This report describes the characterisation of a monoclonal antibody (mAb), AB6, which recognises specifically a cluster of canine leukocyte surface molecules. The immunogen used for obtaining the AB6 mAb was a lysate of canine peripheral blood mononuclear cells (PBMC). This novel mAb belongs to the IgG2a isotype, and reacted in Western blot with four different canine leukocyte glycoproteins with apparent molecular weights of 180, 190, 205 and 220 kDa. The AB6 mAb recognised the majority of canine peripheral blood leukocytes as determined by flow cytometry (97%). It also exhibited a broad reactivity pattern against lymphoid and myeloid cells, inhibited the proliferation of mitogen-stimulated canine PBMC and did not recognise human PBMC and murine splenocytes. The biochemical properties, cell and tissue specificity, and in vitro biological activity of the AB6 mAb indicate that it recognises a canine CD45 homologue. The mAb could become a valuable diagnostic and research tool for the evaluation of immune functions in dogs. 相似文献

In Southern Italy the paucity of water is a very important problem and due to the fundamental importance of the water for the processing tomato, in this study the influence of irrigation on processing tomato quality was investigated in the years 2002 and 2003. A hybrid processing tomato (Lycopersicon esculentum L. Mill cv. Ability) was treated with different water regimes obtained combining amount of water and irrigation interval. The effects of the trials were evaluated taking into account the physical and chemical characteristics of the fruits, as well as the content in antioxidants moieties. Furthermore, the relationship among all the quality parameters and the seasonal amount of irrigation water was evaluated.The results, comparing different water regimes obtained combining amount of water and irrigation interval, gave useful indications on the possibility to improve nutritional tomato quality by reducing irrigation water during tomato cropping. In particular, relatively to the Mediterranean areas of tomato cultivation and to the texture of the soil considered in this study, the best compromise between quality and quantity of the processing tomato fruit was achieved both with the extension of the irrigation interval (L40 or L60) and with the limitation of irrigation volume for the second part of the tomato crop cycle (thesis 100-50). Extending the irrigation interval and limiting irrigation volume for the second part of the tomato crop cycle, appeared to be the best management strategy to optimize the yield and nutritional quality of processing tomato. 相似文献

The objective of this work was to quantify changes in the germination capacity of Juglans nigra seeds following storage and then artificial stratification. Seeds were stored at 5 and –20 °C under 5% and 14% internal moisture content (IMC) for 0, 8, 10, and 12 months under conditions with oxygen or under vacuum. Germination percentage (GP) was affected by storage temperature, IMC, and storage period. Seeds stored with 5% IMC succeeded in germinating regardless of the temperature. Seeds stored with 14% IMC did not germinate (–20 °C) or germinated during storage (5 °C). The GP was reduced with the time of storage in seeds at –20 °C and 5% IMC and this effect was avoided in storage under vacuum. In conclusion, the germination capacity of black walnut seeds did not change for up to 12 months at low above-zero temperature and low IMC (5 °C, 5% IMC), being the most cost-effective alternative for long-term storage of black walnut seeds in commercial nurseries. Individuals were selected for resistance against the imperfect fungus Seiridium cardinale Wag in six common cypress populations (Cupressus sempervirens L.) in Greece. A total of 109 resistant clones and 140 trees from their base populations were surveyed at several allozyme loci. The comparison yielded information on changes in genetic variation due to artificial selection. The genetic structure of the breeding populations was similar to their base populations. Nevertheless, the number of rare alleles among the resistant clones had decreased, showing a similar trend at most loci and for most comparisons. Possible implications for breeding strategies are discussed. 相似文献

The Sinorhizobium -legume interaction is sensitive to a number of environmental factors, soil acidity being one of the most important. In the typic Hapludoll soil of the central-southern region of Córdoba (Argentine) it was found that the nodulation of alfalfa ( Medicago sativaL.) roots was impaired with a reduction in shoot dry matter under conditions of soil acidity (pH 5.5) Our results showed that the addition of lime as dolomitic limestone at a concentration of 1 t ha -1to acid soil caused a significant increase in the nodulation of alfalfa roots inoculated with the strain Sinorhizobium meliloti SEMIA 116 (recommended inoculum for alfalfa) in the greenhouse experiments. The success of the lime treatment may be related not only to an increase in the pH values but also to an increase in the Ca concentration, improving the growth of S. meliloti and its nodulation ability under acidic conditions. In this study, we also demonstrated an increment in the bacterial growth rate as well as in the production of exopolysaccharides and lipopolysaccharides under low pH (5.5) and a high Ca concentration (5 mM) in the culture medium. 相似文献

The effects of dietary oregano essential oil and alpha-tocopheryl acetate supplementation on the oxidative stability of long-term frozen stored turkey meat were investigated. Thirty 12-week-old turkeys, randomly divided into five groups, were given a basal diet or a basal diet supplemented with 200 mg of alpha-tocopheryl acetate kg(-1), or 100 or 200 mg of oregano oil kg(-1), or 100 mg of oregano oil plus 100 mg of alpha-tocopheryl acetate kg(-1) for 4 weeks prior to slaughter. Lipid oxidation in breast and thigh meat was assessed after 1, 3, 6, and 9 months of frozen storage at -20 degrees C prior to or following 7 days of refrigerated storage at 4 degrees C. Results showed that oregano oil increased the oxidative stability of breast and thigh meat during the frozen storage. Dietary oregano oil at the inclusion level of 200 mg kg(-1) feed was significantly (p < 0.05) more effective in delaying lipid oxidation compared to the level of 100 mg kg(-1), but equivalent to dietary alpha-tocopheryl acetate supplementation at 200 mg kg(-1), which in turn was inferior to dietary supplementation of 100 mg kg(-1) oregano essential oil plus 100 mg kg(-1) alpha-tocopheryl acetate that was significantly (p < 0.05) superior to all other treatments. Thigh meat was more susceptible to oxidation than breast meat, although the former contained alpha-tocopherol at markedly higher levels. Mean alpha-tocopherol levels in breast and thigh meat from all treatments decreased during the frozen storage, the decrease being sharper between 1 and 3 months of frozen storage for breast and between 3 and 6 months for thigh meat. Oregano oil supplementation increased (p < 0.05) the retention of alpha-tocopherol in meat, the increase being positively correlated with the supplementation level. However, the retention of alpha-tocopherol in meat could only partly elucidate the antioxidant activity exhibited by dietary oregano oil supplementation. 相似文献

Commercial red wines ( Vitis vinifera L. cv. Shiraz) produced during the 2009 vintage underwent winemaker assessment for allocation grade soon after production. The wines were then subjected to phenolic analysis to measure wine color (total anthocyanin, SO(2) nonbleachable pigment, and wine color density) and tannins (concentration, composition, and average degree of polymerization). A positive relationship was found between wine phenolic concentration and projected bottle price. Tannin compositional analysis suggested that there was specifically a relationship between wine grade and skin-derived tannins. These results suggest that maximization of skin tannin concentration and/or proportion is related to an increase in projected wine bottle price. 相似文献
