The diagnosis of the chronic, human brucellosis is frequently difficult and usually needs experimental methods. This paper describes a lymphocyte stimulation test with a Brucella antigen and the results of this test concerning 45 brucellic or not brucellic patients. It is concluded that this test is interesting, especially for the chronic and sero-negative Brucellosis diagnosis.  相似文献

This case report describes an 11-year-old Belgian Malinois dog with acute onset of cough caused by aspiration of barium-impregnated polyethylene spheres (BIPS) and pneumonia following an episode of suspected gastric dilation. Although bronchoscopic retrieval of the BIPS was largely unsuccessful, the dog recovered uneventfully, with most of the BIPS being coughed out and swallowed over a 1-month period. Aspiration of BIPS should be considered a potential complication of their administration. Furthermore, endoscopic removal of aspirated BIPS is challenging and may not be indicated because of their inert nature and possible self-clearance.  相似文献

Aged plants are more difficult to infect than young plantlets. This modification of susceptibility is described as mature plant resistance (MPR). For potato virus Y (PVY), MPR is known to lead to low infection rates of plants inoculated at the postflowering stage and a decrease in the number of infected daughter tubers. However, the impact of inoculation date on the capacity of PVY to accumulate in daughter tubers has not been studied so far. Field and greenhouse experiments were carried out to better understand PVY epidemiology and to help potato growers to evaluate consequences of early/late infections on the quality of their crops. In field trials, potato plants (cv. Bintje) were covered by insectproof nets from planting to harvest except for a 14-day period to expose plants to natural PVY infections. Under controlled conditions, potato plants were mechanically inoculated with PVY at different dates from preflowering stages (early inoculations) to postflowering stage (late inoculations). At harvest, daughter tubers were individually collected and analysed to define proportions and viral load of infected tubers according to the time between virus inoculation and harvest. Our results showed that although the age of plants at the time of inoculation can modify their susceptibility to PVY infection, in return, early and late PVY inoculations lead to similar rates of infected tubers at the plant scale and equivalent viral accumulation in infected tubers. All together, these data revealed that both early/late infections are high risks for the sanitary quality of potato tubers.  相似文献

Habitat reserves are a common strategy used to ensure viability of wildlife populations and communities. The efficacy of reserves, however, is rarely empirically evaluated. We examined the likelihood that small (650 ha), isolated habitat reserves composed of old-growth Sitka spruce (Picea sitchensis)-western hemlock (Tsuga heterophylla) rain forest (upland-OG) and mixed-conifer peatlands (peatland-MC) would sustain populations of northern flying squirrels (Glaucomys sabrinus) in the absence of immigration or emigration within the Tongass National Forest in Southeast Alaska. We used demographic data obtained from a study of flying squirrels on Prince of Wales Island in Southeast Alaska and litter size from flying squirrels in similar habitat to estimate per capita rate of increase (r) of flying squirrels in upland-OG (r = 0.14, SD = 0.42) and peatland-MC habitats (r = 0.01, SD = 0.39). Our results indicated that peatland-MC habitat was unlikely to sustain populations and that viability of flying squirrel populations in small habitat reserves largely depended on the upland-OG forest component. We subsequently estimated time to extinction (TN) based on r, its variance (v), and the potential population ceiling (K). We used TN to calculate the probabilities (Pt) that squirrel populations would persist in small reserves containing 100%, 50%, and 25% upland-OG habitat for 25, 50, and 100 years. In each scenario, we calculated TN and Pt for 2 levels of v. For the best-case scenario (100% upland-OG forest, lowest variance, t = 25 years), TN was 507 years and Pt was 0.95. For the worst-case scenario (25% upland-OG forest, highest variance, t = 100 years), TN was 237 years and Pt was 0.66. Minimum patch size of upland-OG forest required for a high probability (Pt = 0.95) of sustaining a flying squirrel population in isolation with relatively low demographic variability (v = 0.34) for 25, 50, or 100 years was 578, 5077, and 78,935 ha, respectively. We concluded that it was unlikely that small isolated habitat reserves could sustain populations of flying squirrels for >25 years in the absence of immigration. Consequently, dispersal among small reserves is critical to ensure that they function to support metapopulations of northern flying squirrels.  相似文献

Sixteen (mean age = 1.1 +/- 0.1 yr; mean BW = 478 +/- 34 kg) Brahman-influenced bulls were used to determine the influence of fescue type on sperm characteristics and serum concentrations of prolactin, cortisol, and testosterone. Bulls were blocked by BW, scrotal circumference (SC), and pregrazing sperm characteristics and randomly assigned to graze toxic endophyte-infected (EI; 4 bulls/pasture; 2 pastures) or novel endophyte-infected (NE; 4 bulls/pasture; 2 pastures) tall fescue for 121 d. Semen was collected by electroejaculation, and SC was measured and blood samples collected monthly. Sperm were evaluated for motility and morphology with an integrated visual optical system. Overall mean concentration of prolactin was decreased more (P < 0.01) in EI bulls than NE bulls from May to August. Scrotal circumference was not affected by fescue type (P = 0.58); overall SC averaged 36.7 +/- 2.3 cm. Percentage of live sperm was not different (P = 0.24) between NE bulls (80%) than EI bulls (67%) in July and August. Bulls grazing NE fescue had more (P < 0.06) motile sperm than EI bulls in July and August. Percentages of progressive (57 vs. 38%, NE and EI, respectively; P < 0.06) and rapid (67 vs. 46%, NE and EI, respectively; P = 0.04) sperm were greater from bulls grazing NE than EI bulls in July and August. Average velocity of the smoothed sperm path and progressive velocity in a straight line from the beginning to the end of the sperm track were slower (P < 0.09) in EI bulls than NE bulls and were slower (P = 0.04) in August compared with July. Mean width of head oscillation as the sperm swims was less (P < 0.06) in August than July. Concentrations of cortisol and testosterone were not (P > 0.10) influenced by fescue type. Semen from bulls grazing EI had reduced motility and morphology than bulls grazing NE. Detrimental effects of toxic fescue may not be mediated by cortisol, testosterone, or both. Semen quality of bulls grazing toxic EI tall fescue was decreased with increased maximum ambient temperatures.  相似文献

Intraperitoneal [i.p.] and subcutaneous [s.c.] administration to BALB/C mice with a single dose of 5 mg/kg body weight (wet weight) of live Mycobacterium chelonae (Mch) augmented splenocyte blastogenesis. Similar increases in splenocyte blastogenesis manifested during a single oral administration to mice with 100 mg/kg body weight (wet weight) of this Mycobacterium. When splenocytes issued from these mice are activated by mitogens, a highly significant enhancement of lymphoblastic transformation was observed. On the other hand, multiple oral administrations with 50 mg/kg body weight (wet weight) to Mch/dose did not manifest statistically significant differences in splenocyte blastogenesis as compared to controls. Meanwhile, a highly increased transformation of splenocytes, issued from such mice, is observed in response to lectins and to the mitogenic effect of this microorganism. Splenocyte counts have shown 44.5, 37.6, and 23.2% increases in response to i.p., s.c. and multiple oral administration of this bacterium, respectively, as compared to solvent controls. Repeated s.c. administration of this mycobacterium manifested short lived and weak syndromes of anaphylactic shock during and immediately after the second inoculation of Mch. This phenomenon is not observed during repeated intraperitoneal and oral administrations. In conclusion, parenteral (i.p. and s.c.) and oral administration of Mch stimulates the immune system of mice. This effect is characterised by increased in vivo cell multiplication and by enhanced ex vivo DNA synthesis of murine splenocytes. The need of further studies is eminent to elucidate classes of immunocompetent cells involved in this phenomenon.  相似文献

The identification and the numeration of the lymphocytes in cattle is realized essentially by revealing the membranary properties of these cells: receptors for some lectins (i.e. peanut agglutinin or phytohemagglutinin), receptors for sheep red blood cells (SRBC), surface-specific antigens concerning T-lymphocytes and surface immunoglobulins concerning B-lymphocytes. The technique of the production in the goat of a specific anti-bovine T-lymphocyte antiserum (GABTA) is described. The GABTA shows a specific cytotoxic activity on 55% of the peripheral blood lymphocytes and more than 95% of the thymocytes. It inhibits, at 90%, the formation of the rosetting with SRBC. This reagent allows identification and a common and quick numeration of the T-lymphocytes in cattle.  相似文献

Immunomodulatory properties of a strain of live Mycobacterium chelonae (Mch) was investigated in an in vitro lymphocyte transformation system. Murine splenocyte activation by this bacterium was characterized by polyclonal lymphoproliferative responses in a dose dependent fashion. Optimal doses ranging from 20 to 80 micrograms of Mch (wet weight) per ml of cell suspension induced a very significant mitogenic effect. Higher doses (100 micrograms) of Mch manifested a decreased rate of tritiated thymidine ([3H] TdR) uptake whereas responsiveness of splenic lymphocytes to lower doses (0.156 microgram) was not modified. Contrary to the splenocyte responses activation of murine thymocytes by this mycobacterium is characterised by a decreased proliferation as compared to the background count of unstimulated cells. Simultaneous addition of Mch with optimal doses of Concanavalin A (Con A) and Phytohemaglutinin (PHA) potentiated polyclonal mitogenic responses of murine splenocytes to these two lectins. However, proliferation of these lymphocytes to Lypopolysaccharide (LPS) induction was not modified. BALB/C and DBA/2 spenocytes were found to be more responsive to stimulation by this Mycobacterium as compared to those of C3H/Ou and to a lesser degree to those of C57BL/6 mice.  相似文献

This report describes the fecalith‐induced intestinal obstruction of a free‐ranging red diamond rattlesnake (Crotalus ruber) and the snake's subsequent history following surgical removal of the fecalith. The captured snake exhibited an abnormally distended abdomen and an extremely hard mass, detected via palpation, near its vent. Coeliotomy yielded a 2.5‐cm, 5‐g fecalith from the large intestine. Microscopic dissection of the fecalith revealed no evidence of gastrointestinal parasitic worms. Subsequently, we implanted a radio‐transmitter that allowed us to track the snake's movements for 7 months (until the radio signal vanished), indicating normal behaviour, complete recovery and good health apart from the obstruction. This observation suggests that fecalith development and intestinal obstruction represent potential risks of long‐term faecal retention, an unusual physiological trait well documented among rattlesnakes and other stout, heavy‐bodied terrestrial viperid snakes. Dehydration and decreased gut motility associated with brumation (≈hibernation) may predispose temperate snakes to fecalith formation. Regional drought and a small mammal diet with indigestible hairs might have also promoted fecalith formation in this specimen.  相似文献
