In this work, we investigated the spasmolytic effect of caulerpine, a bisindole alkaloid isolated from marine algae of the Caulerpa genus, on guinea pig ileum. Our findings indicated that caulerpine inhibited phasic contractions induced by carbachol (IC50 = 7.0 ± 1.9 × 10−5 M), histamine (IC50 = 1.3 ± 0.3 × 10−4 M) and serotonin (IC50 = 8.0 ± 1.4 × 10−5 M) in a non-selective manner. Furthermore, caulerpine concentration-dependently inhibited serotonin-induced cumulative contractions (pD′2 = 4.48 ± 0.08), shifting the curves to the right with Emax reduction and slope of 2.44 ± 0.21, suggesting a noncompetitive antagonism pseudo-irreversible. The alkaloid also relaxed the ileum pre-contracted by KCl (EC50 = 9.0 ± 0.9 × 10−5 M) and carbachol (EC50 = 4.6 ± 0.7 × 10−5 M) in a concentration-dependent manner. This effect was probably due to inhibition of Ca2+ influx through voltage-gated calcium channels (CaV), since caulerpine slightly inhibited the CaCl2-induced contractions in depolarizing medium without Ca2+, shifting the curves to the right and with Emax reduction. According to these results, the spasmolytic effect of caulerpine on guinea pig ileum seems to involve inhibition of Ca2+ influx through CaV. However, other mechanisms are not discarded. 相似文献

The objectives of this study were to obtain information about genetic parameters of a partial diallel in common bean, as well as to evaluate the effects of growing seasons, locations and generations in these estimates in order to define the best strategies for the use of diallel analysis as methodology of selection of parents and populations in common bean breeding. Twelve parents were crossed in a partial diallel scheme (5?×?7), and the F1 and F2 generations were evaluated in four experiments (I, II, III, and IV). The effects of seasons were determined in the Experiments I and II, in which the F1 generation was evaluated in two distinct growing seasons. To quantify the effect of locations, we used Experiments III and IV, in which the F2 generation was evaluated in the same season, but at different locations, and to quantify the effect of generations, the experiments II (generation F1) and III (F2 generation) were evaluated in the same season and location. As for grain yield the genetic control varied according to the environment. An effect of interaction on the genetic parameters of the diallel was observed for generation, whereas for grain yield, effects of season and location were observed. For plant architecture, parental combining ability should therefore be estimated using the F1 generation, while for grain yield, both the F1 and F2 can be used. The recombination between the best hybrids is a promising strategy for extraction of superior black bean lines. 相似文献

The antinociceptive and anti-inflammatory activity of caulerpin was investigated. This bisindole alkaloid was isolated from the lipoid extract of Caulerpa racemosa and its structure was identified by spectroscopic methods, including IR and NMR techniques. The pharmacological assays used were the writhing and the hot plate tests, the formalin-induced pain, the capsaicin-induced ear edema and the carrageenan-induced peritonitis. Caulerpin was given orally at a concentration of 100 μmol/kg. In the abdominal constriction test caulerpin showed reduction in the acetic acid-induced nociception at 0.0945 μmol (0.0103–1.0984) and for dypirone it was 0.0426 μmol (0.0092–0.1972). In the hot plate test in vivo the inhibition of nociception by caulerpin (100 μmol/kg, p.o.) was also favorable. This result suggests that this compound exhibits a central activity, without changing the motor activity (seen in the rotarod test). Caulerpin (100 μmol/kg, p.o.) reduced the formalin effects in both phases by 35.4% and 45.6%, respectively. The possible anti-inflammatory activity observed in the second phase in the formalin test of caulerpin (100 μmol/kg, p.o.) was confirmed on the capsaicin-induced ear edema model, where an inhibition of 55.8% was presented. Indeed, it was also observed in the carrageenan-induced peritonitis that caulerpin (100 μmol/kg, p.o.) exhibited anti-inflammatory activity, reducing significantly the number of recruit cells by 48.3%. Pharmacological studies are continuing in order to characterize the mechanism(s) responsible for the antinociceptive and anti-inflammatory actions and also to identify other active principles present in Caulerpa racemosa. 相似文献

The aim of this study was to evaluate the effect of increasing concentrate supplementation levels on the intake, nutrient digestibility, and performance of crossbred steers during the dry period of the year. The experiment was developed on Princesa do Mateiro farm, in the municipality of Ribeirão do Largo, located in the southwest region of Bahia State, Brazil. Forty uncastrated male crossbred (½ Holstein-Zebu) steers with an average body weight (BW) of 232.55?±?24.97 kg were distributed into four treatments in a completely randomized design with ten replicates. The animals were managed in an experimental area formed by Brachiaria brizantha cv. Marandu, in an intermittent grazing system. Treatments consisted of the following supplementation levels: 0.2% BW, with 60% crude protein (CP); 0.3% BW, with 40% CP; 0.4% BW, with 30% CP; and 0.5% BW, with 24% CP. The intakes of forage dry matter in kg/day and %BW and neutral detergent fiber corrected for ash and protein (NDFap) in %BW decreased linearly, whereas the intake of non-fibrous carbohydrates corrected for ash and protein in kg/day and average daily gain increased linearly. Therefore, the use of supplementation at 0.5% BW (24% crude protein) to provide gains of up to 0.500 kg/day is recommended for grazing steers during the post-weaning period in the dry season of the year. 相似文献

Cercosporiosis, or brown eye spot, is currently one of the main diseases of the coffee tree. It is caused by Cercospora coffeicola Berk. & Cooke. Nevertheless, genetic resistance to this disease has not yet been explored in any depth. Our objectives (a) were evaluate the response of 124 accessions from the germplasm collection of the Minas Gerais State (GC), Brazil, and eight commercial cultivars of C. arabica to cercosporiosis and (b) determine the best way to perform early progenies selections via controlled greenhouse experiments. Three controlled greenhouse experiments (1–3) were run in different seasons to determine the best way to proceed the selection. The seedlings were inoculated with a four isolates mixture obtained from different regions. The experimental data were analyzed individually (1–3), in a joint analysis, and as repetitions of a randomized complete block design. In each analysis we estimated genetic parameters and E-BLUP (empirical best linear unbiased predictor) genotypic values of the access. There was genetic variability to C. coffeicola resistance among the coffee tree accessions germplasm collection. Therefore, genetic improvements could be obtained by selection. Experimental repetitions in different seasons increase the selection efficiency and reliability of resistant genotypes with low cercosporiosis severity. The genotype Sarchimor MG 8840 showed the highest resistance level followed by Guatenano and the Timor Hybrid UFV 377-34, Timor Hybrid UFV 376-14 BE 5, and Wush–Wush × Timor Hybrid UFV 366-08. 相似文献

Marine natural products have been the focus of discovery for new products of chemical and pharmacological interest. The aim of this study was to evaluate the antinociceptive activity of the methanolic (ME), acetate (AE), hexanic (HE) and chloroform (CE) extracts obtained from Caulerpa mexicana, and ME, CE and HE obtained from Caulerpa sertularioides. These marine algae are found all over the world, mainly in tropical regions. Models such as the writhing test, the hot plate test and formalin-induced nociception test were used to evaluate antinociceptive activity in laboratory mice. In the writhing test, all the extracts were administered orally at a concentration of 100 mg/kg, and induced high peripheral antinociceptive activity, with a reduction in the nociception induced by acetic acid above 65%. In the hot plate test, treatment with extracts from C. sertularioides (100 mg/kg, p.o.) did not significantly increase the latency of response, although the ME, AE and HE from C. mexicana showed activity in this model. This result suggests that these extracts exhibit antinociceptive activity. In the formalin test, it was observed that ME, AE and HE obtained from C. mexicana reduced the effects of formalin in both phases. On the other hand only CE from C. sertularioides induced significant inhibition of the nociceptive response in the first phase. To better assess the potential anti-inflammatory activity of the extracts, the carrageenan-induced peritonitis test was used to test Caulerpa spp. extracts on cell migration into the peritoneal cavity. In this assay, all extracts evaluated were able to significantly inhibit leukocyte migration into the peritoneal cavity in comparison with carrageenan. These data demonstrate that extracts from Caulerpa species elicit pronounced antinociceptive and anti-inflamatory activity against several nociception models. However, pharmacological and chemical studies are continuing in order to characterize the mechanism(s) responsible for the antinociceptive action and also to identify the active principles present in the Caulerpa species. 相似文献

Background: The health risks of crack cocaine smoking on the oral mucosa has not been widely researched and documented. Objective: The purpose of this study was to analyze the proliferative activity of oral epithelial cells exposed to crack cocaine smoke using silver nucleolar organizer region (AgNOR) staining. Methods: Oral smears were collected from clinically normal-appearing buccal mucosa by liquid-based exfoliative cytology of 60 individuals (30 crack cocaine users and 30 healthy controls matched for age and gender) and analyzed for cytomorphologic and cytomorphometric techniques. Results: Crack cocaine users consumed about 13.3 heat-stable rocks per day and the time consumption of the drug was of 5.2 (± 3.3) years. Mean values of AgNOR counting for case and control groups were 5.18 ± 1.83 and 3.38 ± 1.02 (P<0.05), respectively. AgNOR area and percentage of AgNOR-occupied nuclear area were increased in comparison with the control (P<0.05). There was no statistically significant difference in the mean values of the nuclear area between the groups (P>0.05). Conclusion: This study revealed that crack cocaine smoke increases the rate of cellular proliferation in cells of normal buccal mucosa. Key Words: Crack-Cocaine, Mouth mucosa, Cell proliferation 相似文献
