In recent decades, harmful algal blooms (HABs) producing paralytic shellfish toxins (including saxitoxin, STX) have become increasingly frequent in the marine waters of Alaska, USA, subjecting Pacific Arctic and subarctic communities and wildlife to increased toxin exposure risks. Research on the risks of HAB toxin exposures to marine mammal health commonly relies on the sampling of marine mammal gastrointestinal (GI) contents to quantify HAB toxins, yet no studies have been published testing the stability of STX in marine mammal GI matrices. An understanding of STX stability in test matrices under storage and handling conditions is imperative to the integrity of toxin quantifications and conclusions drawn thereby. Here, STX stability is characterized in field-collected bowhead whale feces (stored raw in several treatments) and in fecal extracts (50% methanol, MeOH) over multiple time points. Toxin stability, as the percent of initial concentration (T0), was reported for each storage treatment and time point. STX was stable (mean 99% T0) in 50% MeOH extracts over the 8-week study period, and there was no significant difference in STX concentrations quantified in split fecal samples extracted in 80% ethanol (EtOH) and 50% MeOH. STX was also relatively stable in raw fecal material stored in the freezer (mean 94% T0) and the refrigerator (mean 93% T0) up to 8 weeks. STX degraded over time in the room-temperature dark, room-temperature light, and warm treatments to means of 48 ± 1.9, 38 ± 2.8, and 20 ± 0.7% T0, respectively, after 8 weeks (mean ± standard error; SE). Additional opportunistically analyzed samples frozen for ≤4.5 years also showed STX to be relatively stable (mean 97% T0). Mean percent of T0 was measured slightly above 100% in some extracts following some treatments, and (most notably) at some long-term frozen time points, likely due to evaporation from samples causing STX to concentrate, or variability between ELISA plates. Overall, these results suggest that long-term frozen storage of raw fecal samples and the analysis of extracts within 8 weeks of extraction in 50% MeOH is sufficient for obtaining accurate STX quantifications in marine mammal fecal material without concerns about significant degradation.  相似文献

The sea louse, Lepeophtheirus salmonis , is an ectoparasitic copepod of Atlantic salmon, Salmo salar L., capable of causing severe damage. This study was conducted to examine the physiological response of salmon to the stress of sea lice infestation. Smoltified salmon were acclimatized in 30‰ saltwater and exposed to high levels of lice infestation. The number of copepods per fish ranged from 15 to 285, with a mean of 106. The infested salmon were sampled six times over the 29-d experimental duration and examined for alterations in the primary and secondary stress indicators, including plasma concentrations of cortisol, glucose, electrolytes, thyroid hormones T3 and T4, as well as the haematocrit level. The results were examined for correlations between the stress indicators, the number of copepods per fish and the life stage of the copepods. The presence of L. salmonis elevated stress indicators in relation to the specific sea lice stage. By day 21, both cortisol (mean 63.1 nmol L−1 controls: 179.8 nmol L−1 for parasitized) and glucose (mean 3.545 mmol L−1 controls: 4.567 mmol L−1 for parasitized) levels were significantly increased due to the presence of the lice. This was believed to be a direct result of the sea lice development into the larger life stages, thus increasing the level of host damage.  相似文献

Poly(ADP-ribose) polymerase-1 (PARP-1) protects the genome by functioning in the DNA damage surveillance network. PARP-1 is also a mediator of cell death after ischemia-reperfusion injury, glutamate excitotoxicity, and various inflammatory processes. We show that PARP-1 activation is required for translocation of apoptosis-inducing factor (AIF) from the mitochondria to the nucleus and that AIF is necessary for PARP-1-dependent cell death. N-methyl-N'-nitro-N-nitrosoguanidine, H2O2, and N-methyl-d-aspartate induce AIF translocation and cell death, which is prevented by PARP inhibitors or genetic knockout of PARP-1, but is caspase independent. Microinjection of an antibody to AIF protects against PARP-1-dependent cytotoxicity. These data support a model in which PARP-1 activation signals AIF release from mitochondria, resulting in a caspase-independent pathway of programmed cell death.  相似文献

Those involved with pollinator risk assessment know that agricultural crops vary in attractiveness to bees. Intuitively, this means that exposure to agricultural pesticides is likely greatest for attractive plants and lowest for unattractive plants. While crop attractiveness in the risk assessment process has been qualitatively remarked on by some authorities, absent is direction on how to refine the process with quantitative metrics of attractiveness. At a high level, attractiveness of crops to bees appears to depend on several key variables, including but not limited to: floral, olfactory, visual and tactile cues; seasonal availability; physical and behavioral characteristics of the bee; plant and nectar rewards. Notwithstanding the complexities and interactions among these variables, sugar content in nectar stands out as a suitable quantitative metric by which to refine pollinator risk assessments for attractiveness. Provided herein is a proposed way to use sugar nectar concentration to adjust the exposure parameter (with what is called a crop attractiveness factor) in the calculation of risk quotients in order to derive crop‐specific tier I assessments. This Perspective is meant to invite discussion on incorporating such changes in the risk assessment process. © 2016 The Authors. Members of the DExH/D superfamily of nucleic acid-activated nucleotide triphosphatases are essential for virtually all aspects of RNA metabolism, including pre-messenger RNA splicing, RNA interference, translation, and nucleocytoplasmic trafficking. Physiological substrates for these enzymes are thought to be regions of double-stranded RNA, because several DExH/D proteins catalyze strand separation in vitro. These "RNA helicases" can also disrupt RNA-protein interactions, but it is unclear whether this activity is coupled to duplex unwinding. Here we demonstrate that two unrelated DExH/D proteins catalyze protein displacement independently of duplex unwinding. Therefore, the essential functions of DExH/D proteins are not confined to RNA duplexes but can be exerted on a wide range of ribonucleoprotein substrates.  相似文献

Little is known about the effects of enriched CO(2) atmospheres, which may exist in the next century, on natural plant-insect herbivore interactions. Larvae of a specialist insect herbivore, Junonia coenia (Lepidoptera: Nymphalidae), were reared on one of its host plants, Plantago lanceolata (Plantaginaceae), grown in either current low (350 parts per million) or high (700 ppm) CO(2) environments. Those larvae raised on high-CO(2) foliage grew more slowly and experienced greater mortality, especially in early instars, than those raised on low-CO(2) foliage. Poor larval performance on high-CO(2) foliage was probably due to the reduced foliar water and nitrogen concentrations of those plants and not to changes in the concentration of the defensive compounds, iridoid glycosides. Adult pupal weight and female fecundity were not affected by the CO(2) environment of the host plant. These results indicate that interactions between plants and herbivorous insects will be modified under the predicted CO(2) conditions of the 21st century.  相似文献
