As a result of screening procedures employed for animals entering the AI service, two bulls were identified as being persistently infected with bovine virus diarrhoea virus by isolation of the virus from blood. Semen was collected on two occasions from these bulls; its quality as measured by density and motility was poor. Gross abnormalities of the sperm head, termed 'collapsed' heads, were seen in 28 to 45 per cent of sperm from one bull and in 1 per cent of sperm from the other. The collapsed heads were small and the whole head or its anterior part had the appearance of a dried pea. Electron microscopy showed the defect to consist of convoluted nuclear material with membrane-bound vacuoles and invaginations containing membranous debris and lamellar structures. In the 'high incidence' bull there was a corresponding increase in enlarged sperm heads. The 'low incidence' bull had sperm with heads of similar mean size to sperm from control bulls but with an increased variance. The semen was diluted in a lactose diluent, frozen and stored in liquid nitrogen. The distribution of viral antigen was determined and virus was isolated from several fractions of the semen, both before and after processing and cryopreservation. In one animal raw semen failed to yield virus but virus was recovered after processing, suggesting that raw semen may not be suitable for the efficient detection of the virus.  相似文献

The stable carbon isotope technique has been widely used to infer the dietary ecology of a range of animal species; however calibration of the technique with animals fed known diets is essential for accurate back-calculation of dietary preferences. The aim of this study was to identify suitable samples and back-calculation methods to predict short-term (2 to 3 week) dietary selection by sheep among plants with C3 and C4 photosynthetic pathways. Variation in integration time of dietary carbon into plasma and faeces; diet-tissue discrimination of carbon isotopes (fractionation) and the importance of accounting for the digestible or indigestible components of the diet was investigated. The results indicate that faecal and rumen samples provided the most accurate prediction of short term dietary changes in sheep selecting between C3 and C4 plants. The most accurate back-calculation method for these samples used δ13C of the C3 and C4 plants and accounted for both diet-tissue discrimination and differences in the indigestibility between the C3 and C4 forage. For faecal samples, the organic matter content of the diet originating from C4 plants could be predicted with a mean error as low as 2.7%. Wool and plasma samples were not conducive to predicting proportion of C4 forage in the diet within 18 days after a change in diet; however plasma could be used to discriminate between animals fed 100% C3 and C4 diets after 3 days. The δ13C technique provides a valuable tool for researchers when designing pastures for dual environmental and production purposes. An understanding of what sheep select allows for development of appropriate grazing management strategies to optimise productivity and/or persistence of target species.  相似文献

In face of climate change and other environmental challenges, one strategy for incremental improvement within existing farming systems is the inclusion of perennial forage shrubs. In Australian agricultural systems, this has the potential to deliver multiple benefits: increased whole-farm profitability and improved natural resource management. The profitability of shrubs was investigated using Model of an Integrated Dryland Agricultural System (MIDAS), a bio-economic model of a mixed crop/livestock farming system. The modelling indicated that including forage shrubs had the potential to increase farm profitability by an average of 24% for an optimal 10% of farm area used for shrubs under standard assumptions. The impact of shrubs on whole-farm profit accrues primarily through the provision of a predictable supply of ‘out-of-season’ feed, thereby reducing supplementary feed costs, and through deferment of use of other feed sources on the farm, allowing a higher stocking rate and improved animal production. The benefits for natural resource management and the environment include improved water use through summer-active, deep-rooted plants, and carbon storage. Forage shrubs also allow for the productive use of marginal soils. Finally, we discuss other, less obvious, benefits of shrubs such as potential benefits on livestock health. The principles revealed by the MIDAS modelling have wide application beyond the region, although these need to be adapted on farm and widely disseminated before potential contribution to Australian agriculture can be realized.  相似文献

On assessment for use in an AI stud, a 12‐month‐old bull was found to produce low volume ejaculates with 41% of the sperm having morphological abnormalities. No left epididymal tail was palpable and the head of the epididymis on the left was twice the size compared with the right. Ultrasound examination showed the left testis to contain a large central area of decreased echogenicity, which could be followed proximally to a 15‐mm echolucent lesion at the site of the epididymal head. Postmortem examination revealed a 15‐mm diameter cyst in the region of the left epididymal head, and absence of the body and tail of the epididymis. The mediastinum testis of the left testis was dilated, corresponding to the area of decreased echogenicity observed on ultrasonography. No left seminal vesicle was present and the ampulla was significantly smaller than the same structure on the right. Histological examination revealed incomplete or absent spermatogenesis involving the majority of seminiferous tubules in the left testis, and a small proportion of those of the right testis. The cystic structure at the site of the left epididymal head was lined by irregular, sometimes attenuated, epithelium and contained sparse spermatozoa. This case demonstrates the adverse impact, which segmental aplasia of the mesonephric duct had on the testicular and epididymal function of a bull, and highlights the importance of careful clinical assessment in its diagnosis.  相似文献

Pasture legumes are important components of both mixed farming rotations and permanent pastures in temperate climates. Breeding of two widely sown pasture legumes, subterranean clover (Trifolium subterraneum L.) and French serradella (Ornithopus sativus Brot.), is constrained by the long generation cycle, typically enabling only one generation per year. We hypothesized manipulation of culture medium and conditions would enable the development of a laboratory‐based protocol for in vitro reproduction in subterranean clover and French serradella. In vitro flowering and viable seed set was induced from both species. For subterranean clover, the most effective treatment was culturing on modified MS medium with 1 μm kinetin and 0·1 m sucrose under a 100 μmol m?2 s?1 light intensity and continuous photoperiod. For French serradella, culture on a hormone‐free B5 medium with 5 mm NH4Cl and 0·1 m sucrose under a 100 μmol m?2 s?1 light intensity and 20 h photoperiod was optimum. It is expected this technique will have application in accelerating generation turnover within breeding programs, for the study of factors influencing flowering in pasture legumes, and for the propagation of valuable yet enfeebled plants such as embryo‐rescued hybrids.  相似文献

Eastern star clover (Trifolium dasyurum C. Presl.) is a new pasture legume developed for use in short phases between extended cropping sequences in Western Australian dryland agriculture. Its delayed germination provides an opportunity to obtain almost complete control of the highly-competitive crop weed rigid ryegrass (Lolium rigidum Gaudin) through non-selective herbicide application and/or grazing. Given the recent development of a commercial cultivar of eastern star clover (cv. AGWEST® Sothis), a complex simulation model is used to evaluate its potential profitability relative to continuous-cropping and rotations employing a popular pasture, French serradella (Ornithopus sativus Brot. cv. Cadiz). The profitability of those sequences containing eastern star clover is robust to high initial ryegrass populations and increasing severity of herbicide resistance. Moreover, the weed control benefits accruing to this pasture's delayed germination are of sufficient magnitude to offset the low establishment cost and higher biomass production of French serradella. This highlights the value of eastern star clover to producers in the Western Australian wheatbelt and offers an additional trait that plant breeders and selectors can exploit when seeking to expand weed control options in land-use sequences.  相似文献

This study examined how sheep develop a learned aversion to the annual legume biserrula (Biserrula pelecinus L.). Merino sheep were grazed on paddocks sown with either (i) biserrula, (ii) biserrula + ryegrass (Lolium rigidum Gaud.), (iii) crimson clover (Trifolium incarnatum L.), (iv) crimson clover + ryegrass or (v) ryegrass, for 4 weeks. Relative preference (Chesson–Manly selection index) was determined by offering randomly ordered monoculture plots of the three forage species. The groups of sheep were then combined into a single group and grazed a paddock containing a monoculture sward of each plant type for 5 d. Relative preference of sheep was tested again after the completion of the combined grazing. The experiment was repeated at three stages of plant phenology: vegetative, reproductive and senesced. Relative preference for biserrula was lower in sheep that had grazed the paddock sown with only biserrula compared with sheep that grazed a combination of biserrula + ryegrass, at both the vegetative (α; −0·11 vs. 0·32) and reproductive (α; 0·18 vs. 0·63) stages. At each stage of phenology, when groups were combined and grazed together, preference for biserrula became uniform. This supported the hypothesis that sheep develop an aversion to biserrula when they graze a pasture containing biserrula. However, we concluded that the familiarity of sheep with biserrula did not result in low relative preference; rather, the aversion was more likely a response to the high proportion of this plant in their diet. Social facilitation may have enabled sheep to overcome rapidly the food aversion.  相似文献

Over the past decade, lambing percentages have risen in conjunction with a rise in the percentage of multiple lambs born. Multiple-born lambs are smaller than their singleton counterparts and are particularly susceptible to starvation-exposure. Any technique that can increase the birthweight or thermoregulatory capability, or both, of otherwise lightweight lambs has the potential to substantially increase survival of multiple-born lambs. In the United Kingdom under housed conditions, shearing during pregnancy has been shown to increase both the birthweight and thermoregulatory capability of newborn lambs. However, shearing during pregnancy under pastoral conditions has failed to consistently affect the newborn lamb's thermoregulatory capability. In contrast, under New Zealand's pastoral conditions shearing during pregnancy has been found to increase birthweight, but results have been inconsistent in both magnitude and birth-rank specificity. Increase in feed intake by the dam, types of shearing comb used and changes in gestation length do not explain the variation observed. When studies involving shearing during pregnancy are collated it becomes apparent that there are two criteria that must be met to achieve a birthweight response. Firstly, the dam must have the potential to respond and secondly, the dam must have the means to respond. Any increase in lamb survival through shearing during pregnancy would be predominantly through an increase in birthweight of otherwise lightweight lambs. However, in the few studies conducted to date, an increase in birthweight has not resulted in a statistically significant increase in survival. For an increase in birthweight to have a positive effect on lamb survival, lambs must be otherwise destined to be born within a birthweight range in which survival rate is below optimum (<4.0 kg), and the increase in birthweight observed must move a significant proportion of otherwise lightweight lambs into a higher range of survival rate.  相似文献
