Egg yolk is routinely used as a cryoprotectant in semen extenders. However, it may contain cryoprotective antagonists, and there are hygienic risks associated with its use. Proteins of plant origin, like soya-lecithin, lack these hazards. The aim of this study was to use soya-lecithin as a cryoprotectant in extender and to investigate its effects on in vitro quality and in vivo fertility of buffalo semen. Semen from three buffalo bulls was frozen in tris-citric extender containing 5.0%, 10% or 15% soya-lecithin or 20% egg yolk. Sperm motility, plasma membrane integrity and viability were assessed post-dilution, pre-freezing and post-thaw. In Post-dilution and pre-freezing, the values for motility, plasma membrane integrity and viability remained higher (p ≤ 0.05) in extenders containing 10% soya-lecithin and control compared with extender containing 5% and 15% soya-lecithin. However, motility, plasma membrane integrity and viability were higher (p < 0.05) in extender containing 10% soya-lecithin compared with control and extenders containing 5% and 15% soya-lecithin. Semen from two buffalo bulls was frozen in tris-citric extender containing either 10% soya-lecithin or 20% egg yolk. Higher (p < 0.05) fertility rate was recorded in buffaloes inseminated with semen containing 10% soya-lecithin (56%) compared with 20% egg yolk (41.5%). The results suggest that 10% soya-lecithin in extender improves the freezability and fertility of buffalo bull spermatozoa and can be used as an alternate to egg yolk in cryopreservation of buffalo semen.  相似文献

The present study was undertaken to investigate the genetic nature of zinc (Zn) accumulation in the ear‐leaves of maize (Zea mays L.) plants. Fourteen inbred lines were evaluated. Out of which, five inbreds were chosen, two high Zn accumulators (Rg‐5 and Rd‐2), two low Zn accumulators (Rg‐8 and G‐307) and a moderate one (K‐64). These five parental inbreds were utilized to obtain all possible ten F1 hybrids (in one direction), 10 F2, 10 Bl, and 10 B2. All genotypes were planted and Zn was estimated at 50% tasseling stage. Zinc accumulation in the present genotypes was found to be genetically controlled and affected by additive genes. Significant values were obtained for the General Combining Abilities (GCA) but not for the Specific Combining Abilities (SCA) and the high Zn accumulating parents were the best general combiners. Four genes were found to be the minimum segregating factors in the (high x low) crosses for Zn accumulation.  相似文献

A factor in Taenia multiceps coenurus fluid (TMCF) has previously been shown to modify the accessory activity of murine macrophages in vivo and in vitro. The factor (TMCF-F24) has been purified by ion exchange in a fast protein liquid chromatography (FPLC) system. This study was conducted to determine whether TMCF-F24 is an antigen in naturally occurring cerebral coenuriasis, and whether it can also modify normal sheep blood monocytes. Specific IgG antibodies to TMCF-F24 were detected, using ELISA, in serum and cerebrospinal fluid of sheep with clinical coenuriasis. Alterations in monocyte accessory activity were detected by an assay which measured the rate of increase in mitogen-induced lymphocyte transformation caused by addition of increasing numbers of the monocytes. Normal monocytes caused a positive increase in lymphocyte transformation. Monocytes incubated with TMCF-F24 caused progressive inhibition of transformation. This factor may therefore modify monocyte-T cell interaction in natural infection.  相似文献

This study was designed to compare the quality of liquid‐stored buffalo bull spermatozoa in soya lecithin based extender Bioxcell® (BIOX), milk (MILK), tris‐citric egg yolk (TEY) and egg yolk‐citrate (EYC) extender at 5°C. Semen was collected from five Nili‐Ravi buffalo (Bubalus bubalis) bulls of 6–7 years of age with artificial vagina over a period of 3 weeks (two consecutive ejaculates once in a week). Semen ejaculates having more than 60% motility were pooled, split into four aliquots, diluted (37°C; 10 × 106 motile spermatozoa/ml), cooled from 37 to 5°C in 2 h (0.275°C/min) and stored for 5 days. Sperm motility, viability, plasma membrane integrity (PMI) and normal acrosomal ridge were studied at first, third and fifth day of storage. Higher values of progressive sperm motility (%), sperm viability (%), sperm PMI (%) and normal apical ridge (%) were observed in BIOX, MILK and TEY extenders at first, third and fifth day of storage than EYC extender. Progressive sperm motility, sperm viability and sperm PMI in BIOX® extender were not different from MILK and TEY extenders at 1st and third day storage period. However, at fifth day of storage, the values for these parameters remained significantly higher (p < 0.05) in BIOX® compared with MILK, TEY and EYC extenders. At fifth day of storage, the semen quality parameters for Bioxcell® were comparable to those with MILK and TEY extenders at third day of storage. In conclusion, motility, viability and PMI of buffalo bull spermatozoa remained similar in Bioxcell®, milk and TEY extender at first and third days of storage at 5°C. Yet, the values for the aforementioned parameters in Bioxcell® were higher compared with milk, TEY and EYC extender at fifth day of storage at 5°C.  相似文献

Tomato wild relatives are important sources of resistance to many pests of cultivated tomato [Solanum lycopersicum L. (syn. Lycopersicon esculentum Mill.)]. Eleven wild tomato accessions previously identified at AVRDC—The World Vegetable Center as resistant to Bemisia tabaci were evaluated for resistance to the two-spotted spider mite [Tetranychus urticae (Koch.)] based on egg numbers using the leaf disc and Tanglefoot no-choice bioassays, and damage scores in choice bioassays. Highest resistance based on choice and no-choice bioassays was identified in AVRDC S. galapagense accessions VI057400, VI045262, VI037869 and VI037239, and S. cheesmaniae accession VI037240, all of which are new sources of T. urticae resistance. In addition, S. pimpinellifolium accession VI030462 exhibited resistance only in the no-choice bioassay based on egg numbers. Resistance to T. urticae based on the number of eggs from the no-choice bioassays was positively correlated with density of type IV glandular trichomes and negatively correlated with densities of type V trichomes. All resistant accessions accumulated high levels of total acylsugars, which were positively associated with type IV trichomes. There was a significant negative relationship between acylsugar content and T. urticae egg numbers from the no-choice bioassays. There was high correlation between the results from the leaf disc test and the Tanglefoot no-choice bioassay. These findings support the possible presence of broad-based insect and mite resistance in accessions closely related to cultivated tomato.  相似文献

The tomato leaf miner (Tuta absoluta) is a serious pest of tomato (Solanum lycopersicum) in the tropics and subtropics. Previous World Vegetable Center studies identified selected accessions of S. galapagense, S. cheesmaniae and S. pimpinellifolium that were resistant to whitefly (Bemisia tabaci Genn.) and spider mite (Tetranychus urticae Koch). Here, we evaluated these accessions for resistance to T. absoluta based on the number of eggs from choice bioassays, and larval mortality and adults emerged percentages in no‐choice feeding bioassays at WorldVeg Eastern and Southern Africa (WorldVeg) and the International Centre for Insect Physiology and Ecology (icipe). At WorldVeg, S. galapagense VI063177 exhibited high resistance in both choice and no‐choice bioassays. There was strong negative correlation between larval mortality and adults emerged percentages in the no‐choice feeding bioassays. Results from the icipe experiments were consistent with those of the WorldVeg screening, except for S. pimpinellifolium accession VI030462 , which was susceptible at icipe. Tuta absoluta is rapidly spreading and the resistance sources reported here will be valuable in breeding tomato varieties resistant to this insect and others.  相似文献
