Genetic variation within and among several Sorghum populations from different agroecological zones in Malawi were investigated using random amplified polymorphic markers (RAPDs). DNA samples from individual plants were analyzed using 35 oligonucleotides of random sequence. Twenty five of these primers allowed amplifications of random polymorphic (RAPD) loci. Overall, 52% of the scored loci were polymorphic. Every accession was genetically distinct. The analysis of molecular variance revealed that the within-region (among accessions) variations accounted for 96.43% of the total molecular variance. Observed variations in allelic frequency was not related to agroecological differences. The degree of band sharing was used to evaluate genetic distance between accessions and to construct a phylogenetic tree. Further analysis revealed that the sorghum accessions analyzed were genetically close despite considerable phenotypic diversity within and among them. It is suggested that all the sorghum landraces currently available in Malawi should be conserved both ex situ and in situ to maintain the current level of genetic diversity.  相似文献

Genetic variation and genetic structure of black spruce (Picea mariana L.) populations growing in wet land (lowlands) and dry lands (uplands) with different levels of metal contaminations were analyzed using ISSR. Polymorphic loci (P%) ranged from 65% to 90% with a mean of 75%. Nei??s gene diversity (h) varied from 0.264 to 0.359 with a mean of 0.310, and Shannon??s index (I) ranged from 0.381 to 0.524 with a mean of 0.449. The level of genetic variation was higher in populations from wet lands than those from dry lands. Variation within populations accounts for most of total genetic variation. The genetic distance among the black spruce (P. mariana) populations ranged from 0.171 to 0.351. The present study indicates that genetic variation and long-term exposure to metals (more than 30 years) are not associated. Cytological analysis of black spruce seeds from metal-contaminated and -uncontaminated areas showed normal mitotic behavior during prophase, metaphase, anaphase, and telophase.  相似文献

Though important, CO2 effluxes from non-agricultural ecosystems are poorly characterised. Given that thicket vegetation is prevalent in Eastern Cape, South Africa, we monitored soil CO2 effluxes from an intact thicket, degraded thicket (DT) and grassland (G) over 10 months, as affected by temperature, moisture and penetration resistance (PR). High soil moisture (16%) reduced PR (4?kg cm?2), which raised winter effluxes (1.2 µmol m?2 s?1), while low moisture (2%) resulted in hard dry soil (14?kg cm?2) that suppressed spring effluxes (0.2 µmol m?2 s?1). There was good interaction between PR and moisture (r = ?0.53), with seasonal effluxes increasing with increasing moisture (r = 0.9, p = 0.0001) and decreasing PR (r = ?0.66, p = 0.02). Temperature effects were significant under unlimited moisture supply. Thus high summer temperature (40 °C) gave lower effluxes in DT and G (<1 µmol m?2 s?1) due to limited moisture (<10%), whereas high autumn temperature (48 °C) and good moisture (16%) accelerated CO2 emissions (averaging 2 µmol m?2 s?1) from all covers. Although semi-arid ecosystems are limited by low moisture and sandy soils, they contribute to CO2 emissions under high moisture and increasing temperature. Quantification of soil carbon (C) and nitrogen (N) fractions in grasslands is vital for estimating C sequestration and climate change studies. We quantified background soil total carbon (TOC) status, recalcitrant carbon (RC), acid hydrolysable labile carbon (AHC), hot- and cold-water extractable carbon (HWC and CWC, respectively) fractions in the grassland in this study. Soil C fractions were as follows: TOC (11,633 to 15,525 mg C kg?1); RC (11,500 to 15,357 mg C kg?1); AHC (132 to 168 mg C kg?1); HWC (57 to 70 mg C kg?1); and CWC (27 to 33 mg C kg?1). Labile C fractions contributed at most 1.2% to total C. Concentrations of total N ranged from 1072 to 1230 g N kg?1. Recalcitrant C contributed higher amounts (>90%) to total C, indicating the incorporation of C into the stable C fraction. Carbon dioxide (CO2) and nitrous oxide (N2O) fluxes were not significantly correlated with total C.  相似文献

Soil Hydraulic Properties: Influence of Tillage and Cover Crops   总被引:1,自引:0,他引:1  
Understanding the effects of cover crops and tillage on soil physical properties is important for determining soil productivity. This study was conducted at Lincoln University's Freeman Center, USA to evaluate the effects of tillage and cover crop management on soil hydraulic properties. The field site included three replicate blocks in a randomized complete block design with each plot measuring 21.3 m in length and 12.2 m in width. Treatment factors were tillage at two levels(moldboard plow tillage vs. no tillage) and cover crop at two levels(cereal rye(Secale cereal) cover crop vs. no cover crop). Soil samples were collected in late spring/early summer from each treatment at 10-cm depth increments from the soil surface to a depth of 40 cm using cores(76.2-mm diameter and 76.2-mm length). Soil bulk density was 13% lower with tillage compared with no-tillage. Volumetric water content was significantly higher at 0.0 and -0.4 k Pa pressures with tillage compared with no tillage. Tillage increased the proportion of coarse mesopores by 32% compared with no tillage, resulting in 87% higher saturated hydraulic conductivity(K_(sat)). Cover crop increased the proportion of macropores by 24% compared with no cover crop; this can potentially increase water infiltration and reduce runoff. As a result of higher macroporosity, Ksat was higher under cover crop compared with no cover crop. This study demonstrated that tillage can benefit soil hydraulic properties in the short term, but these effects may not persist over time. Cover crops may slightly improve soil hydraulic properties, but longer term studies are needed to evaluate the long-term effects.  相似文献

Rye (Secale cereale L. and S. strictum) offers potential to increase the genetic variability and to introduce desirable characters for wheat improvements. Cytogenetic techniques have been used to screen wheat lines containing rye chromatin. These techniques are not adequate since they are highly technical and time consuming. They are not suitable for breeding programs that require rapid screening of large numbers of genotypes. The main objective of this study was to develop and characterize ISSR and SCAR markers that can distinguish wheat from rye genome. Total DNA from wheat, rye, and triticale accessions from different provenances were amplified with ISSR primers in PCR assays. Three wheat-diagnostic sequences were identified. In addition three rye-diagnostic ISSR markers of which, one marker specifically diagnostic for Secale strictum were characterized. Pairs of primers flanking these specific sequences were designed to produce SCAR markers. Two SCAR markers were rye genome-specific. One SCAR was present in all the seven rye chromosome, and another was specific to rye chromosomes two, three, four, and seven. These newly developed ISSR and SCAR markers should be useful to wheat breeders screening genotypes that may contain rye chromatins.  相似文献

Cowpea has been cultured from ancient timesin Africa and Asia and is now widespreadthroughout the tropics and subtropics.Cowpea accessions can be distinguishedphenotypically from one another by theirgrowth habit, time to maturity, yield, andseed size and colour. Data on geneticdiversity of farmer-developed accessionsare lacking. The main objective of thisstudy was to determine the pattern andextend of RAPD marker variation within andamong cowpea populations from differentagroecological zones and to determine thedegree of genetic relationships and geneflow among different landraces used bylocal farmers. Twenty of the 30 RAPDprimers tested allowed amplifications ofrandom polymorphic (RAPD) loci. Overall,80% of the scored loci were polymorphic. The degree of band sharing was used toevaluate genetic distance betweenaccessions and to construct a phylogenetictree. The genetic distance values amongaccessions varied between 0.09 to 0.57.Although some small clusters groupedaccessions of the same growth habits, ageneral lack of agreement betweenclustering and morphological features wasobserved. The analysis of molecularvariance revealed that within-region ortypes (among accessions) variationaccounted for 96% of the total molecularvariance. Utilization of the doubled haploid method of breeding usually shortens the time to cultivar release, and methods of haploid production need evaluation in a breeding programme. Thirty-eight different three-way crosses were tested for anther culture response. On average 5.8 percent of the anthers cultured produced calli. Three crosses were found recalcitrant for callus induction. Overall, the anther culture method produced 0.6 plantlet per 100 anthers cultured. Five crosses with an average of 5.8 and 2.8 percent of anthers producing calli and plantlets, respectively, were compared using anther culture and wheat × maize crosses. Non-responsive genotypes for callus induction and plantlet formation in the anther culture method proved to be good parental material in wheat × maize crosses. The average percentages of embryo formation and plantlet production in wheat × maize crosses were 10.3 and 4.7, respectively. Anther-derived plants were cytologically unstable, whereas all the plants regenerated from wheat × maize crosses were haploids (n = 21 chromosomes). The chromosome numbers of the polyhaploids were doubled with a colchicine treatment. Improvement of the two haploid production methods to facilitate their efficient use in a breeding programme is discussed.  相似文献

Common wheat × hexaploid triticale hybrids were produced and evaluated for tolerance to barley yellow dwarf virus disease (BYD). The BYD tolerance expression varied with wheat × triticale combination. The selection for BYD tolerance increased the recovery of tolerant genotypes in the next generations. Homozygous tolerant and susceptible lines were obtained in advanced generations. The rye chromosomes 1R, 2R, and 4R with 7R were transmitted as disomic or monosomic, disomic, and double disomic substitution to the late generations of ‘Musala’ (common wheat) בMuskox 658’ (triticale), ‘Encruzilhada’ (common wheat) בNord Kivu’ (triticale) and ‘Encruzilhada’× 12th. International Triticale Screening Nursery 267 (12ITSN267) (triticale), respectively. A clear association was established between the 1R chromosome of the ‘Muskox 658’ triticale line and the tolerance to BYDV. Results suggest that the 2R chromosome may be involved in BYD tolerance of ‘Nord Kivu’ triticale line.  相似文献
