Enzymatic coupling of cholesterol intermediates to a mating pheromone precursor and to the ras protein

The post-translational processing of the yeast a-mating pheromone precursor, Ras proteins, nuclear lamins, and some subunits of trimeric G proteins requires a set of complex modifications at their carboxyl termini. This processing includes three steps: prenylation of a cysteine residue, proteolytic processing, and carboxymethylation. In the yeast Saccharomyces cerevisiae, the product of the DPR1-RAM1 gene participates in this type of processing. Through the use of an in vitro assay with peptide substrates modeled after a presumptive a-mating pheromone precursor, it was discovered that mutations in DPR1-RAM1 cause a defect in the prenylation reaction. It was further shown that DPR1-RAM1 encodes an essential and limiting component of a protein prenyltransferase. These studies also implied a fixed order of the three processing steps shared by prenylated proteins: prenylation, proteolysis, then carboxymethylation. Because the yeast protein prenyltransferase could also prenylate human H-ras p21 precursor, the human DPR1-RAM1 analogue may be a useful target for anticancer chemotherapy.     

Pressure regulation in the middle ear cavity of sea lions: a possible mechanism

The mucosa lining the cavity of the middle ear of sea lions contains a complex network of venous channels and sinuses. During dives the pressure within the middle ear may be equalized with that in the external auditory meatus either by the distention or depression of the mucosa due to the presence or absence of blood in the sinuses.     

Stereostructure of the archaebacterial C40 diol

The stereostructure of the archaebacterial C40 diol has been established as (3R,7R,11R,15S,18S,22R,26R,30R)-3,7,11,15,18,22,26,30- octamethyldotriacontane-1,32-diol by stereorational total synthesis. This provides the final evidence necessary to establish the structure of an archaebacterial membrane substance that is a 72-membered-ring tetraether with 18 stereocenters.     

Sonar Signals of the Sea Lion

Tape recordings were made of the underwater noises of captive sea lions swimming in a concrete pool at night. When approaching pieces of fish that were thrown into the water, the sea lions emitted trains of sound signals like those of the bat and the porpoise. A detailed analysis of these noises shows that they meet the criteria of a pulse-modulated sonar system and, in fact, reveal an amazing sophistication so far as echo ranging is concerned.     

Chimeras of two isoprenoid synthases catalyze all four coupling reactions in isoprenoid biosynthesis

The carbon skeletons of over 55,000 naturally occurring isoprenoid compounds are constructed from four fundamental coupling reactions: chain elongation, cyclopropanation, branching, and cyclobutanation. Enzymes that catalyze chain elongation and cyclopropanation are well studied, whereas those that catalyze branching and cyclobutanation are unknown. We have catalyzed the four reactions with chimeric proteins generated by replacing segments of a chain-elongation enzyme with corresponding sequences from a cyclopropanation enzyme. Stereochemical and mechanistic considerations suggest that the four coupling enzymes could have evolved from a common ancestor through relatively small changes in the catalytic site.     

Albumin and immunoglobulins G, A and M in aqueous humour from clinically normal equine eyes

The levels of albumin, immunoglobulin G (including IgG [T]), IgA and IgM in the serum and aqueous humour of 47 horses and ponies with no ophthalmic abnormalities were determined by single radial immunodiffusion. There was a linear relationship between serum and aqueous albumin levels. No relationship of serum and aqueous IgG levels was observed. IgA was detected in only one aqueous sample. IgM was not detected in aqueous humour from any eye in this study. The factors which may influence albumin and immunoglobulin levels in normal aqueous humour are discussed.
