The effects of lipid-rich bovine serum albumin (LR-BSA) on the development of porcine blastocysts produced
in vitro were examined. Addition of 0.5 to 5 mg/ml LR-BSA to porcine blastocyst medium
(PBM) from Day 5 (Day 0 = in vitro fertilization) significantly increased the hatching rates
of blastocysts on Day 7 and the total cell numbers in Day-7 blastocysts. When Day-5 blastocysts were cultured
with PBM alone, PBM containing LR-BSA, recombinant human serum albumin or fatty acid-free BSA, addition of
LR-BSA significantly enhanced hatching rates and the cell number in blastocysts that survived compared with
other treatments. The diameter, ATP content and numbers of both inner cell mass and total cells in Day-6 and
Day-7 blastocysts cultured with PBM containing LR-BSA were significantly higher than in blastocysts cultured
with PBM alone, whereas LR-BSA had no effect on mitochondrial membrane potential. The mRNA levels of enzymes
involved in fatty acid metabolism and β-oxidation (ACSL1, ACSL3,
CPT1, CPT2 and KAT) in Day-7 blastocysts were
significantly upregulated by the addition of LR-BSA. The results indicated that LR-BSA enhanced hatching
ability and quality of porcine blastocysts produced in vitro, as determined by ATP content,
blastocyst diameter and expression levels of the specific genes, suggesting that the stimulatory effects of
LR-BSA arise from lipids bound to albumin. 相似文献

Fipronil (FPN) is a systemic insecticide that antagonizes the gamma-aminobutyric acid type A (GABAA) receptors in insects. Recently, adverse effects of FPN on mammals have been reported, but most of those were caused by high doses of FPN and additives in the products. We investigated the effects of low-dose pure FPN on the emotional behavior of mice. Nine-week-old male mice conducted behavioral tests 24 hr after FPN administration by gavage at doses of 0.05 or 5 mg/kg based on the no-observed-effect level (NOEL), showed a significant increase in locomotor activity and dose-dependent responses on the time they spent in the central zone in the open field test. Pure FPN below the NOEL dose may affect the emotional behavior of mice. 相似文献

Fog water and precipitation were collected and analyzed to study fog and precipitation chemistry. The research was carried out through one year from April 1997 to March 1998 at Mt. Rokko in Kobe. Higher fog occurrence and larger volume of fog water were observed in summer, corresponding to the trend of seasonal variation in precipitation amount. The annual mean pH value of fog water (3.80) was lower by ca. one pH unit than that of precipitation (4.74). The concentration of chemical species in fog water was ca. 7 times that in precipitation. The highest anion and cation concentrations were SO4 2? and NH4 + in fog water and Cl? and Na+ in precipitation, although the Cl?/Na+ equivalent ratio in both fog water and precipitation was almost the same value as that in sea water. It is considered that in the longest fog event, NH4 + and nss-SO4 2? in fog water mainly scavenged as (NH4)2SO4, mainly derived from (NH4)2SO4 (aerosol) in the atmosphere, NH3 was scavenged at the growing stage, and SO2 was also scavenged after the mature stage. Recently, developmental exposure to clothianidin (CLO) has been shown to cause reproductive toxicity in male mice, but the effects in female mice remain to be clarified. Pregnant C57BL/6N mice were given a no-observed-adverse-effect-level (NOAEL) dose of CLO until weaning. We then examined ovaries of 3- or 10-week-old female offspring. In the CLO-administered group, morphological changes, a decrease in the immunoreactivity of the antioxidant enzyme glutathione peroxidase 4 (GPx4), and activation of genes in the steroid hormone biosynthesis pathway were observed in 3-week-old mice, and decreases of GPx4 immunoreactivity, 17OH-progesterone and corticosterone levels were observed in 10-week-old mice, along with high rates of infanticide and severe neglect, providing new evidence that developmental exposure to CLO affects juvenile and adult mice differently. 相似文献
