Bermudagrass (Cynodon dactylon L.) is a warm season perennial that is well adapted in the southern Great Plains. It is one of the region's most important forage crops used for livestock production, and is commonly grown without legume interseeding. Recent research has investigated ways of improving the quality and quantity of this forage. The objectives of this study were to determine the effect of interseeded legumes and phosphorus (P) fertilizer on bermudagrass pasture forage yield and crude protein content. One experiment was initiated in 1993 in eastern Oklahoma in an established bermudagrass pasture. Red clover (Trifolium pratense L.), ladino clover (Trifolium repens L.), and two varieties of alfalfa (Medicago sativah), ’alfagraze’ and'common’, were interseeded by hand into an established stand of bermudagrass. The effect of P on forage yield and crude protein was evaluated using a 30‐kg P ha‐1 rate applied at establishment versus no applied P. Forage yield was collected three times throughout the growing season each year from 1994 through 1997. When both alfalfa varieties were interseeded into a bermudagrass pasture without applying additional P fertilizer, forage yields for the legume‐grass mixtures decreased below those obtained from the monoculture bermudagrass in the first year of the stand. The alfalfa variety ‘alfagraze’ interseeded into established bermudagrass decreased total forage yield over the entire 4‐yr study. Interseeded red clover and ladino clover increased crude protein of the forage compared with monoculture bermudagrass the first two years of the study, with red clover continuing to increase crude protein in the fourth year. However, when 30 kg P ha‐1 was applied to the bermudagrass prior to establishment of the legumes, no change in yield or protein was observed for both alfalfa varieties’ interseeding treatments versus the unfertilized mixtures. Although forage yield may not be increased, interseeding legumes into established bermudagrass could provide an efficient way to improve pasture crude protein without the use of inorganic fertilizers. However, if alfalfa ('common’ or ‘alfagraze') is interseeded, additional P may need to be applied at legume establishment to prevent possible yield decreases.  相似文献

The use of variable rate technology has become increasingly popular for applying plant nutrient elements. The most widely used method for determining variable fertilizer rates is presently based on soil testing and yield mapping. Three field studies (Bumeyville 1995, Burneyville 1996, and Ardmore 1996) were initiated in established Midland bermudagrass [Cynodon dacrylon (L) Pers.] pastures to determine the relationship between spectral radiance at specific wavelengths with forage nitrogen (N) removal and biomass, and to determine field variability of soil test parameters. Variable N (applied to 1.5 × 2.4 m subplots within 2.4 × 45.7 m main plots), fixed N and check treatments were evaluated at each location. Spectral radiance readings were taken in the red (671±6 nm), green (570±6 nm), and near infrared (NIR) (780±6 nm) wavelengths. The normalized difference vegetation index (NDVI) was calculated as NIR‐red/NIR+red. Variable N rates were applied based on NDVI. The highest fixed variable N rate was set at 224, 336, and 672 kg N ha‐1 for Burneyville, 1995, 1996, and Ardmore, 1996, respectively. At Bumeyville, soil samples were collected in all variable rate plots (1.5 × 2.4 m) and analyzed for various soil test characteristics. NDVI, red, green, and NIR spectral radiance readings were correlated with bermudagrass forage N removal and yield. Correlation of forage yield and N removal with red, NIR, and NDVI were best with maximum forage production, however, when forage production levels were low correlation decreased dramatically for the red wavelength compared with NIR and NDVI. Forage yield and forage N removal in variable rate treatments increased when compared to the check while being equal to the half‐fixed and fixed rates where higher N rates were applied. Also, variability about the mean in variable rate plots was significantly lower than half‐fixed and fixed rates which supports adjusting N rates based on indirect NDVI measurements. Variable N rate plots reduced fertilizer inputs by 60% and produced the same yield as fixed rate plots, while fixed and half‐fixed rates did not increase N content in the forage over that of the variable rate treatment. Soil sample data collected from small consecutive plots (<4 m2) was extremely variable indicating that intense sampling would be needed if variable fertilizer application were to be based on soil test results.  相似文献

Fetal protein restriction is potentially associated with organ dysfunctions after birth (e.g. impaired gut growth, glucose tolerance and pancreatic β-cell function). Just after birth, gut growth and maturation is stimulated by enteral food intake, and inhibited by total parenteral nutrition (TPN), in part mediated via differential release of insulino- and intestino-tropic hormones like the Glucagon-Like Peptides 1 and 2 (GLP-1, GLP-2). We hypothesized that short-term co-infusion of GLP-1 and GLP-2 would stimulate pancreatic and intestinal growth in newborn TPN-fed pigs subjected to prenatal protein restriction. Two sows were fed a protein-restricted diet (PR: 8% crude protein during last 50% of gestation) while a third sow was fed a control diet (C: 15% crude protein). PR pigs were killed either at birth (n = 7) or after 3 days TPN with (n = 6) or without (n = 4) intravenous infusion of a mixture of synthetic human GLP-17–37 and GLP-21–33 (each 50 μg/kg/d). At birth, PR piglets did not show reduced body weight, relative to controls (1.45 vs. 1.50 kg), but significantly reduced weight of the small intestine (18.0 ± 0.6 vs. 21.9 ± 0.5 g/kg, P < 0.001) and a marginally reduced pancreas weight (0.85 ± 0.02 vs. 0.93 ± 0.04 g/kg, P = 0.10). Co-infusion GLP-1 and GLP-2 into PR pigs resulted in increased basal glucose levels (5.3 vs. 4.0 mM), and glucose-stimulated insulin release, but did not have any significant effect on body weight, or weight of internal organs (heart, lungs, spleen, kidneys, adrenals, stomach, colon, liver, intestine, pancreas). We conclude that short-term (3 days) infusion of native GLP-1 and GLP-2 does not stimulate gut growth or glucose tolerance in TPN-fed piglets born from protein-restricted mothers. Moderate maternal protein restriction does however cause significant reduction in intestinal growth in newborn piglets which may decrease the neonatal digestive capacity.  相似文献

Studies were conducted to investigate the effects of long-term management practices on level and distribution of soil P and enzyme activities involved in P transformations in soil. Treatments included manure, P, NP, NPK, and NPK plus lime. Cattle manure was applied every 4 years at 269 kg N ha-1 for over a century and chemical fertilizers were applied every year at 67 kg N, 14.6 kg P, and 28 kg K ha-1 for over 69 years. Total soil P increase in soils ranged from 4.5 to 10.3 kg P ha-1 year-1 with the highest increase detected in the P-treated and the lowest in the manure-treated plot. Approximately 77-86% of the applied inorganic fertilizer-P in the past 69-71 years was recovered either in the harvested grain or remained in the top 30 cm of soil, while only 32% of the applied manure-P was recovered. Microbial biomass C and activities of alkaline phosphomonoesterase, phosphodiesterase, inorganic pyrophosphatase, and dehydrogenase were significantly higher in the soil treated with cattle manure. Acid phosphomonoesterase activity, however, was significantly higher in soils treated with chemical fertilizers. Results from this study suggested that manure-P is relatively more mobile than inorganic fertilizer-P. Long-term application of cattle manure promoted microbiological activities and P cycling, but did not result in P accumulation to levels close to those in inorganic P fertilizer-treated soils.  相似文献

Maximum benefit of a precise nitrogen application system for wheat   总被引:1,自引:0,他引:1  
Research is ongoing to develop sensor-based systems to determine crop nitrogen needs. To be economic and to achieve wide adoption, a sensor-based site-specific application system must be sufficiently efficient to overcome both the cost disadvantage of dry and liquid sources of nitrogen relative to applications before planting of anhydrous ammonia and possible losses if weather prevents applications during the growing season. The objective of this study is to determine the expected maximum benefit of a precision N application system for winter wheat that senses and applies N to the growing crop in the spring relative to a uniform system that applies N before planting. An estimate of the maximum benefit would be useful to provide researchers with an upper bound on the cost of delivering an economically viable precision technology. Sixty five site-years of data from two dryland winter wheat nitrogen fertility experiments at experimental stations in the Southern Plains of the U.S.A. were used to estimate the expected returns from both a conventional uniform rate anhydrous ammonia (NH3) application system before planting and a precise topdressing system to determine the value of the latter. For prices of $0.55 and $0.33 kg−1 N for urea-ammonium nitrate (UAN) and NH3, respectively, the maximum net value of a system of precise sensor-based nitrogen application for winter wheat was about $22–$31 ha−1 depending upon location and assumptions regarding the existence of a plateau. However, for prices of $1.10 and $0.66 kg−1 N for UAN and NH3, respectively, the value was approximately $33 ha−1. Pharmacokinetic determinants of spiramycin and its distribution into the respiratory tract were studied in 2 groups of calves, 4 to 10 weeks old. Group-A calves (n = 4) were used to determine pharmacokinetic variables of spiramycin after IV (15 and 30 mg/kg of body weight) and oral administrations of the drug (30 mg/kg) and to measure distribution of spiramycin into nasal and bronchial secretions. Group-B calves (n = 4) were used to determine distribution of spiramycin into lung tissue and bronchial mucosa. Spiramycin disposition was best described by use of an open 3-compartment model. Mean (+/- SD) elimination half-life was 28.7 +/- 12.3 hours, and steady-state volume of distribution was 23.5 +/- 6.0 L/kg. Bio-availability after oral administration was 4 +/- 3%. High and persistent concentrations of spiramycin were achieved in the respiratory tract tissues and fluids. Tissue-to-plasma concentration ratio was 58 for lung tissue and 18 for bronchial mucosa at 3 hours after spiramycin administration and 137 and 49, respectively at 24 hours. Secretion-to-plasma concentration ratio was 4 for nasal secretions and 7 for bronchial secretions, and remained almost constant with time. Thus, spiramycin penetrates well into the respiratory tract, although the value in bronchial secretions is lower than that in lung tissues and bronchial mucosa. Calculations indicate that a loading dose of 45 mg/kg, administered IV, followed by a maintenance dose of 20 mg/kg, IV, once daily is required to maintain active concentrations of spiramycin against bovine pathogens in bronchial secretions.  相似文献

AIM: To detect the presence of bovine herpesvirus (BoHV) type 4 in New Zealand dairy cows with clinical metritis.

METHODS: Serum samples taken from 92 dairy cows with clinical metritis, each from a different farm, were tested for the presence of antibodies against BoHV-4 using a commercially available, indirect ELISA. Peripheral blood mononuclear cells (PBMC) were collected from 10 BoHV-4 seropositive cows, and PBMC were examined by a pan-herpesvirus nested PCR to detect herpesvirus. PCR products were sequenced directly and a proportion of the PCR products were cloned and sequenced to identify the virus present.

RESULTS: Antibodies to BoHV-4 were detected in 23/92 (25%) serum samples. The pan-herpesvirus PCR was positive in 8/10 PBMC samples. Cloning and sequencing identified that all of the eight PCR-positive PBMC contained bovine lymphotropic herpesvirus (BLHV); no BoHV-4 DNA was detected.

CONCLUSIONS: This study reports the finding of the presence of apparent antibodies to BoHV-4, and BLHV DNA in New Zealand dairy cows affected by metritis.

CLINICAL RELEVANCE: Bovine herpesvirus type 4 and BLHV are reported to have the potential to cause reproduction failure in cows. This is the first report of apparent BoHV-4 antibodies, and BLHV in New Zealand. The importance and epidemiology of these viruses in cattle in New Zealand requires further investigation.  相似文献

AIMS: To determine immune responses, and the localisation and persistence of Mycobacterium bovis bacille Calmette-Guérin (BCG) in gut-associated lymphoid tissues (GALT) and other organs in possums vaccinated orally with lipid-formulated BCG vaccine. To determine the duration of excretion and longevity of survival of BCG in the faeces of vaccinated animals.

METHODS: Possums (n=28) were vaccinated with lipid-formulated BCG (1 x 10 8 colony forming units (cfu) of formulated BCG) by the oral route. Control possums (n=17) were fed oral bait pellets containing formulation medium only. Possums were sacrificed at 3 days and at 1, 3, 6 and 8 weeks after vaccination or ingestion of bait. Proliferation responses to bovine purified protein derivative (PPD) were measured in lymphocytes from blood and mesenteric lymph nodes (MLN) and samples of lung, spleen, liver, MLN and Peyer's patches (PP) were cultured for the presence of BCG. The number of BCG organisms excreted in faeces and the duration of excretion were determined in eight vaccinated possums and eight control possums over a 3-week period. In a separate experiment, a further six possums were vaccinated with oral BCG vaccine (5–10 x 10 8 cfu BCG/possum) and their faeces collected over 48–72 h, for culture of BCG. The longevity of survival of BCG in these faeces was determined by storing faecal samples (n=12) under three different conditions: in an incubator (22.5°C), and conditions which simulated the forest floor and open pasture. A proportion (1–2 g) of these faecal samples was collected after storage for 1, 3, 5, 8 or 20 weeks, and cultured for BCG.

RESULTS: Possums vaccinated orally with BCG vaccine showed strong proliferation responses to bovine PPD in peripheral blood lymphocytes at 6–8 weeks post-vaccination (p.v.). Positive lymphocyte proliferation assay (LPA) responses to bovine PPD were first evident in MLN at 3 weeks p.v. BCG was cultured from MLN and PP in a proportion of animals at 3–8 weeks p.v. BCG was not cultured from sections of spleen, lung or liver at any time p.v. BCG was recovered in low to moderate numbers from the faeces of vaccinated possums for up to 7 days, and maximal numbers were cultured in faeces collected 48–72 h p.v. After storage for 1 week, BCG was cultured from all faecal samples placed in the incubator and from a proportion of faeces exposed to conditions similar to those on the forest floor and pasture. With the exception of one faecal sample stored under forest floor conditions which was culture-positive for BCG at 3 and 5 weeks, BCG was not cultured from any other faecal sample stored for more than 1 week.

CONCLUSIONS: Ingestion of oral BCG vaccine by possums was associated with the development of strong cell-mediated immunity in both blood and MLN. Following oral vaccination with BCG, the organisms were localised and persisted in GALT but did not spread to the spleen, liver or lungs. BCG was shed in low to moderate numbers in the faeces for up to 7 days p.v. The viability of BCG excreted in faeces decreased rapidly, particularly when faeces were exposed to an open pasture environment. Oral vaccination of possums with formulated BCG is unlikely to result in undue contamination of the environment with BCG.  相似文献

AIMS: To determine factors that may influence the efficacy of an oral pelleted vaccine containing Mycobacterium bovis bacille Calmette-Guérin (BCG) to induce protection of brushtail possums against tuberculosis. To determine the duration of protective immunity following oral administration of BCG.

METHODS: In Study 1, a group of possums (n=7) was immunised by feeding 10 pellets containing dead Pasteur BCG, followed 15 weeks later with a single pellet of live Pasteur BCG. At that time, four other groups of possums (n=7 per group) were given a single pellet of live Pasteur BCG orally, a single pellet of live Danish BCG orally, 10 pellets of live Pasteur BCG orally, or a subcutaneous injection of live Pasteur BCG. For the oral pelleted vaccines, BCG was formulated into a lipid matrix, and each pellet contained approximately 107 colony forming units (cfu) of BCG, while the vaccine injected subcutaneously contained 106 cfu of BCG. A sixth, non-vaccinated, group (n=7) served as a control. All possums were challenged by the aerosol route with a low dose of virulent M. bovis 7 weeks after vaccination, and killed 7–8 weeks after challenge. Protection against challenge with M. bovis was assessed from pathological and bacteriological findings.

In Study 2, lipid-formulated live Danish BCG was administered orally to three groups of possums (10–11 per group), and these possums were challenged with virulent M. bovis 8, 29 or 54 weeks later. The possums were killed 7 weeks after challenge, to assess protection in comparison to a non-vaccinated group.

RESULTS: The results from Study 1 showed that vaccine efficacy was not adversely affected by feeding dead BCG prior to live BCG. Feeding 10 vaccine pellets induced a level of protection similar to feeding a single pellet. Protection was similar when feeding possums a single pellet containing the Pasteur or Danish strains of BCG. All vaccinated groups had significantly reduced pathological changes or bacterial counts when compared to the non-vaccinated group. In Study 2, oral administration of Danish BCG induced protection against challenge with M. bovis, which persisted for at least 54 weeks after vaccination. Some protection was observed in possums challenged 54 weeks after vaccination, but this protection was significantly less than that observed in groups vaccinated 29 or 8 weeks prior to challenge. There was a strong relationship between the proportion of animals producing positive lymphocyte proliferation responses to M. bovis antigens and protection against challenge with M. bovis.

CONCLUSIONS: Factors considered potentially capable of interfering with vaccination, including feeding dead BCG to possums prior to feeding live BCG, feeding multiple doses of BCG at one time, and changing strains of BCG, were shown not to interfere with the acquisition of protective immune responses in possums. Protection against tuberculosis was undiminished up to 29 weeks after vaccination with BCG administered orally. It is concluded that vaccination of possums by feeding pellets containing BCG is a robust and efficient approach to enhance the resistance of these animals to tuberculosis.  相似文献

Using plant sensing to determine the amount of nitrogen (N) to apply has the potential to increase profits in wheat (Triticum aestivum) production by reducing N cost or by increasing grain yield. The objective of this paper was to determine if yields and profits from experimental trials that used a precision N applicator to apply N were significantly different from trials that applied pre-determined amounts of N. Across Oklahoma, USA, experiments were designed to test 10 N treatments that included two variable rate treatments (VRT), two uniform rate treatments (URT) where the level of N applied was based on optical reflectance measurements (ORM), and six conventional treatments (i.e., pre-determined uniform rates of N). Data included treatments during 2005–2009 from eight different locations. Results indicated no statistical difference in yields between the conventional treatments that apply 90 kg ha−1 of N and the VRT and URT treatments. On average, the conventional treatment that applied 90 kg ha−1 of top-dress N produced the largest yield, with a VRT treatment producing the third largest yield. Profits were calculated for each treatment using a partial budget. On average, the treatment that received 90 kg ha−1 of top-dress N was the most profitable even though the pre-plant N (anhydrous ammonia) had a cost advantage relative to top-dress N (urea and ammonium nitrate).  相似文献
