Two near-isogenic lines of the processing tomato cv. ‘IPA-5’, pheno-typically distinct from each other only in terms of the presence or absence of a jointless fruit pedicel, were used to estimate pleiotropic effects of the recessive gene j-2 on 15 processing and agronomic parameters. Total yield, total yield of mature fruit, total yield of green fruit, total number of fruit, total number of mature fruit, total number of green fruit, mean weight of mature fruit, mean weight of green fruit, mean weight of mature plus green fruit, soluble solids, fruit colour, pH, titratable acidity, sugar: acid ratio, and fruit firmness were compared in jointed (J-2/J-2) and jointless pedicel (J-2/J-2) lines. Statistically significant differences were detected between both isolines for the following parameters: total number of fruit, total yield, total yield of green fruit, and total number of green fruit. In all cases, smaller values were observed in the isoline with the genetic constitution J-2/J-2. These data reinforce the view that they j-2 gene may cause, in some specific tomato backgrounds, significant yield constraints. Furthermore, it was demonstrated that this gene cannot exert any important pleiotropic influence on any of the processing characteristics evaluated.  相似文献

The whitefly-transmitted Tomato chlorosis virus (ToCV) (genus Crinivirus) is associated with yield and quality losses in field and greenhouse-grown tomatoes (Solanum lycopersicum) in South America. Therefore, the search for sources of ToCV resistance/tolerance is a major breeding priority for this region. A germplasm of 33 Solanum (Lycopersicon) accessions (comprising cultivated and wild species) was evaluated for ToCV reaction in multi-year assays conducted under natural and experimental whitefly vector exposure in Uruguay and Brazil. Reaction to ToCV was assessed employing a symptom severity scale and systemic virus infection was evaluated via RT-PCR and/or molecular hybridization assays. A subgroup of accessions was also evaluated for whitefly reaction in two free-choice bioassays carried out in Uruguay (with Trialeurodes vaporariorum) and Brazil (with Bemisia tabaci Middle-East-Asia-Minor1—MEAM1?=?biotype B). The most stable sources of ToCV tolerance were identified in Solanum habrochaites PI 127827 (mild symptoms and low viral titers) and S. lycopersicum ‘LT05’ (mild symptoms but with high viral titers). These two accessions were efficiently colonized by both whitefly species, thus excluding the potential involvement of vector-resistance mechanisms. Other promising breeding sources were Solanum peruvianum (sensu lato) ‘CGO 6711’ (mild symptoms and low virus titers), Solanum chilense LA1967 (mild symptoms, but with high levels of B. tabaci MEAM1 oviposition) and Solanum pennellii LA0716 (intermediate symptoms and low level of B. tabaci MEAM1 oviposition). Additional studies are necessary to elucidate the genetic basis of the tolerance/resistance identified in this set of Solanum (Lycopersicon) accessions.  相似文献

The degree of heterosis for total tuber yield (TTY) and total solids (TS) in 4x‐2x crosses was estimated by comparing the performance of 12 families with their respective parents in two locations in Wisconsin (USA). The parental 2x clones were Phureja‐haploid Tuberosum hybrids with 2n‐pollen production by first‐division restitution. The general combining ability (GCA) and specific combining ability (SCA) were estimated for TTY, TS, vine maturity (VM), length of tuber sprout dormancy (LD), and tuber eye depth (ED). Family performance for TTY ranged from 74 to 146% at Hancock (E#1) and from 77 to 287 at Rhinelander (E#2) when compared with that of the 4x parent group. For VM, the families were late maturing, but a few precocious ones were identified. For TS, the families had heterosis of 5.1% over the 4x parent group. The families had slightly higher ED values than the 4x parents, but families with values within the commercial range were identified. The family average for LD (54 days) was closer to the 2x group (51 days) than to the 4x group (88 days). The direction and magnitude of the parent‐family relationships were variable. The 4x parent TTY was correlated with progeny in E#1 but not E#2. The 2x parent VM had correlation with the offspring at E#2 but not at E#1. The type of gene action had a trait‐specific expression. Significant SCA and GCA variances were observed, suggesting that additive as well as non‐additive genetic effects were operating. The 4x‐2x crosses were able to generate heterotic families for TTY and TS in combination with other useful traits. However, no promising results were found for LD because of the apparent dominance of the short‐dormancy phenotype. This result indicates the need of additional selection and breeding efforts for some specific traits when using S. phureja‐derived germplasm.  相似文献

The relative value of haploid Tuberosum-Solanum chacoense (TCH) vs. Solanum phureja-haploid Tuberosum (PTH) hybrids as male parents in 4x–2x crosses was estimated under two distinct crop management systems. The first experiment (E#1) was carried out with supplementary irrigation (three times a week) at Hancock-Wisconsin (WI) with 27 families derived from a sub-set of crosses involving 11 tetraploid (4x) cultivars and 10 diploid (2x) clones (3 TCH; 6 PTH; and one TCH-PTH hybrid clone). A second experiment (E#2) was conducted at Rhinelander-WI with the same group of families but it was only rainfed with no supplementary irrigation being provided during the entire course of the assay. For comparison, a complete set of 4x and 2x clones (used as parents) was also planted in each location. Three traits were evaluated in both locations: total tuber yield (TTY), haulm maturity (HM), and plant vigor (PV). Altogether, 18 out of 27 and 10 out of 27 families outyielded the corresponding 4x parents at E#1 and E#2, respectively. Significant differences among all 4x–2x families and among families of the TCH group were observed for TTY at both locations. No difference was found for TTY among families of PTH and [TCH–PTH] groups at each location. A total of 75% and 50% of the TCH families outyielded their 4x parents at E#1 and E#2, respectively. The percentage of families from the PTH group that outyielded their 4x parents at E#1 and E#2 was 83.3% and 25%, respectively. The TCH group had on average lower HM scores than PTH (at both locations) indicating some segregation for earliness. However, the HM values of the 4x–2x families were, in general, higher than those observed for the 4x parents. For PV, the PTH group mean was significantly higher than TCH group only in E#1. The PV values of the 4x–2x families were higher than the 4x-parent group at both locations. Our results indicated that TCH hybrids might be as good parents as PTH to increase the TTY of 4x–2x progenies. In addition, the TCH families displayed a slight higher level of performance under more stressful growing conditions than PTH hybrids as indicated by the assay at Rhinelander. Therefore, selection of genetic materials with potential broad range of adaptation seems to be feasible with both hybrids via the 4x–2x breeding scheme. A phenotypically and geographically diverse collection of 45 Arabidopsis thaliana ecotypes was evaluated for its reaction to the northern root-knot nematode (Meloidogyne hapla) under greenhouse conditions. A significant ecotype effect was observed for both criteria used for host reaction assessment: namely, root-gall rating (RGR) and nematode reproductive index (RI). A non-host (non-galling) response was not detected in this germplasm collection. The majority of the ecotypes showed high degrees of RGR and RI. However, some medium-late maturing ecotypes such as ‘CS-1540’ and ‘CS-6028’(both collected in the UK) displayed high RGR but in association with very reduced RI. The confirmation of A. thaliana as an experimental host of M. hapla and the identification of differential levels of response to infection by this endoparasitic nematode provides the opportunity for genetic and molecular characterization of a new set of plant genes expressed during plant-nematode interaction. A. thaliana is the smallest and the best characterized genome among plant species, which may facilitate gene isolation and cloning. This may hasten transgenic transfer to economically important host plants of the genetic factors controlling low RI levels identified in some A. thaliana ecotypes.  相似文献

Summary Inheritance studies were conducted to determine the genetic basis of adult-plant resistance in pepper (Capsicum annuum L.) to Phytophthora capsici. F1, backcrosses and F2 populations were developed using the resistant parent Criollo de Morellos 334 and susceptible parents Agronômico 10-G and Yolo Wonder. Pepper plants, at 36 days post-emergence, were inoculated near the base of the stem with an inoculum suspension of 5×104 zoospores/ml. Segregation ratios in the F2 generation of 13 resistant to 3 susceptible plants fit a 2-gene model for resistance with dominant and recessive epistasis.  相似文献

Summary The genetic basis of the resistance to a pepper strain of potato virus Y (PVY pathotype 1–2) was investigated in two lines: Capsicum annuum Criollo de Morellos 334 and Capsicum chinense PI 159236. F1, backcrosses, and F2 populations were produced using C. annuum Magda as the susceptible parent. Segregation ratios indicated that the resistance in C. annuum Criollo de Morellos 334 is governed by a nuclear, single dominant gene with a very high but incomplete penetrance. On the other hand, the resistant response of C. chinense PI 159236 is associated with the presence of a major recessive gene with an apparently complete penetrance. The failure in recovering or detecting PVY from/in the inoculated leaves of both resistant genotypes is strongly suggestive that both genes are acting by preventing the multiplication of the virus in the leaf tissue. Both mechanisms of resistance closely resemble either an operational immunity or a single-cell hypersensitive reaction to PVY pathotype 1–2. The name Ry 1–2 is suggested for the resistance gene from Criollo de Morellos 334. The gene for resistance to PVY pathotype 1–2 identified in C. chinense PI 159236 may be the same designated et c1, which is mentioned to be allelic to the et av locus. The phenotypic expression of the et av gene was characterized in C. annuum Avelar as a tolerant but not immune or hypersensitive reaction against some potyviruses in Florida. These data suggest that a critical reexamination of the allelic relationships at the locus et should be performed.  相似文献

Recent outbreaks of Verticillium wilt diseases in various vegetable crops have been reported in Brazil. This fact was our initial stimulus to carry out a nationwide survey aiming to determine their causal agent(s). Thus far, Verticillium dahliae has been reported as the predominant species based solely on morphological traits. As other Verticillium species can be associated with wilt diseases, we characterized a collection of 80 isolates, collected across 10 agricultural Brazilian regions, by combining morphological, biological, and molecular traits. A multilocus approach was employed for identification of Verticillium species with information from three genomic regions (ribosomal internal transcribed spacer region, glyceraldehyde-3-phosphate dehydrogenase, and actin genes). Only 21 out of the 80 isolates were unable to produce microsclerotia in culture. The analyses of all genomic regions indicated V. dahliae as the sole species associated with vascular wilt of distinct hosts, including major solanaceous vegetables and other hosts such as strawberry, okra, and cacao. Pathogenicity tests confirmed the infection by the V. dahliae isolates and the development of typical disease symptoms on their original hosts. This is the first nationwide characterization of Verticillium isolates associated with major vegetable crops in Neotropical areas. This provides valuable information to design sound management strategies for these diseases, mainly for establishing efficient rotation systems and for the development of resistant cultivars.  相似文献

The genus Tospovirus was considered as monotypic with Tomato spotted wilt virus (TSWV) being the only assigned species. However, extensive studies with worldwide isolates revealed that this genus comprises a number of species with distinct virulence profiles. The Neotropical South America is one center of Tospovirus diversity with many endemic species. Groundnut ringspot virus (GRSV), TSWV, Tomato chlorotic spot virus (TCSV), and Chrysanthemum stem necrosis virus (CSNV) are the predominant tomato-infecting species in Brazil. Sources of resistance were found in Solanum (section Lycopersicon) mainly against TSWV isolates from distinct continents, but there is an overall lack of information about resistance to other viral species. One-hundred and five Solanum (section Lycopersicon: Solanaceae) accessions were initially evaluated for their reaction against a GRSV isolate by analysis of symptom expression and systemic virus accumulation using DAS-ELISA. A subgroup comprising the most resistant accessions was re-evaluated in a second assay with TSWV, TCSV, and GRSV isolates and in a third assay with a CSNV isolate. Seven S. peruvianum accessions displayed a broad-spectrum resistance to all viral species with all plants being free of symptoms and systemic infection. Sources of resistance were also found in tomato cultivars with the Sw-5 gene and also in accessions of S. pimpinellifolium, S. chilense, S. arcanum, S. habrochaites, S. corneliomuelleri, and S. lycopersicum. The introgression/incorporation of these genetic factors into cultivated tomato varieties might allow the development of genetic materials with broad-spectrum resistance, as well as with improved levels of phenotypic expression.  相似文献
