Chromatin prepared by a method involving limited nuclease digestion contains the same repeating structure as chromatin in the nucleus, whereas chromatin prepared by conventional methods involving shear does not.     

Entamoeba histolytica--the virulence of imported strains

We tested the virulence of 15 strains of Entamoeba histolytica, imported to Czechoslovakia, by intracaecal inoculation of laboratory rats. According to the scoring system of Neal, none of the 15 strains possessed the virulence index greater than 2. This indicates that all the organisms tested should be classified as avirulent. However, it should be noted that all the strains produced infection of the caecum and thus should be considered infective for rat. For 7 strains, isoenzyme patterns were determined for PGM, HK and ME. One imported strain, obtained from student from Congo, demonstrated isoenzyme pattern for PGM and HK indicated that the strain was virulent. This organisms had the index of virulence 1.8 (avirulent) in animal experiment; it was isolated from cysts of clinically asymptomatic patient. Examination of the rectal mucosa of the donor of the strain indicated typical chronic catarrhal proctitis of mild degree. Examination of the patient's serum demonstrated the presence of anti E. histolytica antibodies by CIEP, while the ELISA test was negative. Twenty-one cyst carriers were examined by rectoscopy. Pathologic changes were observed in 20 of these, as follows: altered vascular structure (13X), roughened mucosa (12X), mucosal reddening (10X), decreased glistening (7X), mucus in mucosa (5X), inflammatory pseudopolypes (2X), ulcers (2X), enanthema (1X). Histological biopsies were obtained in 15 cases. One was considered normal. Remaining 14 biopsies exhibited following morphological changes: increased mucus secretion (8X), edema (7X), lymphocytic and plasmocytic infiltration (6X), lymphocytic and plasmocytic infiltration in addition to the presence of eosinophilic granulocytes (6X), presence of mucophages (5X), haemorrhages (4X), increased vascularity (3X), lymphocytic and plasmocytic infiltration with presence of extremely abundant eosinophilic granulocytes (1X), erosive-ulcerative changes of mucosa (1X). The changes observed indicated chronic catarrhal proctitis with expression to greater or less degree of signs of chronic catarrhal inflammation.     

Long term use of hydraulic artificial urethral sphincters in nine dogs from New Zealand with urethral sphincter mechanism incompetence

AIMS: To report on the long-term outcomes of hydraulic artificial urethral sphincter (HAUS) placement for the correction of urethral sphincter mechanism incompetence (USMI) in New Zealand dogs.

METHODS: Retrospective data were obtained from cases of dogs which had a HAUS placed after failed medical and/or surgical management of USMI between August 2012 and November 2016. Owner assessment of urinary incontinence was evaluated by an online survey in May 2017 using a visual analogue scale (0 being normal, 100 being severely affected) for the frequency, volume and severity of any straining to urinate, immediately prior to the placement of the HAUS and at the time of the survey. The number of days between surgery and the completion of survey were recorded.

RESULTS: Seven females and two male dogs, which were all desexed except for one female, were eligible for inclusion in the study. The period of follow-up following HAUS placement ranged from 206–1,685 days. Following HAUS placement, frequency and volume of urinary incontinence decreased for six dogs and were practically unchanged for three dogs. The median frequency score decreased from 70 to 13 and the volume score decreased from 73 to 12. There was no consistent change in the perceived degree of straining to urinate. Complications occurred in three dogs; one required repositioning of a dislodged injection port, one required management for haematuria and a hypoplastic bladder, and one required surgical removal of fibrous tissue around the HAUS cuff.

CONCLUSIONS AND CLINICAL RELEVENCE: HAUS placement was an effective method for the treatment of persistent USMI in most dogs and provided good clinical results based on owner assessment. The technique was associated with few complications and allowed successful long-term control of urinary incontinence without the need for medical management.

The structure of the upper atmosphere of mars: In situ accelerometer measurements from mars global surveyor

The Mars Global Surveyor (MGS) z-axis accelerometer has obtained over 200 vertical structures of thermospheric density, temperature, and pressure, ranging from 110 to 170 kilometers, compared to only three previous such vertical structures. In November 1997, a regional dust storm in the Southern Hemisphere triggered an unexpectedly large thermospheric response at mid-northern latitudes, increasing the altitude of thermospheric pressure surfaces there by as much as 8 kilometers and indicating a strong global thermospheric response to a regional dust storm. Throughout the MGS mission, thermospheric density bulges have been detected on opposite sides of the planet near 90 degreesE and 90 degreesW, in the vicinity of maximum terrain heights. This wave 2 pattern may be caused by topographically-forced planetary waves propagating up from the lower atmosphere.     

Effects of bacterial coinfection on the pathogenesis of avian pneumovirus infection in turkeys

Four- and nine-week-old poults were inoculated with cell culture propagated avian pneumovirus (APV) into each conjunctival space and nostril, followed by inoculation 3 days later with Escherichia coli, Bordetella avium (BA), or Ornithobacterium rhinotracheale or a mixture of all three (EBO). Clinical signs were evaluated on days 3, 5, 7, 9, 11, and 14 postinoculation (PI) of APV. The poults were euthanatized on days 2, 4, 6, 10, and 14 PI, and blood and tissues were collected. The poults that received APV followed by EBO or BA alone developed more severe clinical signs related to nasal discharge and swelling of intraorbital sinuses than did poults inoculated with APV alone or bacteria alone. More severe pathologic changes were found in poults inoculated with APV+BA that extended to the air sacs and lungs, particularly in 9-wk-old poults. Bordetella avium was recovered from tracheas and lungs of birds that were inoculated with APV followed by EBO or BA alone. APV was detected by immunohistochemical staining in the upper respiratory tract longer in the groups of poults inoculated with APV and pathogenic bacteria than in those that received only APV, particularly when BA was involved. Viral antigen was also detected in the lungs of poults that were inoculated with APV followed by administration of EBO or BA alone. Loss of cilia on the epithelial surface of the upper respiratory tract was associated with BA infection and may enhance infection with APV, allowing deeper penetration of the virus into the respiratory tract.     

Salmonella isolated from animals and feed production in Sweden between 1993 and 1997

This paper presents Salmonella data from animals, feedstuffs and feed mills in Sweden between 1993 and 1997. During that period, 555 isolates were recorded from animals, representing 87 serotypes. Ofthose, 30 serotypes were found in animals in Sweden for the first time. The majority of all isolates from animals were S. Typhimurium (n = 91), followed by S. Dublin (n = 82). There were 115 isolates from cattle, 21 from broilers, 56 from layers and 18 from swine. The majority of these isolates were from outbreaks, although some were isolated at the surveillance at slaughterhouses. The number of isolates from the feed industry was similar to that of the previous 5-year period. Most of those findings were from dust and scrapings from feed mills, in accordance with the HACCP programme in the feed control programme. It can be concluded that the occurrence of Salmonella in animals and in the feed production in Sweden remained favourable during 1993-97.
