Abstract. The degree of protection in immunized rainbow trout against the infective larval stages of Diplostomum spathaceum was investigated. Immunized rainbow trout demonstrated a 70–80% reduction in the number of metacercariae establishing in their eye-lenses following challenge with live cercariae compared with unimmunized fish. Both normal trout serum and anti–diplostomule serum weTC cytotoxic to diplostomules in vitro . However, there was no correlation between the degree of protection and the level of serum antibody within individual fish. Reduced establishment of migrating diplostomules, indicative of in vivo killing, occurred in fish possessing activated macrophages and challenged with cercariae opsonized with specific antibody. An antibody–dependent cell–mediated cytotoxic (ADCC) mechanism is proposed, conferring protection in immunized fish. This is supported by the finding that cercarial and diplostomule antigens are capable of activating macrophages in immunized rainbow trout. The role of macrophage activation and specific antibody in conferring protection in immunized fish is discussed.  相似文献

During three consecutive summers, forty spring-calving beef cows and their calves grazed perennial ryegrass-dominant swards receiving 250 kg N ha?1 at one of two annual stocking rates [2.0 (SR 2.0) or 2.5 (SR 2.5) cows ha?1] and one of two sward heights [4–5 (LS) or 7–8 (HS) cm] in a 2 × 2-factorial experiment, replicated twice. Sward heights were maintained from turn-out in spring by weekly adjustment of the area grazed and herbage was cut for silage in June and again in August from the areas not grazed. After the second cut of silage there was no control of sward height. Calves were weaned in early October and cows removed from pasture and housed when sward heights fell to 4 cm in autumn. Cows were fed in groups a variable but measured quantity of silage during winter to achieve a body condition score of 2.0–2.25 at turn-out the following spring. During the period of sward height control the cows on the HS treatment gained more live weight than those on the LS treatment (0.841 vs. 0.496 kg day?1; P<0.01) as did the calves (1.167 vs. 1.105; P<0.05). but the stocking rate treatment had no effect. From the time of second-cut silage to the time of weaning and housing respectively, calves and cows gained more live weight on the SR 2.0 treatment because sward heights were higher. Reproductive performance of cows was not affected by treatment. The quantity of silage produced and consumed per cow was not affected by sward height treatment, but the SR 2.0 treatment produced significantly (P<0.001) more silage (1559 kg dry matter per cow) than the SR 2.5 treatment (833 kg dry matter per cow) and had higher winter silage requirements (1249 vs. 1153 kg dry matter per cow: P<0.05). The overall mean stocking rate at which winter fodder production and requirements would be in balance was calculated as 2.25 cows ha?1 but values were 1.86, 2.60 and 2.28 in each of the three years of the experiment. The results showed that it was possible to control sward height in temperate beef cow systems by adjusting the area available for grazing. Body condition score can be used as a means of determining the feeding levels required to manipulate body condition of cows over winter to achieve prescribed levels of body condition. The experimental approach allows the identification of the stocking rate at which self-sufficiency in winter fodder can be achieved and the year-to-year variation associated with that stocking rate. This approach could be generalized if information on herbage growth rate were available, either from direct measurement or from predictive models.  相似文献

Seed of Adonis microcarpa (pheasant's eye) fed at 5.6 g/kg of the diet induced virtually total feed refusal within 3 d in growing and finishing pigs. It also caused vomiting, rapid and shallow breathing and death in a minority. These effects were probably caused by cardiac glycosides whose structure and effects closely resemble those of digoxin. Feed intake and growth recovered within 2 weeks of removal of the seed.  相似文献

Protection of pregnant swine by vaccination against leptospira infection   总被引:2,自引:0,他引:2  
SUMMARY The protection conferred on pregnant gilts by 2 commercially available Leptospira interrogans serovars pomona and tarassovi bacterins was evaluated. Gilts vaccinated either 3, 6 or 12 months prior to natural challenge with L. interrogans serovar pomona had significantly lower abortion rates (2% vs 69%) and foetal mortality rates (14% vs 57%) than unvaccinated controls. One vaccine was significantly superior to the other and contained approximately twice the number of L. interrogans serovar pomona organisms per vaccine dose. Neither vaccine protected against renal colonisation but vaccination reduced urinary excretion of leptospires. Both vaccines reduced agglutinating antibody response to infection, as measured by the microscopic agglutination (MA) test. This may prevent the detection of a carrier animal by serology. Foetal pigs did not develop specific MA titres. Cultural methods were not reliable in making a diagnosis of foetal infection. Histopathology of foetal liver and kidneys helped in making a diagnosis of foetal infection.  相似文献

The LH surge induces functional and morphological changes in granulosa cells. Mechanistic target of rapamycin (mTOR) is an integrator of signalling pathways in multiple cell types. We hypothesized that mTOR kinase activity integrates and modulates molecular pathways induced by LH in granulosa cells during the preovulatory period. Cows were ovariectomized and granulosa cells collected at 0, 3, 6, 12 and 24 hr after GnRH injection. While RHEB mRNA levels increased at 3 and 6 hr, returning to basal levels by 12 hr after GnRH treatment, RHOA mRNA levels increased at 6 hr and remained high thereafter. Western blot analyses revealed increased S6K phosphorylation at 3 and 6 hr after GnRH injection. Similarly, mRNA levels of ERK1/2, STAR and EGR‐1 were higher 3 hr after GnRH treatment. Rapamycin treatment inhibited mTOR activity and increased AKT activity, but did not alter ERK1/2 phosphorylation and EGR1 protein levels in cultured bovine granulosa cells. Rapamycin also inhibited LH‐induced increase in EREG mRNA abundance in granulosa cells in vitro. However, intrafollicular injection of rapamycin did not suppress ovulation. These findings suggest that mTOR is involved in the control of EREG expression in cattle, which may be triggered by LH surge stimulating RHEB and S6K activity.  相似文献

An experiment was conducted to assess the effects of grazing a perennial ryegrass (Lolium perenne) / white clover (Trifolium repens) sward by sheep or goats on sward composition and structure and on subsequent diet selection, herbage intake and liveweight gain by weaned lambs. From mid-May to late July (phase 1), ewes with twin lambs or yearling Scottish Cashmere goats grazed continuously swards maintained at 4- or 8-cm sward surface height. From mid-August to the end of September (phase 2), weaned lambs continuously grazed the same swards maintained at 4 cm (treatment 4–4) or at 8 cm (treatment 8–8) or which had been allowed to increase from 4 cm to 8 cm (treatment 4–8). By the end of phase 1, swards grazed by goats had higher proportions of white clover in the whole sward (0.377 vs. 0.181; s.e.d 0.0382; P < 0.001) than those grazed by sheep, irrespective of sward height treatment. This resulted in phase 2 in a higher proportion of white clover selected ( P <0.001), higher herbage intakes ( P < 0.001) and higher liveweight gains ( P < 0.001) by weaned lambs grazing swards previously grazed by goats compared with those previously grazed by sheep. There were higher proportions of clover present in the swards from treatment 4–8 at the beginning of phase 2 compared with the other sward height treatments and consequently weaned lambs had, on this treatment, a higher proportion of clover in their diet ( P <0.001), higher herbage intakes ( P <0001) and higher liveweight gains ( P <0.001). It is concluded that goats can be integrated into sequential grazing systems with sheep on grass/clover swards and this can result in an increase in the proportion of clover in swards and increased sheep performance.  相似文献

The objective of this study was to evaluate the effect of retinol (RT) and retinoic acid (RA) on the in vitro development of pre‐implantation goat embryos cultured in potassium simplex optimized medium or synthetic oviduct fluid or cocultured in oviductal cells monolayer either in potassium simplex optimized medium or synthetic oviduct fluid. A total of 2407 cumulus‐oocyte complexes were aspirated from 2 to 6 mm ovarian follicles from slaughtered animals. Selected cumulus‐oocyte complexes were subjected to in vitro maturation in TCM 199 for 24 h at 39°C in an atmosphere of 5% (v/v) CO2 in humidified air. In vitro fertilization was performed in modified defined medium. Eighteen hours after in vitro fertilization, cumulus cells were removed and presumptive zygotes were randomly distributed into experimental groups. In Experiment 1, presumptive zygotes were cultured in potassium simplex optimized medium, potassium simplex optimized medium + RT, potassium simplex optimized medium + retinoic acid, synthetic oviduct fluid, synthetic oviduct fluid + RT and synthetic oviduct fluid + RA at 39°C in a humidified atmosphere of 5% (v/v) CO2, 5% (v/v) O2 and 90% (v/v) N2. In Experiment 2, presumptive zygotes were cocultured in potassium simplex optimized medium + oviductal cells monolayer, potassium simplex optimized medium + RT + oviductal cells monolayer, potassium simplex optimized medium + RA + oviductal cells monolayer, synthetic oviduct fluid + oviductal cells monolayer, synthetic oviduct fluid + RT + oviductal cells monolayer and synthetic oviduct fluid + RA + oviductal cells monolayer in an atmosphere of 5% (v/v) CO2 in humidified air. In both experiments, media were partially changed on day 2 after in vitro fertilization and unfertilized oocytes were excluded from the experiment. Embryos were cultured or cocultured for 8 days. In Experiment 1, there was no effect of RT or RA supplementation on the proportion of oocytes that reached the morula or blastocyst stages. By contrast, Experiment 2 demonstrated that the addition of 0.28 μg/ml RT and 0.5 μm RA to the embryo culture media stimulated (p < 0.05) development to the morula and blastocyst stages under the coculture conditions tested. In conclusion, retinoids play an important role in pre‐implantation development of goat embryos and can be used to enhance in vitro embryo production.  相似文献
