A commercial high-methoxy citrus pectin was treated with a purified salt-independent pectin methylesterase (PME) isozyme isolated from Valencia orange peel to prepare a series of deesterified pectins. A series of alkali-deesterified pectins was also prepared at pH 10 under conditions permitting beta-elimination. Analysis of these pectins using high-performance size exclusion chromatography (HPSEC) with on-line multiangle laser light-scattering, differential viscometer, and refractive index (RI) detectors revealed no reduction in weight-average molecular weight (M(w); 150000) in the PME-treated pectin series, whereas a 16% reduction in intrinsic viscosity (IV) occurred below a degree of esterification (DE) of 47%. In contrast, alkali deesterification rapidly reduced both M(w) and IV to less than half of that observed for untreated pectin. PME treatment of a non-calcium-sensitive citrus pectin introduced calcium sensitivity with only a 6% reduction in the DE. Triad blocks of unesterified galacturonic acid were observed in (1)H nuclear magnetic resonance spectra of this calcium-sensitive pectin (CSP). These results demonstrate that the orange salt-independent PME isozyme utilizes a blockwise mode of action. This is the first report of the preparation of a CSP by PME treatment without significant loss of the pectin's M(w) due to depolymerization.  相似文献

An analytical survey of mutagens, nitrosamines, polychlorinated biphenyls, toxic elements, and gamma-emission, as well as the toxicologically protective constituents zinc, selenium, and vitamin C, in 48 pet foods was conducted. Aside from high concentrations of fluoride and iodide in some samples and the expectedly higher concentrations of mercury and selenium in certain cat foods containing fish, the samples were notably free of the other toxic constituents. Direct-acting and promutagens and nitrosamines were not detectable in any of the samples. gamma-Emission was very low in all of the foods. Polychlorinated biphenyls were only detected in one cat food.  相似文献

Global structure of microwave-assisted flash-extracted sugar beet pectin   总被引:3,自引:0,他引:3  
The global structure of microwave-assisted flash-extracted pectins isolated from fresh sugar beet pulp has been studied. The objective was to minimize the disassembly and possibly the degradation of pectin molecules during extraction. These pectins have been characterized by high-performance size exclusion chromatography with light scattering, viscometric detection, and atomic force microscopy (AFM). Analysis of molecular parameters was performed on 15 and 8 microm size column packings. Samples analyzed with 15 microm packing gave weight-average molar masses that ranged from 532,000 to 1.2 million Da, radii of gyration from about 35 to 51 nm, polydispersities from 1.78 to 2.58, intrinsic viscosities from about 3.00 to 4.30 dL/g, and recoveries from 8.40 to 14.81% of dry weight. Chromatography revealed that a bimodal distribution of high molar mass spherical particles and lower molar mass coils was obtained. AFM images of pectin corroborated this conclusion and further revealed that these strands and spherical particles were integrated into networks. It is demonstrated that microwave-assisted extraction of sugar beet pulp under moderate pressure and at relatively low temperature could extract under acid conditions high molar mass, moderate-viscosity pectin in minutes rather than hours as required by conventional heating.  相似文献

The analysis of exhaled breath is a potentially useful method for application in veterinary diagnostics. Breath samples can be easily collected from animals by means of a face mask or collection chamber with minimal disturbance to the animal. After the administration of a 13C-labelled compound the recovery of 13C in breath can be used to investigate gastrointestinal and digestive functions. Exhaled hydrogen can be used to assess orocaecal transit time and malabsorption, and exhaled nitric oxide, carbon monoxide and pentane can be used to assess oxidative stress and inflammation. The analysis of compounds dissolved in the aqueous phase of breath (the exhaled breath condensate) can be used to assess airway inflammation. This review summarises the current status of breath analysis in veterinary medicine, and analyses its potential for assessing animal health and disease.  相似文献

The aim of the study was to quantify peach scab (Fusicladosporium carpophilum) lesion distribution relative to the point of maximum lesion number on the fruit surface, the relationship between lesion count and distance from the point of maximum lesion density, and establish whether the distribution of lesions was consistent with a splash dispersed pathogen, and to assess the effect of lesion number on fruit size. Fruit of four cultivars, Jerseyqueen, Jefferson, BY07-6428r and Dixiland were collected and the fruit (assumed spherical) sliced taking three horizontal planes across the axis from the point of maximum disease, such that each horizontal zone (Z1-Z4) had the same vertical height, and thus equal surface areas. Lesion counts were analysed using general linear modeling with a Poisson distribution and a log-link function. Zones on the fruit had different numbers of lesions (P < 0.0001), with most lesions found on Z1. Cvs differed in the number of lesions per fruit (P = 0.0042–<0.0001). An analysis of covariance showed that although fruit size varied among most cvs (P = 0.1614–<0.0001), the number of lesions on a fruit did not affect fruit size (P = 0.5654). Measurements of the point of maximum disease relative to the peduncle-flower scar axis of the fruit suggest that fruit are not always held upright when infection occurs, such that up to 40% of fruit showed maximum infection at an angle >90° to the peduncle. This pattern of disease is consistent with observations of the splash-borne nature of conidia, with the most exposed, easily wetted, uppermost portion of fruit developing most disease.  相似文献

Error in estimates of plant disease severity occur and standard area diagrams (SADs) help improve accuracy and reliability. The effects of diagram number in SADs are unknown. The objective of this study was to compare estimates of pecan scab severity made without SADs, and using three‐, five‐, seven‐ or 10‐diagram SADs. Disease severity was estimated to the nearest percent (NPE), or classified to the closest reference diagram value using a scale. Twelve raters assessed 20 images of scabbed pecan valves with and without the SADs using NPEs and the scale method (values were converted to midpoints prior to analysis). Increases in diagram number using NPEs did not necessarily result in more accurate or reliable estimates. Inter‐rater reliability was positively correlated with number of diagrams using NPEs (= 0·3288 (< 0·0001)) or a scale (= 0·2803 (< 0·0001)). The least accurate estimates improved the most using SADs with NPEs, but the gain did not relate to number of diagrams; as few as three diagrams reduced error of inaccurate estimates as much as five, seven or 10 diagrams. When used as a scale, only estimates made with 10‐diagram SADs had similar accuracy and inter‐rater reliability to estimates made using SADs with NPEs. Maximum disease severity and the disease severity range for the pathosystem and the SADs are important factors and will probably influence resulting accuracy and reliability. The ramifications of diagram numbers in SADs and assessment methods on accuracy and reliability of disease estimates are discussed.  相似文献

Pectin gels were induced by monovalent salts (0.2 M) concurrently with deesterification of high methoxy pectin using a salt-independent orange pectin methylesterase (PME). Constant pH was maintained during deesterification and gelation. If salt or PME was absent, the pectin did not form a gel. The gel strength was influenced by both pH and species of monovalent cation. At pH 5.0, the pectin gel induced by KCl was significantly stronger than the NaCl-induced gel. In contrast, a much stronger gel was produced in the presence of NaCl as compared to KCl at pH 7.0. LiCl did not induce pectin gelation at either pH. Molecular weights of pectins increased from 1.38 x 10(5) to 2.26 x 10(5) during NaCl-induced gelation at pH 7. One proposal to explain these pectin molecular weight changes is a hypothetical PME transacylation mechanism. However, these pectin molecular weight changes can also be explained by metastable aggregation of the enzymatically deesterified low methoxy pectin. We postulate that gelation was induced by a slow deesterification of pectin under conditions that would normally salt out (precipitate) low methoxy pectin in the absence of PME.  相似文献
