We examined whether recreational tree climbing (TC) activities would have positive social impacts and rejuvenate an outdoor activity center and surrounding community forest in central Japan. Our case study comprised 3800 adults and children participating in TC activities at the Jyokoji Outdoor Activity Center (JOAC) in Seto, Japan. We focused on 1393 adult participants whom we considered to be the decision makers regarding family recreation, and we explored both the social impacts of TC and various factors that contributed to and distracted from the overall experience. Qualitative and quantitative data indicated that elements such as age, gender, tree preferences, tree appreciation, tree education, and technical instruction influenced the enjoyment of recreational TC and the positive effects on tree climbers. Overall, the climbing program had positive social impacts for the community forest and contributed to local conservation initiatives. Our case study suggests that other areas can benefit from incorporating TC programs into community forest planning to complement aesthetic, ecological, and restoration benefits and provide a new venue for recreation and conservation awareness. 相似文献

The Spontaneously Diabetic Torii (SDT) fatty rat, a model for obese type 2 diabetes,
shows bone quantitative abnormalities, namely low bone mineral density (BMD). The
objective of this study was to evaluate bone morphological changes, in particular
identifying the bone qualitative abnormalities, in the SDT fatty rat. Male SDT fatty rats
showed increases in total trabecular area and trabecular number and decreases in
trabecular thickness in cancellous bones of the proximal tibia, indicating trabecular
miniaturization. The SDT fatty rat is useful for investigation of pathophysiological
changes in bone quality in diabetic osteoporosis. 相似文献

The Japanese crested ibis Nipponia nippon is a critically threatened bird. Accurate sexing is necessary to perform effective management of captive breeding toward a national project for a tentative release of the Japanese crested ibis on Sado Island. A PCR‐based sexing method targeting a 0.6 kb EcoRI fragment (EE0.6) sequence on W chromosome with AWS03 and USP3 primers has been developed for the Japanese crested ibis. However, the primers were selected from the EE0.6 sequences from bird species other than the Japanese crested ibis. In this study, we determined the W‐ and Z‐linked EE0.6 sequences in the Japanese crested ibis, and clarified Japanese crested ibis sequence mismatch in the binding sites of the primers. Further, we found no polymorphism in the primer binding sites among five founder birds for the Sado captive Japanese crested ibis population. These findings validated the PCR‐based sexing method with the AWS03 and USP3 as accurate molecular sexing methods of captive Japanese crested ibis on the Sado Island. Additionally, we designed a primer set for a novel PCR‐based sexing, based on the EE0.6 sequences obtained in this study. This novel sexing method may be useful for future ecological research following the release of Japanese crested ibis on Sado Island. Feline gastrointestinal eosinophilic sclerosing fibroplasia was diagnosed in an 8-month-old Scottish fold that had a primary gastrointestinal mass involving the stomach, duodenum and mesenteric lymph nodes. Histopathologically, the most characteristic feature of this mass was granulation tissue with eosinophil infiltration and hyperplasia of sclerosing collagen fiber. Immunohistochemically, large spindle-shaped cells were positive for smooth muscle actin and vimentin. This case emphasizes the importance of feline gastrointestinal eosinophilic sclerosing fibroplasia as a differential diagnosis of gastrointestinal neoplastic lesions such as osteosarcoma and mast cell tumor in cats. 相似文献

Previously, we developed a simple and efficient method to isolate liver macrophages from a mixed primary culture of adult rat liver cells. To extend the applicability of this method, we isolated macrophages from mixed primary cultures of bovine liver cells. Macrophage cells proliferated on the cell sheet of mixed bovine liver cells after 8-16d of culture. These cells were detached by shaking of the culture flasks. Subsequent transfer and brief incubation in plastic dishes resulted in selective adhesion of macrophages. After rinses with PBS, attached macrophages were harvested. More than 10(6) cells could be harvested from the culture flask at intervals of 2-3d for more than three weeks. The isolated cells were strongly positive for bovine macrophage markers, such as CD68, CD172a and Iba-1. These cells exhibited functional properties of macrophages, including active phagocytosis of polystyrene microbeads, proliferative response to recombinant bovine granulocyte-macrophage colony-stimulating factor, upregulation of specific inflammatory cytokine genes upon stimulation with lipopolysaccharide, and formation of multinucleated giant cells. The shaking and attachment method provides a simple and efficient alternative to obtain bovine liver macrophages without requiring complex equipment or specialized technical skills. 相似文献

Effect of methotrexate exposure at late gestation on development of telencephalon in rat fetal
brain

Pregnant rats were treated with 30 mg/kg of methotrexate (MTX) on gestation day (GD) 16, and fetal brains
were examined time-dependently. On GD 20, the appearance of the telencephalon in the MTX group was different
from that in the control group, and the major axis of the telencephalon of the MTX group was shortened,
compared to that of the control group. In the sagittal section of the telencephalon in the MTX group on GD 20,
histopathological findings of deformation and narrowing of the cerebral ventricle, the disturbance of the
arrangement of the marginal cell layer of subventricular zone (SVZ) and thickening of telencephalic wall,
cortical plate and ventricular zone (VZ)/SVZ were possibly attributable to neuronal migration disorders by
MTX. Through all the experimental period, few pyknotic cells or TUNEL-positive cells were observed in the
VZ/SVZ of the telencephalic wall and striatum in the control group. On the other hand, in the VZ/SVZ of the
telencephalic wall and striatum in the MTX group, pyknotic cells or TUNEL-positive cells were observed on GD
17, and they increased significantly on GD18 and then decreased to the control levels from GD 19 onward. The
phospho-Histone H3-positive rate decreased remarkedly in the VZ/SVZ of the telencephalic wall and striatum of
the MTX group on GDs 17 and 18, compared to the control group, but they recovered on and after GD 19. These
results suggested that there was a high possibility that development of the telencephalon in this period

Genetically modified corn Bt11 is insect-resistant and expresses Cry1Ab toxin, an insecticidal protein, in kernels. Although Bt11 corn is considered safe based on animal performance, there are no reports available on the clinico-biochemical effects of feeding it to cattle. In this study, we evaluated the effects of feeding Bt11 to calves, using blood and ruminal clinico-biochemical parameters. Our three-month-long feeding experiment demonstrated that calves (n=6), fed with a ration containing 43.3% of Bt11 corn kernels as dry matter, did not develop any discernible clinical, hematological, biochemical, or ruminal abnormalities as compared with control calves (n=6) fed non-Bt11 corn. The results suggest that the transgenic Bt11 has no negative clinico-biochemical effects on calves. 相似文献

Floral scent attracts pollinators. We investigated the floral scent compounds recognized by pollinators in six Brassica crop species, including allogamous species with different genomes and autogamous species with two parental genomes and radish (Raphanus sativus). Biologically active compounds recognized by honeybees were screened from all floral compounds by combined gas chromatography–electroantennogram analysis and their profiles were determined by gas chromatography–mass spectrometry. Fourteen of the 52 compounds were active. All accessions had more than two active compounds, but the compounds greatly differed between the two genera. On the basis of similarities in whether active compounds were presence or absence, their amount and their composition ratio, we divided the Brassica accessions into three to five groups by cluster analyses. Most groups were composed of a mixture of allogamous and autogamous species sharing same genome, indicating that the variation depended on genome, not species. These results suggest that all species require pollinator visits for reproduction, despite their different reproductive systems. However, the inter-genus and intra-specific variations shown by the multiple groups within a species might cause different visitation frequencies by pollinators between genera and among accessions within a species, resulting in insufficient seed production in some accessions or species. 相似文献
