OBJECTIVE: To determine the efficacy of trilostane, a 3beta-hydroxysteroid dehydrogenase inhibitor, in dogs with pituitary-dependent hyperadrenocorticism (PDH). ANIMALS: 11 dogs with PDH. PROCEDURE: The initial dose of trilostane was 30 mg, PO, q 24 h for dogs that weighed < 5 kg and 60 mg, PO, q 24 h for dogs that weighed > or = 5 kg. A CBC count, serum biochemical analyses, urinalysis, ACTH stimulation test, and ultrasonographic evaluation of the adrenal glands were performed in each dog 1, 3 to 4, 6 to 7, 12 to 16, and 24 to 28 weeks after initiation of treatment. RESULTS: All dogs responded well to treatment. All had reductions in polyuria-polydipsia and panting and an increase in activity. Polyphagia decreased in 9 of 10 dogs, and 9 of 11 dogs had improvement of coat quality and skin condition. Concentration of cortisol after ACTH stimulation significantly decreased by 1 week after initiation of treatment. After treatment for 6 months, clinical signs resolved in 9 dogs. In the other 2 dogs, marked clinical improvement was reported for 1 dog, and moderate improvement was reported in the other dog. Ultrasonographically, there was a considerable change in the parenchyma and an increase in size of the adrenal glands. Adverse effects consisted of 1 dog with transient lethargy and 1 dog with anorexia. CONCLUSIONS AND CLINICAL RELEVANCE: Trilostane is an efficacious and safe medication for treatment of dogs with PDH. Additional studies in a larger group of dogs and characterization of progressive changes in adrenal glands are needed.  相似文献

Objective: To review the current understanding of dopamine and its use in the prevention and treatment of acute renal failure (ARF). Data sources: Original research articles and scientific reviews. Human data synthesis: Low‐dose dopamine administration has been shown to increase natriuresis and urinary output in both healthy individuals and in a few small studies in human patients with renal insufficiency. However, in several large meta‐analyses, dopamine treatment did not change mortality or the need for dialysis. Due to the potential side effects, the use of dopamine for prevention and treatment of ARF is no longer recommended in human medicine. Veterinary data synthesis: Low‐dose dopamine increases urinary output in healthy animals and animal models of ARF if given before the insult. There are no available studies looking at the effect of low‐dose dopamine therapy in naturally occurring ARF in dogs or cats. Conclusion: Due to the potential side effects of low‐dose dopamine therapy, the results from large human trials, and the lack of information in veterinary medicine, the use of dopamine for treatment of ARF in veterinary patients should be further evaluated.  相似文献

An antigen found only in neuronal nuclei of Drosophila melanogaster is revealed by staining with a monoclonal antibody (Mab44C11). This antigen appears early in development, before neurons show any other signs of antigenic or morphologic differentiation, and persists throughout development. This nuclear staining permits reliable detection of neurons in developmental studies of wild-type and mutant flies. Protein immunoblot analyses and immune precipitation experiments show that the neuronal nuclear antigen is a 50-kilodalton polypeptide.  相似文献

The T-cell receptor beta-chain gene has a nuclease hypersensitive site in several kinds of T cells, which does not appear in B cells expressing immunoglobulins. Conversely, the kappa immunoglobulin gene shows a known hypersensitive site at its enhancer element in B cells, as expected, but this site is absent in T cells. As is the case with immunoglobulin genes, the T-cell receptor site lies within the gene, in the intron separating joining and constant region segments. These nuclease hypersensitive DNA configurations in the introns of active T-cell receptor and immunoglobulin genes may arise from control elements that share ancestry but have diverged to the extent that each normally acts only in lymphoid cells which use the proximal gene product.  相似文献

Seminal, seminal vesicular, urethral and preputial fluids from bulls of two different age groups were assessed for quantitative differences in immunoglobulins. Selected markers were measured in individual samples to differentiate locally derived immunoglobulins from those present as a result of trauma or secretions from other accessory glands.Immunoglobulin levels in reproductive tract fluids from older bulls (5–6 years) were higher than those of younger bulls (3–4 years) and preputial fluids contained the highest concentration of immunoglobulins of all fluids examined. Similarities existed, however, among all fluids in the relative concentrations of immunoglobulins. IgG was generally in highest concentration, though the predominant subclass varied. A marked predominance of IgG2 over IgG1 occurred in preputial fluid samples of older bulls. IgA was in second highest concentration, and levels were often equal to or greater than those in serum. IgM was in low concentration and occasionally undetectable. IgG/IgA ratios did not exceed 5 in most of the reproductive fluids, whereas serum ratios were usually over 100. Proportional contents of albumin and immunoglobulin in reproductive tract fluids by comparison with those in serum indicated that substantial quantities of IgG as well as IgA were synthesized locally or derived by selective transport. Increased numbers of plasma cells in the lamina propria of the preputial and penile mucosa of older bulls were correlated with higher immunoglobulin concentrations in preputial fluid from older bulls, suggesting that differences in local synthesis were responsible.  相似文献

Fruit pigmentation in ripening cv. Gala, Schniga’ apples was monitored under the influence of fertigation weekly during 21 July–31 August 2009, i.e. 34 days before and 7 days after the optimal harvest date in Slovenia. The soil was a heavy loam clay with high humus content (2.8 %) and high acidity (low pH 4.9). The spectral indices NDVI (Normalized Difference Vegetation Index) and NAI (Normalized Anthocyanin Index) were obtained by a PA 1101 on attached fruit on the transition between the ground and blush colour of the fruit surface. NAI increased slowly from 0.25 to 0.6 two weeks prior to optimum maturity. NDVI values fell from 0.9 to 0.2 before and during ripening, with significant changes starting ca. 3 weeks before and during the week after the optimum harvest date. Fertigation delayed fruit ripening viz harvest date by ca. 4 days. Fertigation delayed changes in the intensity of the fruit’s epidermal pigments, expressed as NDVI for 1 week and NAI for 4 weeks, respectively, but had no significant effect on fruit colour or NDVI and NAI values at the optimal harvest date. At this time, those fruits from the fertigated trees were firmer with a lower starch index compared with the un-fertigated control. Statistically, highly significant correlations were found (p?≤?0.01) (r?=?0.35 to r?=?0.91, R2?=?0.11–0.83) between all the studied spectral and standard parameters i.e. fruit fresh firmness, soluble solids content and starch index.  相似文献
