Sir2 (silent information regulator 2) is a nicotinamide adenine dinucleotide-dependent deacetylase required for longevity due to calorie restriction in yeast and Drosophila. In mammals, calorie restriction induces a complex pattern of physiological and behavioral changes. Background

In the less-sensitive mouse model, Shiga toxin-producing Escherichia coli (STEC) challenges result in shedding that reflect the amount of infection and the expression of virulence factors such as Shiga toxins (Stx). The purpose of this study was to characterize the contribution of STEC diversity and Stx expression to shedding in beef feeder calves and to evaluate the effectiveness of a prebiotic, Celmanax®, to alleviate STEC shedding. Fecal samples were collected from calves at entry and after 35 days in the feedlot in spring and summer. STECs were evaluated using selective media, biochemical profile, serotyping and Stx detection. Statistical analysis was performed using repeated measures ANOVA and logistic regression.

Results

At entry, non-O157 STEC were dominant in shedding calves. In spring, 21%, 14% and 14% of calves acquired O157, non-O157 and mixed STEC infections, respectively. In contrast, 45%, 48% and 46% of calves in summer acquired O157, non-O157 and mixed STEC infections, respectively. Treatment with a prebiotic, Celmanax®, in spring significantly reduced 50% of the O157 STEC infections, 50% of the non-O157 STEC infections and 36% of the STEC co-infections (P = 0.037). In summer, there was no significant effect of the prebiotic on STEC infections. The amount of shedding at entry was significantly related to the number and type of STECs present and Stx expression (r2 = 0.82). The same relationship was found for shedding at day 35 (r2 = 0.85), but it was also related to the number and type of STECs present at entry. Stx - producing STEC infections resulted in 100 to 1000 × higher shedding in calves compared with Stx-negative STECs.

Conclusions

STEC infections in beef feeder calves reflect the number and type of STECs involved in the infection and STEC expression of Stx. Application of Celmanax® reduced O157 and non-O157 STEC shedding by calves but further research is required to determine appropriate dosages to manage STEC infections.  相似文献

Context

Animal movements are inherently linked to landscape structure. Understanding this relationship for highly-mobile species requires documenting their responses to spatiotemporal variability of resources. To that end, characterizing movement behaviors and resource distributions using the principles of habitat connectivity facilitates coordinated landscape planning efforts within highly modified landscapes.

Objectives and methods

We tracked locations and movements for 156 dunlin (Calidris alpina) and 109 long-billed dowitchers (Limnodromus scolopaceus) overwintering in two regions with distinct water distributions in California’s Central Valley. We then compared residency rates, functional connectivity to other regions, and associations between movement distances and average habitat availability and structural connectivity of habitat at multiple temporal and spatial scales.

Results

A widespread yet highly variable regional water distribution was associated with lower residency rates and substantially higher functional connectivity to nearby regions when compared to a stable regional water distribution characterized by a large, contiguous wetland complex. Longer movements were associated with decreasing average availability and spatial aggregation of surface water. Movement models suggested shorebirds primarily responded to habitat availability at smaller scales (<?10 km) and structural connectivity at larger scales (≥?10 km).

Conclusions

Differences in movement behaviors suggested that wintering shorebirds will avoid long distance movements and remain resident within a wetland region when possible. Conservation and management efforts should reliably flood individual wetlands and agricultural lands from November to April and prioritize locations that maximize structural wetland connectivity and limit spatiotemporal variability of surface water throughout the Central Valley.

BACKGROUND: Bromus rigidus is a common weed species that has increased in cropping fields owing to limited control options. During a random field survey in Western Australia, six B. rigidus populations that had survived in‐crop weed control programmes were collected. The study aimed to determine the resistance profile of these six populations. RESULTS: Based on dose–response studies, all six B. rigidus populations had a low‐level resistance to sulfosulfuron and sulfometuron (both sulfonylurea herbicides) while remaining susceptible to herbicides with other modes of action. ALS in vitro activity assays revealed no differences in enzyme sensitivity between susceptible and resistant populations, while the use of malathion (a cytochrome P450 inhibitor) in combination with sulfosulfuron caused the resistant populations to behave like the susceptible population. CONCLUSION: This study established that these six B. rigidus populations have a low‐level resistance to the ALS‐inhibiting sulfonylurea herbicides, but are able to be controlled by other herbicide modes of action. The low‐level, malathion‐reversible resistance, together with a sensitive ALS, strongly suggest that a non‐target‐site enhanced metabolism is the mechanism of resistance. Previous studies have made some progress with the use of microbial community properties as assessment criteria for rehabilitation success of post-mining areas. Currently, there is a need for reference ranges of specific properties in rehabilitated post-mining sites to make this approach more practical. The aim of this investigation was to compare assessment parameters indicative of microbial community function (enzymatic assays) and structure (phospholipid fatty acid (PLFA) analysis) in rehabilitated asbestos and coal discard sites and to establish ranges of minimum and maximum values for these parameters in both types of sites. The range established for dehydrogenase activity in coal discard sites was 24.3–339.5 μg INF g−1 2 h−1 and for asbestos 44.5–544.6 μg INF g−1 2 h−1. Ranges were also established for β-glucosidase, urease, acid phosphatase and alkaline phosphatase. Complete PLFA profiles were determined and ranges established for major PLFA groups and ratios in both types of discard. From the PLFA profiles, viable microbial biomass was determined as 6,080–29,851 and 8,128–47,242 pmol g−1 dry weight for the coal and asbestos discard sites, respectively. While similar ranges were observed for both types of discard, a canonical correspondence analysis that accounts for functional and structural characteristics showed that sites clustered according to the origin of the samples.  相似文献

We describe an experiment where cattle urine was applied at a rate of 420 kg N ha−1, equivalent to 10 L m−2, to mesocosm swards of a high lipid genetically modified perennial ryegrass line (HME) and its wild type (WT). We measured N pools and fluxes in the plant and soil, soil microbial populations and N2O production. HME plants produced 21% greater biomass than WT (p = .02), resulting in greater N uptake (27% higher in HME, p = .05). Urine N recovery in total plant biomass during the experiment in HME and WT swards were 54.7% and 33.9% respectively. The nitrification potential of soil was significantly lower (p = .01) in HME than WT. Partial least square-discriminant analysis using microbial gene abundance data indicated that HME and WT plant growth induced distinct microbial populations in the soil. These differences in plant soil microbial interactions between HME and WT swards resulted in significantly lower N2O emissions from the HME sward. Total N2O emissions over the 4 weeks after urine application was 38% lower (p < .03) in HME swards than in WT swards.
