management plan.

? Method

Fresh bark peeling rate was recorded with a systematic sampling survey from 2004 to 2007. The covered territory was then divided into 63 distinct hunting zones of area ranging from 1,000 to 25,000 ha. About 5,000 plots were monitored annually. Each zone was characterized with a large number of explanatory variables. The explanatory variables were integrated firstly into fixed linear models using a stepwise procedure, and then into a mixed model.

? Results

The significant variables included in the model (R 2?=?44 %) are (by decreasing order of importance) red deer densities, proportion of coniferous stands and agricultural areas, snow cover, distance to urban habitats, and species diversity in the understory.

? Conclusion

The models revealed the impacts of several factors on bark peeling: deer density, deer-carrying capacity of the territory, landscape structure, and severity of winter conditions. The adjusted model allowed subtracting the impact of winter conditions in order to produce a relevant indicator for hunting management. In addition, the model was used to assess the sensitivity of a forested area to bark peeling based on its environmental characteristics.  相似文献
