Due to the potential for anti-erythrocyte membrane antibodies as possible enhancers of erythrocyte destruction, the presence of serum anti-erythrocyte membrane antibodies in 31 dogs with Babesia gibsoni infection admitted to a veterinary hospital was investigated by an enzyme linked immunosorbent assay (ELISA) and immunoblotting analyses. This infection resulted in an increase of anti-erythrocyte membrane antibodies in 84% (IgG) and 74% (IgM) of 31 infected dogs, respectively. This was confirmed by the similarity in the protein profiles of the erythrocyte membrane antigens immunoblotted with rabbit antiserum to dog erythrocyte membrane antigens and infected dog serum. These results suggest the production of anti-erythrocyte membrane antibodies was induced by B. gibsoni infection.  相似文献

Fibrotic degeneration was examined in the kidneys of ICR-derived glomerulonephritis (ICGN) mice, a novel inbred mouse line with a hereditary nephrotic syndrome of unknown etiology considered to be a good model of human idiopathic nephrotic syndrome. In the present study, we histochemically revealed changes in accumulation of extracellular matrix (ECM) components and in localization of integrins, cellular receptors for ECM, in the kidneys of ICGN mice with the progression of renal failure. Excessive accumulation of basement membrane (laminin and collagen IV) and interstitial (type III collagen) ECM components were demonstrated in the glomeruli and tubulointerstitum of ICGN mice. Marked deposition of type I collagen and tenascin was seen only in the glomeruli of ICGN mice but not in those of ICR mice as normal controls. Increased expression of integrin alpha1-, alpha2-, alpha5- and beta1-subunits in glomeruli with fibrotic degeneration and abnormal distribution of alpha6-subunit were noted in the kidneys of ICGN mice. Excessive laminin, a ligand of alpha6beta1-integrin, was demonstrated on the tubular basement membrane, but alpha6-subunit diffusely disappeared on the basal side of the tubular epithelial cells. We presumed that abnormal integrin expression in renal tubules causes epithelial cell detachment, and consequently tubular nephropathy, and results in disorder of ECM metabolism causing excessive accumulation of ECM components in the kidneys of ICGN mice.  相似文献

Chuzan virus agglutinated erythrocytes of several species of animals including bovine. The hemagglutinating (HA) activity against bovine erythrocytes was dependent on NaCl molarity and was expressed best at 0.6 M, but it was independent of pH and temperature. Three strains of Chuzan virus isolated from 2 cows and a pool of culicoides midges had indistinguishable HA antigenicity. All cattle infected with the virus developed high titers of hemagglutination inhibiting (HI) antibody which changed in parallel with neutralizing (NT) antibody titers. Correlation between HI and NT antibodies was very high and the antibodies persisted for one year or more. Therefore it was concluded that the HI test is applicable for survey of Chuzan virus infection among cattle in place of the NT test.  相似文献

To identify possible sites of viral attenuation, the complete nucleotide sequences of two isolates of Zucchini yellow mosaic virus (ZYMV) were determined; a severe isolate Z5-1 and an attenuated isolate from Z5-1 (designated ZYMV-2002). The viral genome of both isolates consisted of 9593 nucleotides in size and contained an open reading frame encoding a single polyprotein of 3080 amino acids. Comparison of the nucleotide sequences for Z5-1 and ZYMV-2002 revealed 14 nucleotide mutations, resulting in seven amino acid substitutions with four in the HC-Pro region, two in the CI region, and one in the NIb region. These results provide a genetic basis for future manipulation of the ZYMV reverse genetics system. The water flow in larval rearing tanks has been indicated to cause mass mortality of the seven-band grouper Epinephelus septemfasciatus larvae. Therefore, a new aerating method was tested in an actual scale intensive rearing tank (8.0 m in diameter, 1.87 m of water depth, 100 m3 of volume), in which an aerator was positioned at the center of the rearing tank surrounding cylindrical drain (1.2 m in diameter) to generate the flow field, and seven larval rearing trials were performed. The survival rate with the former aeration methods were compared, in which several aerators were located in the rearing tank. The survival rate at 10 days after hatching with the new aeration method (61.5±5.1%, n=7) was approximately three times higher than the former methods (21.2±13.7%, n=6). The flow environment of rearing tanks was also examined by quantifying the flow field, and the relationship between the flow field in the rearing tank, behavior of larvae and survival discussed. It was confirmed that the vertical circulating flow was observed in rearing tanks, and determined effectively the survival and the behavior of grouper larvae in patchiness.  相似文献

To evaluate the impact of temporal variation of primary productivity on the recruitment of Japanese sardine (Sardinops melanostictus) in the Sea of Japan, the phenology of sea surface phytoplankton abundance was estimated from 8 day multiple satellite (SeaWiFS, MODIS‐Aqua, MERIS, and VIIRS) derived sea surface chlorophyll (SSChl) a concentrations from January 1998 to December 2015. Because relationships between SSChl a and in situ chlorophyll a concentrations were significantly different among periods based on the satellite combinations used, maximum and minimum SSChl a concentrations of 1 year were relativized as 1 and 0, respectively. Spatio‐temporal variation of relativized SSChl a concentrations was determined by using empirical orthogonal function (EOF) analysis. Scores in the first EOF mode denoted the basin‐scale variations of SSChl a concentrations in the Sea of Japan, and the major peak from the end of February to the end of May displayed the spring bloom. The logarithm of recruitment per spawner (LNRPS) for sardine was positively affected by delays in the start and end dates of the spring phytoplankton bloom. The delay of the date of the lowest sea surface temperature contributed to the delay of the end of the spring bloom during the period 1998–2015 and elevated the LNRPS during the period 1982–2015. Sardine spawns in the southern Sea of Japan from April to May, hence, delays of the spring bloom prolonged its overlap with sardine larval periods, and thus improved the recruitment of Japanese sardine in the Sea of Japan.  相似文献

Increasing food loss and waste (FLW) is a global problem, and efforts are being made to use waste food as potential livestock feed material. The amount of self-supplied feed is lower in Japan than in other countries, and the government recommends FLW use for animal feed. Sake (Japanese rice wine) is a traditional alcoholic beverage. During the sake manufacturing process, large amounts of squeezed solids or “lees” (sake lees) are generated. Sake lees are nutritious and functional, but are prone to spoilage. In this study, we investigated whether sake lees should be mixed with animal feed immediately or after drying. To assess the usefulness of sake lees as a poultry feed ingredient and determine the effect of sake lees on intestinal immunity, we performed a feeding trial with three treatments: a raw sake lees (RSL) diet, dried sake lees (DSL) diet, and control diet. Three-week-old broilers were fed these diets (n=8 per group) for two weeks. We then calculated feed efficiency and performed RT-qPCR to assess the effects of diet on intestinal immunity. The growth performance in the RSL diet group was equivalent to that in the control diet group. The DSL diet became difficult for broilers to eat, resulting in decreased growth performance. In the ileum of RSL-diet broilers, the mRNA expression levels of TGF-β1 and avian β-defensin (AvBD)12 were significantly increased compared to those of control diet broilers (p<0.05), and a significant correlation was observed between the two genes (p<0.05). Our results indicated that sake lees should not be dried and should be mixed immediately with feed, and this sake lees when fed to chicken activates the intestinal immunity. However, sake lees have a lower fat content than corn, and it is thus important to combine sake lees with high-energy feed.  相似文献
