Genistein, like other phytochemicals, has beneficial health effects, but its bioavailability is limited. This research studied the effect of complexation of genistein with starch on genistein bioavailability. Genistein release from these complexes was tested in vitro under simulated intestinal conditions and in vivo in rats fed high-amylose corn starch (HACS)-genistein complexes (experimental group) as compared to those fed a physical mixture of HACS and genistein (controls). In vitro results showed that genistein release is sustained and fits the normal transit time of food in the intestine. The genistein concentration in the plasma was twice as high in the experimental group versus controls; the genistein concentration in the urine was also higher in the experimental group but lower in the feces. These results indicate that starch-genistein complexes increase genistein bioavailability and suggest that starch can affect the bioavailability of additional food components.  相似文献

Purpose

This paper aims to elucidate urban development-induced processes affecting the sediment and the distribution of contaminating metals in a seasonal pond located in the highly populated Israeli Coastal Plain. The paper demonstrates how an integrated approach, including geochemical, sedimentological, geochronological, mathematical, historical, and geographical analyses, may decipher a complicated and dynamic metal pollution history in a sedimentary environment controlled by anthropogenic activity.

Materials and methods

Three short sediment cores were collected from the margins and center of a small urban pond (Dora, Netanya), located within the Israeli Coastal Plain. Profiles of grain size, organic matter (OM), trace metals (Pb, Zn, V, Ni, Cu, Cr and Co), Pb isotopic ratios, and 210Pb activities (center and southern cores) were determined and a geochemical mixing model was employed (southern core). The watershed contour was calculated, and aerial photos and satellite images were examined.

Results and discussion

Construction activities in the watershed were chronologically associated with coarse sediment transport and deposition in the margins of the pond. The upper sandy layers were superimposed on layers rich in fine particles and OM, high concentrations of trace metals, and with Pb isotopic composition of more recent petrol. In the 210Pb-dated southern core, deep metal-rich layers with petrol-related Pb isotopic ratios were inconsistent with metal emissions history. These findings point to mobility and migration of recent contamination metals through the coarse upper sediment layers and into deeper denser layers, confirmed also by a geochemical mixing model. Conversely, in the center of the pond, homogeneous fine particles were deposited with metal profiles consistent with regional emissions.

Conclusions

A small urban pond was found to provide an important case study for understanding heavy metal pollution records in highly populated regions. The margins of the pond depicted the surrounding urban development and the induced coarse sediment erosion, accompanied with post-depositional metal mobility. Due to the proximate developing residential areas, high metal concentrations accumulated in the margins, overshadowing regional atmospheric pollution levels recorded by sediment at the center of the pond.

‘Granny Smith’ apples are highly susceptible to superficial scald, a symptom of chilling injury. For many crops, low temperature storage results in oxidative stress and chilling injury, caused by increased production of superoxide anions which in turn leads to the generation of other dangerous reactive oxygen species (ROS). Application, prior to cold storage, of low oxygen (LO2, <0.5%) atmospheres, ethanol (<2% vapour) or 1-methylcyclopropene (1-MCP, 0.5 μL L−1) at 20 °C, was effective in reducing superficial scald in fruit following 24 weeks of cold storage. ROS levels were measured by confocal laser-scanning microscopy of apple peel treated with the fluorescent probe 2′,7′-dichlorodihydrofluorescein diacetate. In control fruit, ROS levels increased during cold storage and shelf-life and were very high after only 8 weeks, whereas in 1-MCP-, ethanol- and LO2-treated fruit, ROS levels remained low throughout storage. Gene-expression levels of ROS-scavenging enzymes were induced by the various pretreatments: catalase (MdCAT) was induced by LO2 treatment, whereas Mn superoxide dismutase (MdMnSOD) was induced by 1-MCP treatment. Polyphenol oxidase (MdPPO) gene expression levels were associated with scald symptom development and were highest in control fruit. Ethylene levels and expression of ethylene biosynthesis genes were correlated with α-farnesene levels and <alpha>-farnesene synthase (MdAFS) gene expression in the variously treated fruit. Accumulation of the α-farnesene oxidation product, 6-methyl-5-hepten-2-one (MHO), was highest in control fruit after 8 weeks, in accordance with ROS accumulation. The LO2 pretreatment mechanism might involve production of anaerobic metabolites, causing a delay in ethylene and α-farnesene biosynthesis and oxidation; this is different from the mechansism of action of 1-MCP, even though both consequently reduce ROS accumulation and scald symptoms.  相似文献

Complexes of amylose or high-amylose corn starch (HACS) with genistein were prepared by the acidification of an alkali solution to yield a V 6IotaIotaIota structure. The amylose-genistein complexes exhibited significantly higher genistein content (11 mg/100 mg of complex) than HACS-genistein complexes (9 mg/100 mg of complex). The effect of genistein on the amylose complexes was examined in different genistein-amylose ratios, and a model for genistein organization in the amylose complexes was suggested. The complexes were stable at different pH values, with <10% of the complexed genistein released, and were stable at 30 and 50 degrees C. Lower stability was observed at 80 degrees C as shown by the extensive release of genistein. All complexes showed high retention of genistein in simulated stomach conditions and released genistein upon digestion in pancreatin solution. It is therefore suggested that the complexes can be used as carriers for the slow release of genistein.  相似文献

Protocols for in vitro regeneration and production of in vitro-propagated plants and a transformation system were developed for Mirabilis jalapa (Nyctaginaceae). Among the types of explants and the different media tested, consistent shoot regeneration was obtained only from nodal segments grown in a regeneration medium consisting of Murshashige and Skoog medium supplemented with 2 mg l−1 6-benzyladenine, 2 mg l−1 zeatin and 1 mg l−1 indole acetic acid. Regeneration efficiency was dependent on the type of plant – white or pink flowers – used as the source of explants. Stable transformation was obtained following inoculation of nodal segments with Agrobacterium tumefasciens strain EHA105, which harbours the binary plasmid pAD1339 containing both nptII and gus genes under the control of the 35S promoter. Transformation was confirmed by PCR and Southern blot analysis of genomic DNA from mature regenerated plants. β-Glucuronidase (GUS) activity was observed only in tissues regenerated from in vitro-grown plants and not in tissues originating from greenhouse-grown plants. GUS expression was not uniform in regenerated leaves and showed a chimera pattern.  相似文献

Pomegranate juice is well known for its health beneficial compounds, which can be attributed to its high level of antioxidant activity and total polyphenol content. Our objective was to study the relationships between antioxidant activity, total polyphenol content, total anthocyanins content, and the levels of four major hydrolyzable tannins in four different juices/homogenates prepared from different sections of the fruit. To this end, 29 different accessions were tested. The results showed that the antioxidant activity in aril juice correlated significantly to the total polyphenol and anthocyanin contents. However, the homogenates prepared from the whole fruit exhibited an approximately 20-fold higher antioxidant activity than the level found in the aril juice. Unlike the arils, the antioxidant level in the homogenates correlated significantly to the content of the four hydrolyzable tannins in which punicalagin is predominant, while no correlation was found to the level of anthocyanins.  相似文献
