In developed countries, most of hepatitis E human cases are of zoonotic origin. Swine is a major hepatitis E virus (HEV) reservoir and foodborne transmissions after pork product consumption have been described. The risk for HEV-containing pig livers at slaughter time is related to the age at infection and to the virus shedding duration. Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) is a virus that impairs the immune response; it is highly prevalent in pig production areas and suspected to influence HEV infection dynamics. The impact of PRRSV on the features of HEV infections was studied through an experimental HEV/PRRSV co-infection of specific-pathogen-free (SPF) pigs. The follow-up of the co-infected animals showed that HEV shedding was delayed by a factor of 1.9 in co-infected pigs compared to HEV-only infected pigs and specific immune response was delayed by a factor of 1.6. HEV shedding was significantly increased with co-infection and dramatically extended (48.6 versus 9.7 days for HEV only). The long-term HEV shedding was significantly correlated with the delayed humoral response in co-infected pigs. Direct transmission rate was estimated to be 4.7 times higher in case of co-infection than in HEV only infected pigs (0.70 and 0.15 per day respectively). HEV infection susceptibility was increased by a factor of 3.3, showing the major impact of PRRSV infection on HEV dynamics. Finally, HEV/PRRSV co-infection – frequently observed in pig herds – may lead to chronic HEV infection which may dramatically increase the risk of pig livers containing HEV at slaughter time.

Raphanus raphanistrum (Brassicaceae) is considered amongst the world's worst agricultural weeds. We address critical issues in its management by studying the pathway of colonisation at local scales. For this, we assessed the small‐scale spatial genetic structure of 231 samples collected from three different sites across the Cape Floristic Region, South Africa, using 11 nuclear microsatellite markers. Although natural pollen and seed dispersal were expected to be restricted, we found no significant relationship between genetic and geographical distance within sites. Instead, our results suggest that R. raphanistrum had colonised new habitats via jump dispersal, rather than through natural diffusive dispersal at local scales. We did not find evidence for road verges as dispersal corridors, as evidenced by a lack of isolation‐by‐distance at local scales. Instead, the absence of spatial genetic structure suggests that R. raphanistrum had rapidly spread throughout its current range, possibly facilitated by human‐mediated actions. Management plans addressing containment or suppression of the weedy species R. raphanistrum (and possibly other weedy species) should take the high degree of connectivity between distant geographical localities into account.  相似文献

Hepatitis E virus (HEV) can cause enterically-transmitted hepatitis in humans. The zoonotic nature of Hepatitis E infections has been established in industrialized areas and domestic pigs are considered as the main reservoir. The dynamics of transmission in pig herds therefore needs to be understood to reduce the prevalence of viremic pigs at slaughter and prevent contaminated pig products from entering the food chain. An experimental trial was carried out to study the main characteristics of HEV transmission between orally inoculated pigs and naïve animals. A mathematical model was used to investigate three transmission routes, namely direct contact between pigs and two environmental components to represent within-and between-group oro-fecal transmission. A large inter-individual variability was observed in response to infection with an average latent period lasting 6.9 days (5.8; 7.9) in inoculated animals and an average infectious period of 9.7 days (8.2; 11.2). Our results show that direct transmission alone, with a partial reproduction number of 1.41 (0.21; 3.02), can be considered as a factor of persistence of infection within a population. However, the quantity of virus present in the environment was found to play an essential role in the transmission process strongly influencing the probability of infection with a within pen transmission rate estimated to 2 ⋅ 10− 6g ge− 1d− 1(1 ⋅ 10− 7; 7 ⋅ 10− 6). Between-pen environmental transmission occurred to a lesser extent (transmission rate: 7 ⋅ 10− 8g ge− 1d− 1(5 ⋅ 10− 9; 3 ⋅ 10− 7) but could further generate a within-group process. The combination of these transmission routes could explain the persistence and high prevalence of HEV in pig populations.  相似文献

In a context of the global major changes, it is mandatory to enlarge the range of crops supporting food security and pay great attention to neglected and underutilized species. However, basic knowledge of the biology of many neglected and underutilized species is still lacking to increase their yields. In this study, the mating system of white fonio [Digitaria exilis (Kippist.) Stapf], a West African minor and promising cereal, is assessed. Progenies arrays from both homozygous and heterozygous mothers were genotyped with microsatellites markers. The rate of genotyping errors in the experiments was assessed and a likelihood framework was used to determine the probability of different mating systems: outcrossing, self-fertilization and apomixis. The results suggested that white fonio has a highly selfing reproductive system with a possible outcrossing rate of 1.7%. Understanding the reproduction system of white fonio opens up opportunities for more effective breeding programs and a wider use of this cereal for food security improvement.  相似文献

Significant efforts have been undertaken in West Africa to increase biofuel production with the expectation to alleviate the dependency on fossil energies and to reduce rural poverty by diversifying cultivated crops. In this context, Jatropha curcas L., a shrub belonging to Euphorbiaceae family, has gained great interest because of its oil which can be converted to biodiesel. It is also highly adaptable to marginal soils due to its drought-tolerant characteristics. Characterisation of J. curcas germplasm in Senegal could be an important input for its better management and in identifying genotypes that could be used in breeding program. Genetic diversity of 103 accessions including 82 accessions from different agro ecological zones in Senegal and 21 exotic accessions was assessed through 33 microsatellite markers. All the markers gave amplifications at the expected band size. Only one microsatellite marker, JCT17, was polymorphic showing 3 alleles and allows distinguishing 2 accessions from Burkina Faso. The surprisingly low level of genetic variation might be because introduction of J. curcas in Senegal seems to have been done from one or a few origins and the species has not regained genetic diversity since then due to vegetative propagation. Cultivation of J. curcas at large scale may face to vulnerability to pests and require many cautions. They are necessity to widen the genetic base of J. curcas in Senegal via new introductions from its centre of origin.  相似文献
