Over a series of seven separate experiments 76 different lupin (Lupinus angustifolius) meals were assessed for their digestible dry matter, protein, amino acid and energy characteristics when fed to rainbow trout. Two reference diets (a common basal diet and a reference lupin meal) were also included in each experiment. Minimal variance in the digestibility parameters of both reference diets was observed among the experiments ensuring that there was a high degree of robustness in the across-experiment evaluations. Using simple and multiple-regression techniques, principal diet and ingredient composition factors affecting diet and ingredient digestibilities and ingredient digestible values were explored within the dataset. Using simple linear regression, it was shown that nitrogen digestibility of the lupin meals was negatively influenced by ingredient lignin content, but positively affected ingredient protein (N × 6.25) content. The energy digestibility of the lupin meals was positively affected by a range of compositional features including protein, sum of amino acids and negatively affected by carbohydrate content. The digestible nutrient and energy content of the meals reflected the combined effects of both ingredient digestibilities and ingredient composition. The digestible nitrogen content of the lupin meals was positively affected by protein (N × 6.25), sum of amino acids and energy content, but was negatively affected by lignin and carbohydrate content. The digestible sum of amino acids was also positively affected by protein, sum of amino acids, but only negatively affected by carbohydrate content, not lignin content. The digestible energy content of the lupin meals was also positively affected by protein, sum of amino acids and their own energy density, but only negatively affected by carbohydrate content. Multiple linear regression modelling supported that together ingredient protein and lignin content were the strongest predictors of digestible protein value, explaining close to 60% of the variability in this parameter. Discrete reassessment of these relationships using non-linear analysis methods provided a stronger interpretation of the ingredient composition effects. In recognition of this, a functional model including terms for ingredient protein and lignin content on the digestible protein value was defined. This study demonstrates that within one raw material type that not only does significant variability in the digestible value of the raw materials exist, but that it is possible to identify compositional features of that raw material that are intrinsically influencing their own digestible value.  相似文献

The influence on wheat flour gluten-starch separation of a xylanase from Aspergillus aculeatus (XAA) with hydrolysis selectivity toward water extractable arabinoxylan (WE-AX) and that is not inhibited by wheat flour xylanase inhibitors was compared to that of a xylanase from Bacillus subtilis (XBS) with hydrolysis selectivity toward water unextractable arabinoxylan (WU-AX) and that is inhibited by such inhibitors. XAA improved gluten agglomeration through degradation of WE-AX and concomitant reduction in viscosity, which in the laboratory scale batter procedure with a set of vibrating sieves (400, 250, and 125 microm), increased protein recoveries on the 400 microm sieve. In contrast, XBS had a negative effect as it decreased gluten protein recovery on this sieve, probably as a result of the viscosity increase that accompanied WU-AX solubilization. Hence, it was active even if most likely a considerable part of its activity was prevented by xylanase inhibitors. A combination of XAA and XBS at a low dosage yielded a distribution of gluten proteins on the different sieves comparable to that of the control. At a high combined dosage, the gluten agglomeration was better than that with XAA alone, indicating that both WE-AX and WU-AX have a negative impact on gluten agglomeration. Finally, experiments with endoxylanase addition at different moments during the separation process suggest that the status of the arabinoxylan population during dough mixing is far less critical for its impact on gluten agglomeration than that during the batter phase.  相似文献

The marine environment is an excellent resource for natural products with therapeutic potential. Its microbial inhabitants, often associated with other marine organisms, are specialized in the synthesis of bioactive secondary metabolites. Similar to their terrestrial counterparts, marine Actinobacteria are a prevalent source of these natural products. Here, we discuss 77 newly discovered alkaloids produced by such marine Actinobacteria between 2017 and mid-2021, as well as the strategies employed in their elucidation. While 12 different classes of alkaloids were unraveled, indoles, diketopiperazines, glutarimides, indolizidines, and pyrroles were most dominant. Discoveries were mainly based on experimental approaches where microbial extracts were analyzed in relation to novel compounds. Although such experimental procedures have proven useful in the past, the methodologies need adaptations to limit the chance of compound rediscovery. On the other hand, genome mining provides a different angle for natural product discovery. While the technology is still relatively young compared to experimental screening, significant improvement has been made in recent years. Together with synthetic biology tools, both genome mining and extract screening provide excellent opportunities for continued drug discovery from marine Actinobacteria.  相似文献

Imazapyr absorption, translocation, root release and metabolism were examined in leafy spurge (Euphorbia esula L.). Leafy spurge plants were propagated from root cuttings and [14C]imazapyr was applied to growth-chambergrown plants in a water + 28% urea ammonium nitrate + nonionic surfactant solution (98.75 + 1 + 0.25 by volume). Plants were harvested two and eight days after herbicide treatment (DAT) and divided into: treated leaf, stem and leaves above treated leaf, stem and leaves below the treated leaf, crown, root, dormant and elongated adventitious shoot buds. Imazapyr absorption increased from 62.5% 2 DAT to 80.0% 8 DAT. Herbicide translocation out of the treated leaf and accumulation in roots and adventitious shoot buds was apparent 2 DAT. By the end of the eight-day translocation period only 14% of applied 14C remained in the treated leaf, while 17% had translocated into the root system. Elongated and dormant adventitious shoot buds accumulated 3.2- and 1.8-fold more 14C, respectively, 8 DAT than did root tissue based on Bq g?1 dry weight. Root release of 14C was evident 2 DAT, and by 8 DAT 19.4% of the 14C reaching the root system was released into the rooting medium. There was no metabolism of imazapyr in crown, root or adventitious shoot buds 2 DAT; however, imazapyr metabolism was evident in the treated leaf 2 and 8 DAT. Imazapyr phytotoxicity to leafy spurge appears to result from high imazapyr absorption, translocation to underground meristematic areas (roots and adventitious shoot buds), and a slow rate of metabolism.  相似文献

beta-Hydroxy-beta-methyl butyrate(HMB) has been shown to counteract many of the negative effects of intensive animal production methods and results in increased growth and protection against diseases. In the present study, the effect of HMB on the immunocompetence cell activity in rainbow trout (Oncorhynchus mykiss) and carp (Cyprinus carpio) was examined. Pronephric phagocytes and lymphocytes were isolated from the fish and grown in culture medium (RPMI-1640) containing either 0, 0.1, 1, 5, 10, 25, 50 or 100 microg HMB/ml of medium. The effects of HMB on the respiratory burst activity (RBA) stimulated by phorbol myristate acetate (PMA), the potential killing activity (PKA) and lymphocyte proliferation stimulated by either concanavalin A (Con-A) or lipopolysaccharide (LPS) were examined. The addition of HMB to the culture medium increased the RBA by up to 84% (p<0.01) over that of cells grown without HMB. Similarly, the PKA of the phagocytes was also increased with HMB addition to the medium by up to 140% (p<0.01) over that of cells grown without HMB. Lymphocyte proliferation stimulated by both ConA and LPS was also increased approximately two-fold (p<0.01) when HMB was added to the culture medium at concentrations between 10 and 100 microg HMB/ml in both rainbow trout and carp. The greatest effects of HMB on RBA and PKA activities were observed at a concentration >50 microg HMB/ml while lymphocyte proliferation was maximally stimulated at 25 microg HMB/ml. In conclusion, the current study shows that HMB could potentially improve immunocompetence cell activity in fish through increased cell proliferation and functionality.  相似文献

To derive optimal benefits from intercropping timber, farmers should make important initial decisions on tree species and planting density with a good understanding of their tradeoffs. Complex and data-intensive models used by researchers should be supplemented with simpler models based on easily measured parameters and easily understood competition functions. In experiments in the Philippine uplands, growth parameters of three popular farm-forestry species (Eucalyptus deglupta, E. torelliana, and Paraserianthes falcataria) were measured, along with intercropped and non-intercropped yields of maize and vegetables. The commonly used forestry parameter of stand basal area had a significant negative correlation with intercrop yields (as a percentage of non-intercropped yields). The slope of the regression line differed between species; in this study, percent yield loss per unit stem basal area growth was in the order E. deglupta > E. torelliana > P. falcataria. The relationship between stand basal area and intercrop-yield decline was tested on an independent data set from China. Intercrop yields had significant negative correlations with stand basal area of Paulownia elongata. We propose that adaptive tree-screening trials evaluate competitiveness in addition to evaluating growth and mortality. Stand basal area may be better suited to this task than more mechanistic indices such as leaf-area index as it is easy to measure, calculate, and understand, and it may serve as a better index of total (aboveground + belowground) competition. Basal area is also directly related to tree volume, and allows farmers to more easily evaluate the economic tradeoffs between tree growth and intercrop-yield declines. Studies have shown that in both in vitro and in vivo tests, β‐hydroxy‐β‐methylbutyrate (HMB) increases the nonspecific cellular and humoral immune response and protection against diseases in animals. The present study examines the influence of HMB on nonspecific humoral defense mechanisms and protection against furunculosis in pikeperch (Sander lucioperca). β‐hydroxy‐β‐methylbutyrate was fed in a pelleted ration of 50 mg kg?1 feed day?1 for 4 weeks. Blood was drawn from 12 HMB‐fed and control‐fed pikeperch. The lysozyme and ceruloplasmin activities in the plasma, total immunoglobulin (Ig) levels, and total serum protein were analysed prior to and then after 2 and 4 weeks of HMB ingestion. After 4 weeks of HMB ingestion, a challenge test was performed by injecting the fish with live pathogenic Aeromonas salmonicida bacteria. β‐hydroxy‐β‐methylbutyrate at a dose of 50 mg kg?1 feed resulted in a statistically significant (P<0.05) increase in the lysozyme activity of the plasma, total Ig, and serum protein levels. Additionally, reduced mortality (40%) after the in vivo challenge with pathogenic A. salmonicida suggested that HMB‐activated nonspecific protection against furunculosis in pikeperch.  相似文献

In recent years the consumption of tea and herbal infusions has increased. These hot drinks are consumed as daily drinks as well as for medicinal purposes. All tea varieties (white, yellow, green, oolong, black and puerh) originate from the leaves of the tea plant, Camellia sinensis. All extracts made of plant or herbal materials which do not contain Camellia sinensis are referred as herbal infusions or tisanes. During processing and manufacturing fungal contamination of the plant materials is possible, enabling contamination of these products with mycotoxins. In this study a multimycotoxin UPLC-MS/MS method was developed and validated for the analysis of the raw tea and herbal infusion materials as well as for their drinkable products. The samples were analyzed by ultra-high-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS), with a mobile phase consisting of variable mixtures of water and methanol with 0.3% formic acid. The limits of detection for the different mycotoxins varied between 2.1 μg/kg and 121 μg/kg for raw materials and between 0.4 μg/L and 46 μg/L for drinkable products. Afterward 91 different tea and herbal infusion samples were analyzed. Only in one sample, Ceylon melange, 76 μg/kg fumonisin B(1) was detected. No mycotoxins were detected in the drinkable products.  相似文献
