Various factors affecting in vitro regeneration like different carbon sources, different gelling agents, and growth additives were assessed comprehensively for callus induction and plant regeneration for five Indian wheat cultivars using mature embryos as the explants for the first time. The tissue culture responses of cultivars WH-1105, HD-2967, and PBW-343 have not been reported earlier. Besides, the effect of different concentrations of the cytokinin, zeatin has also been optimized. Using the optimized factors, the efficiency of five different varieties, i.e., HD 2967, C 306, RAJ 3765, WH 1105, and PBW 343 was evaluated for regeneration. Modified MS basal medium containing dicamba reduced precocious germination of the embryo and induced embryogenic callus more efficiently. Removal of embryogenic calli from non-regenerable structures during early callus phase improved plant regeneration. These calli on zeatin (1.0 mgl-1) and dicamba (0.1 mgl-1) containing medium showed the highest regeneration frequency (98%) with a maximum of 8-9 shoots per calli. Maltose had the maximum callusing and regeneration percentage than other carbon sources. Various gelling agents did not have any significant difference on the regeneration. Of all the varieties, C-306 and HD-2967 were found to be more regenerative and can be used in transformation experiments. 相似文献

Spot blotch (SB), caused by Bipolaris sorokiniana, is a devastating disease of wheat globally, especially in South Asia and South America. Understanding the genetics of resistance to SB is important for developing breeding strategies to improve resistance. A panel of 301 genotypes from Afghanistan was phenotyped over two crop seasons using a mixture of virulent B. sorokiniana isolates and genotyped using DArTSeq to obtain genome-wide markers. Fifty genotypes (16.6%) showed disease scores less than the resistant control. Principal component analysis using the genotypic data clustered the genotypes into five different groups. Among models used for genome-wide association mapping, the multilocus mixed model, and fixed and random model circulating probability unification algorithms were most effective in identifying significant marker-trait associations (MTA). Twenty-five MTAs at p ≤ .001 were identified on chromosomes 1A, 1B, 1D, 2B, 2D, 3A, 3B, 4A, 5A, 5B, 6A, 7A, and 7D, indicating the quantitative nature of resistance to SB. Phenotypic variation explained by these markers ranged from 2.0% to 17.7%, and genomic regions on the chromosomes 1D, 2D, 3A, 3B, 4A, 5A, and 5B coincided with loci identified in previous studies. Three single nucleotide polymorphism (SNP) markers on chromosomes 1B (SNP 1113207) and 5A (SNPs 5411867 and 998276) were significant in both crop seasons as well as in the combined analysis across seasons. Marker 5411867 is close to Vrn-A1, shown to be associated with SB in previous studies. Furthermore, among known SB resistance genes, Sb2 on chromosome 5B was predicted to be significant in this panel. 相似文献

Identification of a small core germplasm set representing the available genetic diversity is essential for its proper evaluation and subsequent utilization in rice improvement programmes. For constituting a small diverse mini‐core panel of Indian rice germplasm, a representative set of 6912 accessions drawn based on their geographic origin from the whole rice germplasm collection available in the National Gene Bank was genotyped using 36 microsatellite markers. Automated fragment analysis of amplicons yielded a total of 435 alleles, with an average 12.4 and range of 3–29 alleles per locus. Polymorphism information content (PIC) ranged from 0.08 (RGNMS190) to 0.86 (RM552) with an average of 0.528. Based on genotyping data, a mini‐core consisting of 98 genotypes was identified. Ninety‐four per cent of the alleles present in the core set were present in the mini‐core. The identified small but diverse panel will be useful for further intensive trait‐specific evaluation and utilization in allele mining. 相似文献

We determined some basic properties of stars that produce spectacular gamma-ray bursts at the end of their lives. We assumed that accretion of the outer portion of the stellar core by a central black hole fuels the prompt emission and that fall-back and accretion of the stellar envelope later produce the plateau in the x-ray light curve seen in some bursts. Using x-ray data for three bursts, we estimated the radius of the stellar core to be approximately (1 - 3) x 10(10) cm and that of the stellar envelope to be approximately (1 - 2) x 10(11) cm. The density profile in the envelope is fairly shallow, with rho approximately r(-2) (where rho is density and r is distance from the center of the explosion). The rotation speeds of the core and envelope are approximately 0.05 and approximately 0.2 of the local Keplerian speed, respectively. 相似文献

Present study was undertaken to study the effect of cypermethrin on oxidative stress after chronic dermal application. The insecticide was applied dermally at 50 mg/kg body weight in different groups of Wistar rats of either sex weighing 150~200 g. Significant (p < 0.05) increase in catalase activity was observed after 30 days of exposure. However, the superoxide dismutase activity declined significantly after 60 days of exposure. The activity of glutathione peroxidase and blood glutathione levels declined significantly (p < 0.05) after 30 days of cypermethrin dermal application. However, the activity of glutathione S-transferase increased significantly (p < 0.05) in all groups after 60 days of dermal exposure. Significant increase in lipid peroxidation was observed from 30 days onwards and reached a peak after 120 days of application. 相似文献

In current study, dry–matter accumulation (DMA), pre– and post–anthesis nitrogen (N) accumulation, N translocation (NT) and dry–matter partitioning by sunflower seeds was investigated under three sowing dates (January 20, February 10 & March 2), two intra–row spacings (30 & 24 cm) and four nitrogen doses (0, 45, 60 & 75 kg ha–1) in two alluvial soils. Early sowing resulted in higher DMA and NT; leading to higher nitrogen use efficiency (NUE) that could be associated with higher pre–anthesis N accumulation. The closer intra–row spacing resulted in higher DMA by all plant parts except seed. Each graded N dose improved DMA, but improvement in dry–matter partitioning to seed was significant up to 60 and 75 kg N ha–1 during 2014 and 2015, respectively owing to higher NT under respective treatments. NUE was highest at 60 kg N ha–1 during both years. 相似文献

Fusarium head blight (FHB) is a major threat to wheat production globally, causing not only yield losses but also food and feed contamination. FHB research began at the International Maize and Wheat Improvement Center (CIMMYT) in the early 1980’s, and since then, large-scale FHB screening has been conducted to identify and incorporate new resistance genes into elite CIMMYT germplasm. Promising lines with good FHB resistance were regularly compiled as a Fusarium Head Blight Screening Nursery (FHBSN) and distributed worldwide. The first FHBSN was assembled in 1985, and the most recent two were the 13th and 14th FHBSN that were released in 2011 and 2012, respectively. Candidate lines for a FHBSN came mainly from different CIMMYT wheat breeding programs and were tested for three consecutive years before being included in an FHBSN. FHBSN screening was conducted under strictly standardized field conditions at El Batán, where CIMMYT headquarters is located, using artificial inoculation of F. graminearum strains, whose aggressiveness and DON chemotypes had been previously identified. FHB index was scored at 31 days after inoculation for all lines and DON concentration was measured only for elite lines in their 2nd and 3rd year of evaluation. Haplotyping is a new tool for genetic characterization of FHBSN entries and helps to identify new resistance sources with novel resistance genes and to better target crosses toward diversifying and/or pyramiding resistance. The 13th FHBSN was taken as an example in this paper to show the procedure and strategy for the development of new FHB resistant lines. 相似文献

Aim of the present study was in vitro expansion and characterization of caprine wharton’s jelly derived mesenchymal stem cells (cWJ-MSCs) to investigate their tissue healing potential in xenogenic animal model. Plastic adherent fibroblastoid cell populations with distinctive homogeneous morphology were isolated from caprine wharton’s jelly explants. These wharton’s jelly derived cells were found positive for the surface markers CD-73, STRO-1 and CD-105, whereas they were negative for hematopoetic stem cell marker CD-34. In vitro cultured cWJ-MSCs also showed differentiation properties into osteogenic, adipogenic and chondrogenic lineages as demonstrated by von Kossa, Oil Red- O and Alcian blue staining respectively, which was further confirmed and quantified by flow cytometric analysis. Furthermore, these well characterized cWJ-MSCs were evaluated for the wound-healing potential in full-thickness skin wounds in rabbit model for 28 days. Caprine WJ- MSCs treated skin wounds showed significantly (P?<?0.05) higher percentage of wound contraction especially at the 21st day post transplantation when compared to PBS treated control group animals. Further, we observed better healing potential of cWJ-MSCs in terms of histo-morphological evaluation, epithelialisation and collagenization with matured vascularization stage by day 28 as compared to control. In conclusion, cWJ- MSCs provide an alternative inexhaustible source of mesenchymal stem cells and also unravel new perspectives pertaining to the therapeutic use of these cells in different species. 相似文献

We sequenced the 29,751-base genome of the severe acute respiratory syndrome (SARS)-associated coronavirus known as the Tor2 isolate. The genome sequence reveals that this coronavirus is only moderately related to other known coronaviruses, including two human coronaviruses, HCoV-OC43 and HCoV-229E. Phylogenetic analysis of the predicted viral proteins indicates that the virus does not closely resemble any of the three previously known groups of coronaviruses. The genome sequence will aid in the diagnosis of SARS virus infection in humans and potential animal hosts (using polymerase chain reaction and immunological tests), in the development of antivirals (including neutralizing antibodies), and in the identification of putative epitopes for vaccine development. 相似文献
