This study investigated whether the increase in wheat resistance to blast, caused by Pyricularia oryzae, potentiated by silicon (Si) is linked to changes in the activity of antioxidative enzymes. Wheat plants (cv. BR 18) were grown in hydroponic culture with either 0 (–Si) or 2 mm (+Si) Si and half of the plants in each group were inoculated with P. oryzae. Blast severity in the +Si plants was 70% lower compared to the ?Si plants at 96 h after inoculation (hai). Superoxide dismutase (SOD), catalase (CAT), peroxidase (POX), ascorbate peroxidase (APX) and glutathione‐S‐transferase (GST) activities were higher in the leaves of the ?Si plants compared with the +Si plants at 96 hai. This indicates that other mechanisms may have limited P. oryzae infection in the +Si plants restricting the generation of reactive oxygen species, obviating the need for increased antioxidative enzyme activity. In contrast, glutathione reductase (GR) activity at 96 hai was higher in the +Si plants than in the ?Si plants. Although the inoculated plants showed significantly higher concentration of malondialdehyde (MDA) than the non‐inoculated plants, lower MDA concentrations were observed in the +Si plants compared with the ?Si plants. The lower MDA concentration associated with decreased activities of SOD, CAT, POX, APX and GST, suggest that the amount of reactive oxygen species was lower in the +Si plants. However, GR appears to play a pivotal role in limiting oxidative stress caused by P. oryzae infection in +Si plants.  相似文献

Although magnesium (Mg) is considered an essential element for wheat growth, its importance for disease control has often been overlooked, and the physiological features of diseased plants mediated by Mg remain elusive. In this study, the effect of three Mg concentrations (0·25, 2·5 and 4 mm ) on wheat resistance to leaf blast (Pyricularia oryzae), leaf gas exchange, invertase activity, cellular damage and foliar concentration of photosynthetic pigments and nutrients was investigated. Foliar Mg increased from 1·9 to 3·9 g kg?1, whereas calcium (Ca) decreased from 7·8 to 4·9 g kg?1 as the applied Mg increased from 0·25 to 4 mm . Blast severity increased from 11·3 to 39·6% as the applied Mg increased from 0·25 to 4 mm . Photosynthesis, stomatal conductance, transpiration and photosynthetic pigment concentrations decreased in inoculated plants compared to non‐inoculated plants regardless of the Mg concentration; however, the reductions were more pronounced for plants grown with 4 mm Mg than those grown with 0·25 mm Mg. On the other hand, a higher internal CO2 concentration, invertase activity and malondialdehyde concentration was recorded in inoculated plants grown with 4 mm Mg compared to those grown with 0·25 mm Mg. In conclusion, reduced Ca uptake may partially explain the increased susceptibility of wheat to leaf blast with the highest Mg concentration. Mg‐induced susceptibility to leaf blast appeared responsible for the photosynthetic impairments. These were most probably due to biochemical constraints because plants grown with the highest Mg concentration suffered extensive cellular damage and degradation of photosynthetic pigments as a result of high disease severity.  相似文献

This study aimed to develop and validate a standard area diagram set (SADS) to quantify the severity of blast, caused by Pyricularia oryzae, on wheat leaves. The SADs has ten levels: 0.1, 1, 5, 10, 22, 32, 42, 52, 62 and 72 % blast severity. To validate the SADs, 12 inexperienced raters estimated disease severity on 50 images of leaves from cultivars BR-18 (susceptible) and BRS-229 (partially resistant). Blast severity was first estimated without the use of the SADs on 50 leaves with a range of blast severity. The same raters evaluated the same 50 leaves using the SADs as an aid. The SADs improved accuracy (coefficient of bias, C b ?=?0.88 and 0.99, without and with SADs, respectively) and agreement (Lin’s concordance correlation coefficient, ρ c ?=?0.84 and 0.96 without and with SADs, respectively) of the estimates of severity. The absolute error was (-) 52 % without the SADs and (-) 24 % when using SADs as an aid. Severity estimates were more reliable when using SADs (R2?=?0.87 unaided and R2?=?0.92 with SAD). The SADs proposed in this study will improve accuracy and reliability of estimates of blast severity on wheat leaves.  相似文献
