Seasonal pattern of shoot emergence and its endogenous control in horsenettle (Solanum carolinense L.)

Controlling established horsenettle plants is achieved by suppressing shoot emergence from root systems. The seasonal pattern of shoot emergence and its possible endogenous control in horsenettle ( Solanum carolinense L.) were investigated. The shoot emergence period in an undisturbed population was limited to a seven-week period from mid-April, and a little longer in tilled conditions. Detached roots showed very high shoot-sprouting ability under 15–30°C throughout the year. In shoot clipping experiments, new shoots sprouted only from the stem and not from the root when attached to shoots, whether above-ground or underground. On the contrary, new shoots sprouted from the roots when all parts of the shoots were clipped off. From these results, the limited shoot emergence period in horsenettle is thought to be initiated by temperatures necessary for sprouting and is ended by a growth correlation effect between early emerged and matured shoots.     

8S globulin of mungbean [Vigna radiata (L.) Wilczek]: cloning and characterization of its cDNA isoforms, expression in Escherichia coli, purification, and crystallization of the major recombinant 8S isoform

Three isoforms of the cDNA of the major 8S globulin of mungbean, 8Salpha, 8Salpha', and 8Sbeta, were isolated, cloned, and characterized. The cDNA sequences of 8Salpha, 8Salpha', and 8Sbeta had open reading frames of 1362, 1359 or 1362, and 1359 bp, respectively, which code for 454, 453 or 454, and 453 amino acids corresponding to molecular weights of 51 973, 51 627 or 51 758, and 51 779, respectively. Homology in terms of cDNA and amino acid sequences was 91-92% between 8Salpha and 8Salpha', 87% between 8Salpha and 8Sbeta, and 86-88% between 8Salpha' and 8Sbeta. The signal peptide was found to be 1-25, 1-24 or 25, and 1-23 for 8Salpha, 8Salpha', and 8Sbeta, respectively, using the signalP website (Nielsen, H.; Engelbrecht, J.; Brunak, S.; von Heijne, G. Protein Eng. 1997, 10, 1-6). The propeptide was determined to be IVHREN. A single site for glycosylation (N-X-S/T) was observed about 90 amino acids from the C terminus. Homology between mungbean 8S isoforms and other 7-8S proteins ranged from 45 to 68% within members of the legume family and 29 to 34% for crops of different species. The major isoform 8Salpha was expressed in Escherichia coli and purified by successive ammonium sulfate fractionation, hydrophobic interaction, and Mono Q column chromatography. The recombinant 8Salpha, but not the native form, was successfully crystallized producing rhombohedral crystals.     

Effects of protein engineering of canola procruciferin on its physicochemical and functional properties

The primary structure of Brassica napus procruciferin 2/3a was engineered to elucidate structure-function relationships and to improve the functionality of cruciferin. The following mutants were constructed: (1) C287T, (2) DeltaII, variable region II was deleted; (3) C287T/DeltaII, mutation involving (1) and (2); (4) DeltaIV + A1aIV; and (5) DeltaIV + A3IV, variable region IV was replaced with variable region IV containing many charged residues from soybean glycinin A1aB1b and A3B4 subunits. Differential scanning calorimetry analysis revealed that the A1aIV region has a more favorable interaction with the procruciferin molecule than does A3IV as well as the original regions. On the basis of heat-induced precipitation analysis, it was concluded that replacement of the free cysteine residue with threonine (C287T) and insertion of charged regions (DeltaIV + A1aIV and DeltaIV + A3IV) could lead procruciferin to form soluble aggregates after heating. Low solubility was observed in mutants DeltaIV + A3IV, DeltaII, and C287T/DeltaII, especially between pH 4 and 6 at mu = 0.08, but not in DeltaIV + A1aIV, indicating that the number of acidic amino acid residues and the high number of glutamine residues are important factors for solubility at mu = 0.08. None of the mutants showed any improvements in emulsifying ability, indicating that destabilization and addition of the hydrophilic region are not effective for emulsification. The insertion of the A1aIV region in procruciferin made the molecule more susceptible to alpha-chymotrypsin.     

A statistical approach for predicting accuracies of soil properties measured by single, double and dual gamma beams

A statistical analysis is developed to describe the accuracies of soil properties measured through single, double and dual gamma-beam methods. The gamma sources are ¹³⁷Cs for the high-energy beam (662 keV) and ²⁴¹Am for the low-energy beam (60 keV). The theoretical confidence intervals of the thicknesses or water contents of samples are determined using a modification of the method of Gardner el al. (1972), who derived theoretical variances using standard error propagation formulae. The finding of Gardner et al. that, in the double-beam method, errors in the soil bulk density and water content measurements are primarily caused by random emission from the sources, is confirmed by the theoretical confidence intervals and is applied to the single-beam and dual-beam methods. The effects of interferences between gamma rays in the dual-beam method on the accuracies of estimations are examined. The minimum measurement time to attain a required accuracy can be predicted from the theoretical confidence intervals.     

Chromosomal regions controlling anther culturability in rice (Oryza sativa L.)

Diallel analysis has revealed that anther culturability in rice (Oryza sativa L.) is a quantitative trait controlled by the nuclear genome. Mapping of anther culturability is important to increase the efficiency for green plant regeneration from microspores. In the previous study, we detected distorted segregation of RFLP markers in rice populations derived from the anther culture of an F₁ hybrid between a japonica cultivar ‘Nipponbare’ and an indica cultivar ‘Milyang 23’. To clarify the association between chromosomal regions showing distorted segregation and anther culturability, the anther culturability of doubled haploid lines derived from the same cross combination was examined, and the association between alleles of the RFLP markers, which exhibiting the most distorted segregation on chromosomes 1, 3, 7, 10 and 11, and the anther culturability was evaluated. One region on chromosome 1 was found to control callus formation from microspores, and one region on chromosome 10 appeared to control the ratio of green to albino regenerated plants. In both regions, the Nipponbare allele had positive effects. Three regions on chromosomes 3, 7 and 11, however, showed no significant effect on anther culturability. An outbreak of rabbit hemorrhagic disease (RHD) caused by Lagovirus europaeus GI.2/rabbit hemorrhagic disease virus 2 (RHDV2) in Ehime, Japan

A total of ten 1–2-year-old rabbits died within 2 weeks at a facility in Ehime prefecture in May 2019. Necropsy revealed liver discoloration and fragility, hemorrhage of some organs and blood coagulation failure. On histopathologic examination, necrotizing hepatitis was a common finding, together with fibrin thrombi in the small vessels and hemorrhage in some organs. Rabbit hemorrhagic disease (RHD) virus gene was detected in liver samples, and viral particles of approximately 32 nm in diameter were found in the cytoplasm of degenerated hepatocytes by electron microscopy. Phylogenetic analysis based on the partial VP60 gene sequence classified it as Lagovirus europaeus GI.2/RHDV2. This is the first confirmed outbreak of RHD caused by globally emerging GI.2/RHDV2 in Japan.

The evaluation of piromidic acid as an antibiotic in fish: an in vitro and in vivo study

Abstract. The activity of piromidic acid (PA, 8-ethyl-5, 8-dihydro-5-oxo-2-prior lidinopyrido [2,3–d] pyrimidine-6-carboxylic acid) against bacteria in fish was evaluated in vitro and in vivo .
In vitro , PA was effective against a wide range of pathogenic bacteria stocked in this laboratory and freshly isolated from fish. PA was active against organisms showing multiple drug-resistance and showed no cross-resistance to chloramphenicol, tetracycline and sulfamonomethoxine.
In vivo , orally administered PA was effective against experimental infections with Aeromonas hydrophila and Vibrio spp. in goldfish and eels. The efficacy of PA against these infections was equal to or higher than that of chloramphenicol, and higher than that of tetracycline. Distribution of PA in the Ash was investigated by bioassay and autoradiography. In the bioassay, orally administered PA was easily absorbed and distributed in the blood and tissues of major organs within 1 h after the administration. Peak levels were attained 2 to 4 h post dosing, and no residue was detected in the blood and tissues 48 h post dosing. The results obtained from the autoradiographic investigation in goldfish were similar to those from the bioassay. Therapeutic levels of PA were well tolerated by fish when administered with feed.     

The evaluation of the curd forming ability of milk replacers

Inadequate milk curd formation in the abomasum of newborn calves causes malnutrition and diarrhea. In order to define the factors of inadequate curd formation, we compared the curd forming ability among 9 kinds of milk replacers, bulk milk (raw milk), and skim milk both in vitro and in vivo . When rennet was added, the raw milk and one milk replacer formed firm curds. The rest of the milk replacers and skim milk did not form any curd. When a solution of HCl was added, raw milk, three milk replacers and skim milk formed the curd at pH 4.5, but the other milk replacers did not. When HCl was added following the rennet, raw milk, one milk replacer and skim milk formed the curd. In vivo , raw milk, two milk replacers and skim milk showed good curd formation whereas the other milk replacers showed poor curd formation inside the abomasums of the calves. This study showed that most of the milk replacers sold in Japan could not form the curd with rennet.
