Efficient water and fertilizer use is of paramount importance both in rain-fed and irrigated rice cultivation systems to tread off between the crop water demand during the dry spell and the fertilizer leaching. This lysimeter study on paddy in a lateritic sandy loam soil of the eastern India, to simulate the water and solute transports using the HYDRUS-1D model, reveals that this model could very well simulate the soil depth-specific variations of water pressure heads and nitrogen (N) concentrations with the efficiency of >86 and 89%, respectively. The change in the level of water ponding depth did not have a significant effect on the time to peak and the temporal variability of N concentration in the bottom soil layer. The lysimeter-scale water balance analysis indicated that the average deep percolation loss and crop water use were 35.01 ± 2.03 and 39.74 ± 1.49% of the total water applied during the crop growth period, respectively. Similarly, the amount of N stored in the plant and lost through soil storage, deep percolation, and other losses (mineralization, denitrification, and gaseous N loss to the atmosphere through plant leaves) were 1.60 ± 0.16, 0.17 ± 0.04, 12.00 ± 0.48, and 86.23 ± 0.41% of the total applied nitrogen, respectively. The simulation results reveal that a constant ponding depth of 3 cm could be maintained in paddy fields to reduce the N leaching loss to 7.5 kgN/ha. 相似文献

Herd and individual animal prevalence along with associated risk factors of bovine brucellosis was assessed on dairy farms located in 22 districts in Haryana and Punjab of India. Examination of 4,580 female animals of 119 dairy farms by Rose Bengal test and enzyme-linked immunosorbent assay revealed that an overall herd prevalence was 65.54 % (78/119) and individual animal prevalence was 26.50 % (1,214/4,580). Herd prevalence in Haryana was 62.79 % which was not significantly different (P?=?0.4208) from Punjab (72.72 %). Individual animal prevalence was 34.15 % in Punjab which was significantly higher (P?<?0.0001) than Haryana (22.34 %). Prevalence of brucellosis on dairy farms was detected in 20 of the 22 districts, of which 12 districts were in Haryana and 8 in Punjab. Risk factors such as species of animals, age of animals, herd size, awareness of dairymen for brucellosis, entry of a purchased animals on the farm, abortion on farm, use of calving pens, type of breeding, and type of farming were analyzed. The study indicated that the odds ratio (OR) was significant with risk factors of species (OR?=?1.63; 95 %CI?=?1.40–1.90; P?<?0.0001), age (OR?=?0.22; 95 %CI?=?0.17–0.29; P?<?0.0001), awareness of brucellosis among dairymen (OR?=?21.65; 95 %CI?=?2.63–178.04; P?=?0.0042), entry of purchased animal on the farm (OR?=?9.16; 95 %CI?=?2.38–35.18; P?=?0.0012), abortion in animals (OR?=?5.57; 95 %CI?=?1.92–16.10; P?=?0.0015) and on cow farm (OR?=?3.43; 95 %CI?=?1.33–8.82; P?=?0.0105). A buffalo disease, called "Degnala", causing lameness, edema, gangrenous ulceration of hooves or tail, emaciation, recumbency and eventual death, occurs in Eastern Nepal. Clinical examinations manifested lice eggs on hairs, bradycardia, hypothermia, dehydration, exanthema and icterus. Hematologically, increase of band neutrophil, giant platelet, hypoalbuminemia and hyperglobulinemia were characteristics. The status of cell-mediated immunity (CMI) after pesticide exposure was assessed in mice with the help of skin sensitivity and graft versus host reaction tests. It was observed that at 24 hours post-challenge CMI values did not differ significantly from the control, indicating no effect of quinalphos treatment in mice. Goats receiving monocrotophos at a dose rate of 1.0 mg kg-1 body mass for 40 days gave a similar result when CMI was tested with the help of the chemical sensitizer dinitrofluorobenzene (DNFB). Protection of Procambarus clarkii against white spot syndrome virus using recombinant subunit injection vaccine expressed in Pichia pastoris 总被引:1,自引:0,他引:1
ABSTRACT: The potentiality of injection vaccine against white spot syndrome virus (WSSV) in crayfish Procambarus clarkii was investigated. WSSV envelope proteins VP19 and VP28 were expressed in yeast Pichia pastoris GS115. The purified recombinant proteins (2 µg/g of crayfish) were injected intramuscularly, and the same dose injected as a booster shot on fifth day after vaccination. The vaccinated crayfish were divided into two even groups and later challenged orally by WSSV-infected dead crayfish muscle (2 g/individual) on the third and 21st days after the booster shot. The relative percent survival (RPS) in the third-day group was the highest in VP28 (91%), followed by VP19 + VP28 (84%), and VP19 (45%). The RPS for the 21st-day group was the highest in VP28 (78%), followed by VP19 + VP28 (76%), and VP19 (17%). Development of vaccine by using recombinant proteins VP19 and VP28 expressed in yeast is feasible. 相似文献

The growth hormone (GH) gene isolated and cloned from various Labeo species (L. rohita, L. calbasu, L. fimbriatus, L. gonius, L. bata, and L. kontius) is shown to contain a single copy in the haploid genome, with an overall size of ∼2.5 kb. The GH gene in all the Labeo species studied has five exons and four introns of various sizes with the exon/intron boundary sequence of GT/AG. The length
variation of the GH gene between the species is found to be due to length variation in the form of several deletions in the
third intron. The length of individual exons is the same in all the species with an open reading frame (ORF) of 630 bp (210
amino acids) except in L. rohita, which has a 9 bp deletion in the fourth exon, resulting in a shorter GH of 621 bp (207 amino acids). The similarity in the
nucleotide and amino acid sequences between the different Labeo species is greater than 97%, in spite of eight amino acids being altered in the GH protein of Labeo that reside outside the conserved domain sequence required for its function. Nucleotide substitutions are seen in the form
of 20 transitions and three transversions in the ORF of the GH gene. Both types of transitions (A–G; T–C) and only one type
of transversion (A–C) are detected in the GH gene. Codon preference in GH gene shows a strong preference for G and C in the
wobble position of the codons. Genetic interrelationships determined between Labeo and other species of fishes using nucleotide sequence of GH cDNA supports the overall teleost classification of Nelson (Fishes
of the World. Wiley, New York, 1984) with separate clades for Ostariophysi, Protacanthopterygii, and Acanthopterygii. Besides, the unweighted pair group method
with arithmetic means (UPGMA) analysis clearly distinguishes between the species having five exons and four introns in the
GH gene from the species having six exons and five introns in the same gene. The Labeo species analyzed in the present study could be clustered into two groups using the maximum-parsimony method on the intron

Periodic changes in reproductive hormone levels, gonadal histology and gonadosomatic index (GSI) of snow trout, Schizothorax richardsonii, were examined to ascertain annual cycle of gonadal development and reproductive status in their natural habitat. In females, there were coherent changes in plasma 17β‐oestradiol and vitellogenin along with GSI, oocyte maturation and vitellogenic progression, collectively indicating two distinct maturation peaks during the months of September and February. Coinciding with this, in males, plasma 11‐keto testosterone was also noticeably higher during September and February, with highest GSI values in September. However, plasma 17α, 20β‐dihydroxyprogesterone levels in males were found to be persistently high from September to February. This observation suggests the potential presence of matured oozing males over a longer period, unlike in females. Overall, the close association between reproductive hormone levels, GSI and gonadal maturation stages in males and females (particularly, the presence of postovulatory follicle complexes) with apparent natural synchronization clearly indicates that S. richardsonii breeds twice in a year, possibly during late September to early November and late February to early April in the coldwater riverine habitats of the Indian Himalayan region. 相似文献

Calli were obtained from leaf, cotyledon and internode explants of in vitro-grown plants of Indian cultivar of Withania somnifera in MS medium supplemented with 2, 4-D (2.0 mg l−1) and Kinetin (0.2 mg l−1). The brown, semi-friable callus (500 mg FW) derived from leaf explants produced higher number of primary adventitious roots (9 roots/callus) in half strength MS medium fortified with IBA (0.5 mg l−1) and NAA (0.1 mg l−1). The primary adventitious roots with an inoculum mass of 15 g FW were cultured for 6 weeks in the same medium for secondary adventitious root proliferation. Elicitation of abiotic elicitor, aluminium chloride at 10 mg l−1 at the end of 4 weeks culture with 4 h exposure time enhanced withanolides productivity. Under similar culture conditions, the biotic elicitor, chitosan at 100 mg l−1 stimulated higher production of all withanolides when compared to aluminium chloride treatment. This is the first report on the use of callus-derived adventitious root culture for the enhanced production of withanolides upon chitosan elicitation. 相似文献
