Veterinary Research Communications, 21 (6), 381-407Steptococcus suis is a Gram-positive, facultatively anaerobic coccus that has been implicated as the cause of a wide range of clinical disease syndromes in swine and other domestic animals. In swine, the disease has spread worldwide but is more prevalent in countries with intensive swine management practices. The disease syndromes caused by S. suis in swine include arthritis, meningitis, pneumonia, septicaemia, endocarditis, polyserositis, abortions and abscesses. S. suis has also been implicated in disease in humans, especially among abattoir workers and swine and pork handlers. In humans, S. suis type 2 can cause meningitis, which may result in permanent hearing loss, septicaemia, endocarditis and death. The pathogenic mechanism of S. suis is not well defined. Several virulence factors have been identified, but their roles in pathogenesis and disease have not been well elucidated. Much work is in progress on characterization of virulence factors and mechanisms, with emphasis on the control of the disease. Because of the non-availability of suitable immunoprophylaxis, control of S. suis infection has depended mainly on the use of antimicrobials.  相似文献

Virulence factors responsible for acute diarrhea in greyhounds have not been well established. The objective of this study was to determine if a correlation exists between disease and the presence of the Escherichia coli toxin genes in non-diarrheic and diarrheic greyhound feces. DNA extracted from broth cultures was evaluated for the presence of Shiga toxin and enterotoxin genes and broth samples were evaluated for Shiga toxin and heat-labile enterotoxin. Shiga toxin (stx1 and stx2) and enterotoxin (et and estA) genes were identified in both non-diarrheic and diarrheic samples after in vitro cultured of swabs at 37 degrees C for 16-24h. The stx1 gene was present in 3% of non-diarrheic and 15% diarrheic samples and the stx2 gene was identified in 36 and 23%, non-diarrheic and diarrheic samples, respectively. Shiga toxin was present in 48% diarrheic and 25% of the non-diarrheic in vitro cultured samples. The elt gene was detected in vitro cultured swabs in 12% of the non-diarrheic and 7% of the diarrheic samples. Labile toxin was present in the feces of small numbers of both groups of dogs. A significant correlation existed between the presence of both stx1 genes and Shiga toxin in feces, and lack of disease in non-diarrheic (P=0.01) and presence of disease in diarrheic (P=0.024) greyhounds. Correlation between production of Shiga toxin and detection of stx1 or stx2 was significant in both the diarrheic and non-diarrheic feces (P=0.03); however, only the presence of stx1 correlated with diarrhea in both groups of samples (P<0.008). The incidence of toxigenic E. coli in both non-diarrheic and diarrheic greyhounds indicates a zoonotic potential from dogs to humans and requires further study.  相似文献

The design of a human immunodeficiency virus-1 (HIV-1) immunogen that can induce broadly reactive neutralizing antibodies is a major goal of HIV-1 vaccine development. Although rare human monoclonal antibodies (mAbs) exist that broadly neutralize HIV-1, HIV-1 envelope immunogens do not induce these antibody specificities. Here we demonstrate that the two most broadly reactive HIV-1 envelope gp41 human mAbs, 2F5 and 4E10, are polyspecific autoantibodies reactive with the phospholipid cardiolipin. Thus, current HIV-1 vaccines may not induce these types of antibodies because of autoantigen mimicry of the conserved membrane-proximal epitopes of the virus. These results may have important implications for generating effective neutralizing antibody responses by using HIV-1 vaccines.  相似文献

The objective of this study was to assess the genetic diversity and to infer the mode of reproduction of Botrytis elliptica and B. tulipae in the Netherlands. First, three molecular typing methods were compared for their ability to differentiate isolates of B. tulipae, B. elliptica, and B. cinerea. The methods compared were multilocus sequencing, restriction analysis of the ribosomal intergenic spacer (IGS) region, and amplified fragment length polymorphism (AFLP) analysis. AFLP fingerprinting provided the most efficient method to differentiate isolates within each Botrytis species and therefore this method was used for population analyses of B. elliptica and B. tulipae. Isolates of both species were sampled during successive growing seasons in experimental field plots in Lisse and other locations in the Netherlands. Among 174 B. elliptica isolates, 105 genotypes could be discriminated and 87 genotypes were found only once, reflecting high genotypic variation. Clonal genotypes were found only within growing seasons and in one location. Linkage disequilibrium analyses indicated that between 9.4% and 19.3% of the loci in clone-corrected samples were linked. The multilocus association index provided no evidence for random mating. We conclude that sexual recombination occurs in the B. elliptica population. Among the 170 B. tulipae isolates, 25 genotypes could be discriminated and four genotypes were found only once, reflecting a low genotypic variation. Clonal genotypes were frequently found in different growing seasons and different locations. Linkage disequilibrium analyses indicated that between 25.2% and 48.6% of the loci in clone-corrected samples were linked. We conclude that the B. tulipae population is mainly clonal with some recombination.  相似文献

Nineteen Streptococcus suis type 2 isolates that had been analyzed previously for hemolysin production, ribotype, and virulence in pigs were examined for presence of the gene coding for suilysin by PCR amplification, and southern blot and hybridization techniques. Based on southern blot and hybridization analysis, all isolates tested contained at least a portion of the suilysin gene. PCR amplification of the entire gene resulted in gene fragments from five of the seven highly virulent isolates and none of the moderately virulent or avirulent isolates. Additional PCR analysis showed that mutation or deletions at the 5′ end of the suilysin gene in the less virulent isolates prevented amplification of the sly gene fragment from those isolates. The MRP+ (muramidase-released protein) EF+ (extracellular protein) phenotype was also expressed by the same five highly virulent/sly+ isolates.  相似文献

Stereological techniques were used to assess seasonal influences on morphometric characteristics of hair follicles, sweat and sebaceous glands in abattoir pelts of ponies (PN), thoroughbred (TB) and non-thoroughbred (NTB) horses. Volume density of sweat glands increased significantly from winter (0.061) to summer (0.098) in TB, and showed no change in NTB and a positive tendency in PN. There might be a body surface area : volume effect for sweat gland parameters as PN had smaller values than either TB or NTB, probably attributable to control of heat loss in winter. In summer, the skin remained thick and the volume density of sebaceous glands was increased in NTB, in contrast to TB where both were decreased. It is possible that in summer, sebum has a particular importance in NTB to enhance wicking of sweat through the pelt. TB showed significantly higher volume measurements of sebaceous glands than NTB and PN for winter: sebum has probably a special importance for water-proofing in TB in winter. PN showed no significant seasonal changes in sebaceous glands, but had a thinner summer skin. Winter values for hair follicle volume density between equine groups were similar (TB, NTB 0.066; PN 0.059), as was skin thickness (1.14-1.19 mm). The volume density lowered significantly in summer in TB and NTB. The volume of hair follicles under a unit area of skin surface decreased significantly in TB and nonsignificantly in NTB and PN. The seasonal adaptations of the skin shown here were most pronounced in TB and differed between breeds.  相似文献
