The effects of crude extracts and an isolated compound from the leaves of milkweed, Pergularia daemia (Forssk) Choiv., on the antifeedant activity against two important lepidopteran pests, Helicoverpa armigera (Hub.) and Spodoptera litura (F.), were studied. Maximum antifeedant activity was recorded in ethyl acetate crude extract against H. armigera (70.3%) and S. litura (71.82%) at 1% concentration. Ethyl acetate crude extract was further subjected to column chromatography, which was performed using hexane as initial solvent and then by increasing the polar strength using ethyl acetate. Fractions collected at hexane and ethyl acetate (80:20) yielded 6-(4,7-hydroxy-heptyl) quinone, a novel compound which showed significant antifeedant activity against H. armigera (80.22% at 2000 ppm) and S. litura (68.31% at 2000 ppm).  相似文献

Groups of turkeys were exposed to different isolates of avian influenza virus from wild mallard ducks and domestic turkeys by the intracerebral, intravenous, intratracheal, and intra-airsac routes, and pathogenicity indices were calculated. For the intracerebral pathogenicity study, body weight was also measured. For intravenous, intratracheal, and intra-airsac pathogenicity studies, necropsy lesions were scored and serological responses were recorded. Only the intracerebral pathogenicity index and body weight gain post intracerebral infection demonstrated any differences between isolates. The other procedures failed to demonstrate any pathogenicity whatsoever. There was a correlation (R = 0.73) between intracerebral pathogenicity index and reduced weight gain postinfection. These studies suggest that growth suppression may be an objective measure of pathogenic potential of influenza viruses found to be nonpathogenic by other methods.  相似文献

A method is described for determining residues of 1,2-dibromoethane (EDB) in milled wheat products. The residues are first extracted with benzene, then recovered azeotropically for reaction with alkali and subsequent determination of liberated bromide ion by a Volhard titration. Freshly milled flours from wheat fumigated with 65.0 and 325 mg EDB/litre at 37°C were found to contain 10.6 to 39.4 mg EDB/kg and the bran contained from 51.4 to 153 mg EDB/kg.  相似文献

Comparative serological evaluation of avian influenza vaccine in turkeys   总被引:1,自引:0,他引:1  
Four- and six-week-old turkeys were vaccinated subcutaneously using avian influenza virus (AIV) A/Duck/613/MN/79 (H4N2) killed oil-emulsion vaccine. Sequential serological tests using agar gel precipitin (AGP), hemagglutination inhibition (HI), and enzyme-linked immunosorbent assay (ELISA) for measuring antibodies to AIV were performed up to 4 weeks postvaccination, when birds were challenged intranasally using A/Turkey/MN/80 (H4N2) live AIV. The ELISA was 25 to 1600 times more sensitive than the HI test and was able to detect antibody production earlier than the HI test. All turkeys with an ELISA titer of greater than or equal to 800 were protected against homologous challenge, as measured by virus recovery 3 days postchallenge. Four turkeys out of 20 serologically negative by AGP and HI tests but ELISA-positive were protected.  相似文献

The ethanol and methanol extracts of Cassia auriculata flowers were screened for antioxidant activity. The antioxidant activity was determined by an improved assay based on the decolorization of the radical monocation of 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging method. The ethanol and methanol extracts of C. auriculata flowers showed antioxidant activity in both assays.  相似文献

Verticillium dahliae is a soilborne fungus that causes Verticillium wilt on multiple crops in central coastal California. Although spinach crops grown in this region for fresh and processing commercial production do not display Verticillium wilt symptoms, spinach seeds produced in the United States or Europe are commonly infected with V. dahliae. Planting of the infected seed increases the soil inoculum density and may introduce exotic strains that contribute to Verticillium wilt epidemics on lettuce and other crops grown in rotation with spinach. A sensitive, rapid, and reliable method for quantification of V. dahliae in spinach seed may help identify highly infected lots, curtail their planting, and minimize the spread of exotic strains via spinach seed. In this study, a quantitative real-time polymerase chain reaction (qPCR) assay was optimized and employed for detection and quantification of V. dahliae in spinach germplasm and 15 commercial spinach seed lots. The assay used a previously reported V. dahliae-specific primer pair (VertBt-F and VertBt-R) and an analytical mill for grinding tough spinach seed for DNA extraction. The assay enabled reliable quantification of V. dahliae in spinach seed, with a sensitivity limit of ≈1 infected seed per 100 (1.3% infection in a seed lot). The quantification was highly reproducible between replicate samples of a seed lot and in different real-time PCR instruments. When tested on commercial seed lots, a pathogen DNA content corresponding to a quantification cycle value of ≥31 corresponded with a percent seed infection of ≤1.3%. The assay is useful in qualitatively assessing seed lots for V. dahliae infection levels, and the results of the assay can be helpful to guide decisions on whether to apply seed treatments.  相似文献

Insect infestations in stored wheat affect the chemical characteristics and baking qualities of wheat flour, and insect‐infested flours are unacceptable in the baking industry. The efficiency of the soft X‐ray method to detect infestations caused by Cryptolestes ferrugineus (Stephens), Tribolium castaneum (Herbst), Plodia interpunctella (Hübner), Sitophilus oryzae (L.), and Rhyzopertha dominica (F.) in wheat kernels was determined in this study. Wheat kernels infested by different insects were prepared by artificial implantation of insect eggs or by introducing adult insects in wheat samples. Kernels infested by different stages of the insects were X‐rayed until the adults emerged from the kernels. A total of 57 features using histogram groups, histogram and shape moments, and textural features were extracted from the X‐ray images and a linear‐function parametric classifier was used to identify the insect‐infested kernels. The parametric classifier identified more than 84% of infestations due to C. ferrugineus and T. castaneum larvae. The infestations by C. ferrugineus pupae‐adults and P. interpunctella larvae were identified with >96% accuracy. Kernels infested by different stages of S. oryzae and R. dominica larvae were identified with >98% accuracy. Using the Berlese funnel method, 67, 51, and 81% of first, second, and third instars of C. ferrugineus, respectively, were extracted in 6 hr. The same infested kernels were all categorized as infested by the parametric classifier. When kernels infested by different insects were pooled together, the parametric classifier correctly identified 74% of uninfested and 94% of infested kernels by the internal and external grain feeders. The 26% false positives identified from the independent test was caused by one sample infested by T. castaneum. When that sample was removed from the training set, the false positives were reduced to 16%, and 92.7% of infested kernels by different insects were correctly identified.  相似文献

The objective of the present study was to establish the changes in plasma concentrations of LH, FSH, estradiol 17-beta (E2) and progesterone (P4), as well as to understand their temporal relationships during oestrus in mithun (Bos frontalis). The experiment was conducted on 11 mithuns during third or fourth postpartum oestrous cycle. Since oestrus onset the jugular vein blood samples were collected every 2 h for 72 and 96 h, respectively from the animals without and with standing heat. The LH, FSH, E2 and P4 concentrations were estimated in plasma. The P4 concentration was fluctuated throughout the oestrus period and the average P4 concentration was found significantly (p<0.05) lower on the day of oestrus onset. The multiple rises in LH and FSH concentrations above the basal level in spike like fashion were observed throughout the oestrus period irrespective of the occurrence of standing heat. A significant (p<0.01) gradual increase in the average daily E2 concentration was observed till day 2 following oestrus onset irrespective of the occurrence of standing heat. A significant (p<0.05) simultaneous increase in LH, FSH and E2 concentrations and a transient increase in P4 concentration at approximately the time of standing heat onset were observed. During investigation a definite temporal coupling between LH and FSH rises was absent throughout the oestrus period. The results suggest that (1) the multiple short-duration low-amplitude LH and FSH surges during oestrus may be crucial for the final maturation of ovulatory follicle and subsequent ovulation in mithun; (2) a differential mechanism for controlling LH and FSH secretions probably exists in mithun.  相似文献
