Summary We developed a method for the selection of aluminium-tolerant cell lines by using a liquid medium which more closely simulates
acid soil conditions than media used previously. This medium has a pH of 4.8 and contains Al2(SO4)3 instead of Al-EDTA as the selective agent to avoid the toxic effects of EDTA. It is shown that Al2(SO4)3 exhibits a similar toxic effect on the growth of intact plants and cultured cells. With this medium, potato cell lines tolerating
2 mmol/l aluminium have been selected. 相似文献

Summary Cell cultures were developed from dihaploid clones ofSolanum tuberosum L. Selection in cell suspensions as well as plating of cells on selective medium supplemented with 5 mmol Al produced tolerant
cell lines. The constancy of Al-tolerance of cell lines was confirmed by culturing the calli for 3 months in Al-free medium
and then transferring them back to selective medium. 4 tolerant regenerant clones were obtained which maintained Al-tolerance
also after subculture in control medium. Two of the 4 clones that constantly maintained Al-tolerance, originated from cell
lines subcultured for 5 months under stress conditions. However, the regeneration rate of these cell lines was low compared
with that of lines obtained after a shorter selection period. 相似文献

To understand the genetic diversity and differentiation of Vietnamese melon (Cucumis melo L.), we collected 64 landraces from the central and southern parts of the country and assessed molecular polymorphism using simple sequence repeat and random amplified polymorphic DNA markers. The Vietnamese melon was divided into seven cultivar groups, namely “Dua le”, “Dua vang”, “Dua bo”, “Dua gang-andromonoecious”, “Dua gang-monoecious”, “Dua thom”, “Montok”, and the weedy-type melon “Dua dai”. Among these, Dua le, Dua vang, Dua bo, and Dua gang-andromonoecious are cultivated on plains and they formed cluster II along with the reference accessions of Conomon and Makuwa. Based on genetic distance, Dua le and Dua vang were regarded as Makuwa and Dua bo and Dua gang-andromonoecious as Conomon. In contrast, Dua thom and Montok are cultivated in highlands, and they formed cluster III along with landraces from the southern and eastern foot of the Himalayas. Dua gang-monoecious which is commonly cultivated in the southern parts of Vietnam, exhibited the greatest genetic diversity, as explained by its possible origin through the hybridization between Dua gang-andromonoecious and Montok. Genetic differences in melon landraces between plains and highlands and hybridization between these two geographical groups have contributed to the enhancement of genetic diversity in Vietnamese melon. 相似文献

A 90‐day feeding experiment was conducted in order to test spent brewer's yeast as a replacement for fishmeal in diets for giant freshwater prawn (Macrobrachium rosenbergii) reared in either a recirculating aquaculture system (RAS) or a biofloc system. Triplicate groups, each of 20 prawn (6.7 ± 0.03 g), were randomly assigned to 24 identical tanks. Twelve tanks were connected to the RAS, and the other 12 tanks were connected to the biofloc system which were initially supplemented with Bacillus subtilis, molasses and blood meal to achieve a C:N ratio of 20:1. Four iso‐nitrogenous (350 g/kg) and iso‐energetic (19 MJ/kg) diets where 0%, 20%, 40% and 60% of fishmeal protein was replaced by spent brewer's yeast. Neither growth nor survival was affected (p > 0.05) by increasing levels of brewer's yeast. The exception, being fish given 60% replacement with brewer's yeast, reared in biofloc, which displayed a higher (p < 0.05) growth than fish given 40% replacement, reared in clear water. The feed conversion ratio of prawn reared in the biofloc environment was lower than in the clear water. We concluded that spent brewer's yeast represents a possible 60% substitute for fishmeal in giant freshwater prawn diets, especially for prawn reared in a biofloc environment. 相似文献
