The COVID-19 pandemic presents a continued public health challenge. Veterinary diagnostic laboratories in the United States use RT-rtPCR for animal testing, and many laboratories are certified for testing human samples; hence, ensuring that laboratories have sensitive and specific SARS-CoV2 testing methods is a critical component of the pandemic response. In 2020, the FDA Veterinary Laboratory Investigation and Response Network (Vet-LIRN) led an interlaboratory comparison (ILC1) to help laboratories evaluate their existing RT-rtPCR methods for detecting SARS-CoV2. All participating laboratories were able to detect the viral RNA spiked in buffer and PrimeStore molecular transport medium (MTM). With ILC2, Vet-LIRN extended ILC1 by evaluating analytical sensitivity and specificity of the methods used by participating laboratories to detect 3 SARS-CoV2 variants (B.1; B.1.1.7 [Alpha]; B.1.351 [Beta]) at various copy levels. We analyzed 57 sets of results from 45 laboratories qualitatively and quantitatively according to the principles of ISO 16140-2:2016. More than 95% of analysts detected the SARS-CoV2 RNA in MTM at ≥500 copies for all 3 variants. In addition, for nucleocapsid markers N1 and N2, 81% and 92% of the analysts detected ≤20 copies in the assays, respectively. The analytical specificity of the evaluated methods was >99%. Participating laboratories were able to assess their current method performance, identify possible limitations, and recognize method strengths as part of a continuous learning environment to support the critical need for the reliable diagnosis of COVID-19 in potentially infected animals and humans. 相似文献

Successful selection of interspecific hybrid progenies with superior ability to regenerate shoots from apical meristems was
performed in sunflower which now allows for the development of lines for improved biotechnological applications. Early generations
of interspecific hybrids originating from crosses between the two H. annuus CMS lines ‘HA89’ and ‘Baso’, and 9 wild species were screened for their ability to regenerate in vitro. Evaluation of 36 progenies allowed to identify seven progenies from crosses involving H. mollis, H. giganteus, H. strumosus, and H. decapetalus which showed a significantly higher regeneration potential than the commercial hybrid ‘Albena’ regarding the number of shoots
per explant. Among these progenies, 47.2 to 62.4% of explants produced shoots with an average of 2.3 to 3.5 shoots per cultured
explant. Regeneration in vitro was significantly determined by the genotype. More than half of the investigated interspecific hybrids performed better than
the inbred ‘HA89’ demonstrating that the high regeneration potential available in the wild species can be efficiently transferred
to cultivated sunflower. The seven progenies with high regeneration potential in vitro were characterised by agronomic performance in the field. Two of the interspecific hybrids derived from H. strumosus and H. decapetalus not only showed a superior regeneration potential but also proved to be competitive to commercial hybrids with regard to
important agronomic traits, e.g. fat content and TGW.

Late Cretaceous (Cenomanian) fossils discovered in the Kem Kem region of Morocco include large predatory dinosaurs that inhabited Africa as it drifted into geographic isolation. One, represented by a skull approximately 1.6 meters in length, is an advanced allosauroid referable to the African genus Carcharodontosaurus. Another, represented by a partial skeleton with slender proportions, is a new basal coelurosaur closely resembling the Egyptian genus Bahariasaurus. Comparisons with Cretaceous theropods from other continents reveal a previously unrecognized global radiation of carcharodontosaurid predators. Substantial geographic differentiation of dinosaurian faunas in response to continental drift appears to have arisen abruptly at the beginning of the Late Cretaceous. 相似文献

A silicon micromechanical magnetometer was constructed and successfully used in 60-tesla pulsed magnetic fields of less than 100-millisecond duration. The device is small, inexpensive to fabricate, and easy to use. It features a fast mechanical response (up to 50,000 hertz) and extremely high sensitivity yet is relatively robust against electrical and mechanical noise. The development of woodlots as an additional source of livelihood and as a land management option for small-scale farmers is a challenging issue in the cereal-based subsistence farming systems of north-western Ethiopia. There is a need to understand why and which factors determine the decisions of small-scale farmers to grow short-rotation woodlots on their land. Data used in this study were collected from a survey of 200 randomly selected households in the region. A Tobit regression model was used to determine predictor variables for farmers’ decisions to allocate land to planting Acacia decurrens (J.C. Wendl.) Willd. and at what density trees are planted on the respective plots. The most important motivations for planting A. decurrens were income, soil fertility management, and soil and water conservation. Having a male head of household, long distance to markets and plots being on marginal land, among other factors, increased the allocation of land to A. decurrens woodlots. Having a male head of household, access to credit and plots being on marginal land, among other factors, increased tree planting density. Age had a negative effect on both allocation of land to woodlots and tree density, whereas farm size had an inverted U-shaped relationship with both decisions. These results suggest that wider expansion of A. decurrens-based plantation systems could be achieved through improving extension, credit access and road infrastructure to connect small-scale farmers to markets and finance. 相似文献

Agricultural extension is an approach to rural development and agricultural transformation in which training, demonstration and technology transfer are key to reducing rural poverty, ensuring food security, and sustainably managing natural resources. During recent decades, different extension approaches have been tested and validated by the Ethiopian government and non-governmental organizations to stimulate participation in the agricultural extension system (AES). The most recent was a German-funded project entitled “Integrated Soil Fertility Management Project” (ISFM+), which employed a novel approach to piloting and upscaling proven technology and best practice. The purpose of this study was to analyze and document the modalities of ISFM+ and illustrate its effects on technology uptake and dissemination. The study used a mixed methods approach to collect data. ATLAS.ti and SPSS were used for data management and analysis. Farmer Research and Extension Groups and Farmer Field Schools were found to be central to the participation process. Also, the ISFM+ was found to aid technology transfer and helped to increase grain and residue yields as well as farmer livelihoods. Based on these empirical findings, it is argued that the ISFM+ approach and technology should be integrated and institutionalized in the mainstream AES in order to promote their extensive application. 相似文献

Horizontal gene transfer plays a major role in microbial evolution. However, newly acquired sequences can decrease fitness unless integrated into preexisting regulatory networks. We found that the histone-like nucleoid structuring protein (H-NS) selectively silences horizontally acquired genes by targeting sequences with GC content lower than the resident genome. Mutations in hns are lethal in Salmonella unless accompanied by compensatory mutations in other regulatory loci. Thus, H-NS provides a previously unrecognized mechanism of bacterial defense against foreign DNA, enabling the acquisition of DNA from exogenous sources while avoiding detrimental consequences from unregulated expression of newly acquired genes. Characteristic GC/AT ratios of bacterial genomes may facilitate discrimination between a cell's own DNA and foreign DNA. 相似文献

Effect of Initial GnRH and Duration of Progesterone Insert Treatment on the Fertility of Lactating Dairy Cows 下载免费PDF全文
The study compared response to prostaglandin F2α (PG), synchrony of ovulation and pregnancy per AI (P/AI) in a 5‐ vs a 7‐day Ovsynch + PRID protocol and investigated whether the initial GnRH affects P/AI in lactating dairy cows. Two hundred and seventy‐six cows (500 inseminations) were assigned to one of four timed‐AI (TAI) protocols: (i) PRID‐7G; 100 μg GnRH im, and a progesterone‐releasing intravaginal device (PRID) for 7 days. At PRID removal, PG (500 μg of cloprostenol) was given im. Cows received the second GnRH treatment at 60 h after PRID removal and TAI 12 h later. (ii) PRID‐5G; as PRID‐7G except the duration of PRID, treatment was 5 days and PG was given twice (12 h apart). (iii) PRID‐7NoG; as PRID‐7G except the initial GnRH, treatment was omitted. (iv) PRID‐5NoG; as PRID‐7NoG except the duration of PRID, treatment was 5 days. Response to treatments and pregnancy status at 32 and 60 days after TAI was determined by ultrasonography. The percentage of cows ovulating before TAI was greatest in PRID‐7G (17.1%), and the percentage of cows that did not have luteal regression was greatest in PRID‐5G (9.5%). The overall P/AI at 32 and 60 days did not differ among TAI protocols. However, during resynchronization, cows subjected to the 5‐day protocols had greater (p < 0.05) P/AI (45.3% vs 33.6%) than cows subjected to the 7‐day protocols. Pregnancy loss between 32 and 60 days tended (p = 0.10) to be greater in cows that did not receive initial GnRH (14.8%) compared to those that received GnRH (8.2%). In conclusion, the PRID‐5G protocol resulted in fewer cows responding to PG, but P/AI did not differ among TAI protocols. A 5‐day protocol resulted in more P/AI in resynchronized cows, and cows that did not receive initial GnRH tended to experience more pregnancy losses. 相似文献
