AIM: To investigate the effect of cGMP on voltage-gated potassium channel in pulmonary artery smooth muscle cells (PASMCs) from rats exposed to chronic hypoxia. METHODS: (1) Wistar rats were randomly divided into control group (group A) and chronic hypoxia group (group B). Then group B received hypoxia 8 hours per day for 4 consecutive weeks. (2) Single PASMC was obtained via acute enzyme separation method. (3) Conventional whole-cell patch clamp technique was used to record resting membrane potential (Em) and ion currents of voltage-gated potassium channel. The changes of ion currents of voltage-gated potassium channel before and after applying cGMP (1 mmol/L), an agonist of protein kinase G (PKG), and cGMP plus H-8 (1 mmol/L), an inhibitor of PKG were compared between two groups. RESULTS: The Em of group B were significantly lower than that of group A. The ion currents of voltage-gated potassium channel in group A and group B were all significantly inhibited by cGMP [control group: from (118.0±5.0) pA/pF to (89.9±16.5) pA/pF, n=6, P<0.05;chronic hypoxia group: from (81.0±5.0) pA/pF to (56.8±9.1) pA/pF, n=6, P<0.05]and these effects were reversed by H-8 [control group: from (119.2±10.3) pA/pF to (117.8±9.1) pA/pF, n=6, P>0.05;chronic hypoxia group: from (96.8±6.2) pA/pF to (98.0±2.2) pA/pF, n=6, P>0.05]. CONCLUSIONS: The currents of voltage-gated potassium channel was inhibited by chronic hypoxic. The inhibitory effect of cGMP on currents of voltage-gated potassium channel in PASMCs from both normal and chronic hypoxic rats may be probably through the phosphorylation of voltage-gated potassium channel.  相似文献

为探讨富铁酵母对家兔红细胞免疫功能的影响,将32只家免随机分成富铁酵母高、低剂量组,FeSO4组及空白对照组,分别灌服富铁酵母(Fe元素8 mg/kg、4 mg/kg)、FeSO4(Fe元素4 mg/kg)及空白生理盐水;连续5 d后,检测红细胞C3g受体花环率和复合物(IC)花环率.结果:富铁酵母较FeSO4及空白组能显著提高红细胞C3h受体花环和红细胞复合物(IC)花环的百分率,但高、低剂量间无显著性差异.结论:富铁酵母能显著提高红细胞免疫功能.  相似文献

在气候变化和人类活动的综合影响下,青海省生态环境发生了明显变化。在此背景下,以GIMMS NDVI 3g.v1为数据源,采用Sen+Mann-Kendal方法研究青海省1982-2015年植被覆盖区域NDVI时空变化,将趋势分析和R/S(rescaled range analysis)分析叠加,研究植被生长季NDVI变化的持续性特征,并揭示植被对气候变化及人类活动的响应规律。结果表明:1)近34年青海省植被NDVI整体呈从西北到东南的增加趋势;且变异系数显示,波动性较大地区集中在柴达木盆地周边和青南牧区西北部等植被NDVI较低的区域,波动性较小地区集中在祁连山东部、东部农业区和青南牧区东南部等植被NDVI较高的区域。2)近34年青海省植被NDVI整体呈增加趋势,增长率为0.38%·10a~(-1);且NDVI变化具有明显的阶段性,存在1994年和2000年两个突变点。3)近34年青海省植被改善区域(75.4%)远大于退化区域(24.6%),其中显著改善面积占植被覆盖区域面积的40.9%,退化区随时间变化在空间上表现出明显的转移现象。4)Hurst指数表明,青海省植被变化反持续性较强,趋势分析与Hurst指数叠加得出,由退化转为改善的区域占植被覆盖区面积的13.7%,由改善转为退化的区域占植被覆盖区面积的44.3%,另41.5%的区域无法确定未来变化趋势。5)青海省植被生长季NDVI受气候变化和人类活动的双重影响,且不同植被类型对气候变化的响应存在较大差异。  相似文献

目的 测定吴茱萸生药和提取物中总皂苷、总黄酮和总酚酸的含量.方法 采用80%乙醇提取,正丁醇萃取,乙醚脱脂的方法提取吴茱萸;用分光光度法测定总皂苷、总黄酮和总酚酸的含量.结果 实验用的两种吴茱萸生药中总皂苷、总黄酮和总酚酸的百分含量分别为5.78、5.24、30.23和12.36、10.99、24.36;提取物中总皂苷、总黄酮和总酚酸的百分含量分别为29.56、52.14、81.88.结论 不同吴茱萸药材中总皂苷、总黄酮和总酚酸的含量差异显著(t检验,P<0.01);吴茱萸提取物中含有大量的皂苷类、黄酮类和酚酸类成分.  相似文献

In order to identify the optimum period of spring water-restrictive irrigation for winter wheat(Triticum aestivum L.) in the Haihe Plain, China and elucidate its effects on flag leaf senescence and yield formation, field experiments were conducted at the Xinji Experimental Station of Hebei Agricultural University from 2016 to 2019 by using different irrigation regimes in spring, including the conventional regime involving two irrigation periods(control(CK), the 3-leaf unfolding stage and the anthesis stage) and a series of single, restrictive irrigation regimes(SRI) comprising irrigation at the 3-leaf unfolding stage(3 LI), 4 LI, 5 LI, and 6 LI. There are five major findings:(1) The senescence(determined by the green leaf area, GLA) in the 4 LI treatment occurred moderately earlier than that in CK, showed no significant difference with that in 5 LI and 6 LI, and occurred significantly later than that in 3 LI.(2) Compared with other SRI treatments, the GLA value and photosynthetic rate in 4 LI were 14.82 and 20.1% higher, respectively. Microstructural analysis of flag leaf also revealed that the mesophyll cells and chloroplasts were irregularly arranged under drought stress in 3 LI and 6 LI; however, drought stress had minimal negative effects on the microstructure in 4 LI and 5 LI.(3) Postponed irrigation in spring could significantly increase superoxide dismutase(SOD) and catalase(CAT) activities in the early stage of grain filling; however, these activities would subsequently decrease. Among the four SRI treatments, the overall enzyme activities were the highest in 4 LI, and the combined malondialdehyde(MDA) content in flag leaves in 4 LI and 5 LI was 14.5% lower on average than that in 3 LI and 6 LI.(4) The soluble sugar(SS) and proline(Pro) contents in 4 LI were the highest among the four SRI treatments; however, they were lower than those in CK. The abscisic acid(ABA) hormone content in 4 LI and 5 LI was lower than that in 3 LI and 6 LI, respectively, suggesting a smaller drought stress effect in 4 LI and 5 LI.(5) In two growing seasons, there was a larger number of spikes per unit area in 4 LI(i.e., 13.4% higher than that in 5 LI and 6 LI) and the 1 000-grain weight in 4 LI was the highest among the four SRI treatments(i.e., 6.0% higher than that in the other three SRI treatments). Therefore, a single restrictive irrigation regime at the 4-leaf unfolding stage is recommended to be effective in slowing down the senescence process of flag leaves and achieving high yield.  相似文献

利用RT-PCR技术成功扩增出IBV AH1-99分离株的N基因全长cNDA,将其克隆到pMD18-T载体上。经酶切分析、PCR鉴定及核苷酸序列测定,成功获得该分离株N基因的重组质粒。序列分析结果表明,分离株N基因全长1 230个核苷酸,编码409个氨基酸。同部分IBV参考毒株相比,核苷酸同源性为85.8%~89.9%,氨基酸同源性为86.6%~91.0%,在进化关系树中,AH1-99与国内分离株X、LX4亲缘关系较近。  相似文献

AIM:To observe pathomorphological changes in cerebral cortex and hippocampus in the mouse with synthetic vascular dementia.METHODS:The synthetic vascular dementia model was produced in the mouse. Animals were killed 7 d, 15 d, and 30 d after the operation, brain tissues were removed and embedded in paraffin. Section of 8μm thickness were stained with hematoxylin-eosin(HE)and Nissl methods, and observed with light microscope.RESULTS:The cerebral cortex in the mouse became thinner on the seventh day, karyopyknosis in partial nervous cells was formed, the number of local neurons was reduced, sieve structure was observed, and glial cells pro liferated, with the similar results 15 d and 30 d afteroperation.Model mouseπs hippocampal cells in CA1 area were reduced and almost disappeared 30 d after operation.At the same time, glial cells were abundantly proliferated, tu bercles were formed.Cells in CA2, CA3 area were also reduced and hippocampal sclerosis occurred.CONCLUSION:Delayed necrosis of hippocampal pyramidal cells may be the pathological basis of ischemia cerebral vascular dementia.  相似文献
