Mesoamerica provides a unique context for biodiversity conservation in managed landscapes because of its geography, history of human intervention, and present conservation and development initiatives. The long and narrow form of the Mesoamerican landmass, and its division by a central mountain range, has served as both a bridge and a barrier. Conservation efforts in Mesoamerica are unique for the emphasis they place on regional connectivity through the Mesoamerican Biological Corridor and on biodiversity conservation in managed landscapes. The emphasis on conservation in agricultural systems has fostered innovations in payment for ecosystem services, and provides novel insights on the functional role that biodiversity plays in the provisioning of ecosystem services. The increasing rate of economic development in the region and the advent of new payment for ecosystem service schemes have provided new opportunities for forest regeneration and restoration. However, the small scale of private landholdings and the diversity of land uses featured in the region, while contributing to biodiversity conservation due to their structural and floristic complexity, present challenges for biodiversity monitoring and management.  相似文献

In this study, we combined the analysis of a coenocline with Huisman–Olff–Fresco (HOF) modelling of species behaviour to describe colonization patterns of woody species on reclaimed coal wastes in northern Spain. We hypothesize that fine‐scale variation in abiotic factors along the forest‐grassland gradient that appears after mining restoration affects colonization pattern of woody species. To understand these factors, we analysed changes in floristic composition and abundance of the main woody colonizers according to the spatial distance from forest limit, as well as the spatial variability of some edaphic and microclimatic parameters. Our results showed how the primary coenocline extracted from DCA1 reflected a gradient in environmental conditions influencing the species composition of the woody plant communities, from the more shade, moist and dense forest on acid and rich soils to the open, dry and oligotrophic grassland. The colonization pattern of woody species is affected by fine‐scale variations in abiotic factors, such as the increase in pH, P, light, and the decrease in K, N, C/N, organic matter and soil moisture, from the forest to the restored mine. Most species HOF modelled showed monotone responses with decreasing trends along the environmental gradient from forest to mine, being Quercus petraea the species clearly dominant in the forest. On the contrary, Cytisus scoparius and Genista florida colonize intensely in the mine area, reaching the maximum abundance around 15 and 10 m distance from the forest limit, respectively. We conclude that the composition of woody species in the forest is not a good predictor of the colonizing intensity, not even of the species composition in the mine area. A liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed for the quantification of the four major enterolignan precursors [secoisolariciresinol, matairesinol, lariciresinol, and pinoresinol] in foods. The method consists of alkaline methanolic extraction, followed by enzymatic hydrolysis using Helix pomatia (H. pomatia) beta-glucuronidase/sulfatase. H. pomatia was selected from several enzymes based on its ability to hydrolyze isolated lignan glucosides. After ether extraction samples were analyzed and quantified against secoisolariciresinol-d8 and matairesinol-d6. The method was optimized using model products: broccoli, bread, flaxseed, and tea. The yield of methanolic extraction increased up to 81%, when it was combined with alkaline hydrolysis. Detection limits were 4-10 microg/(100 g dry weight) for solid foods and 0.2-0.4 microg/(100 mL) for beverages. Within- and between-run coefficients of variation were 6-21 and 6-33%, respectively. Recovery of lignans added to model products was satisfactory (73-123%), except for matairesinol added to bread (51-55%).  相似文献

We detected and cultivated isolates of Trypanosoma (Megatrypanum) theileri from cattle and water buffaloes in S?o Paulo state, southeastern Brazil, which were characterized by comparing morphological, growth and molecular features. Although isolates from cattle and water buffalo were morphologically indistinguishable, they differed in their growth characteristics. A dendrogram based on randomly amplified polymorphic DNA (RAPD) patterns indicated close-genetic relationships among all isolates from both species, which were all tightly clustered together and distant from Megatrypanum species from wild mammals. In addition, isolates within the T. theileri-cluster were clearly segregated into two host-associated groups. The sequence of a synapomorphic RAPD-derived DNA fragment (Tth625), which was shared by all T. theileri trypanosomes from cattle and buffalo but not detected in any of 13 other trypanosome species, was used as target for a conventional T. theileri-specific PCR assay. We also defined RAPD fragments (Tthc606 and Tthb606) that distinguished cattle from buffalo isolates. Thus, distinct growth features and genetic variability distinguished between isolates from cattle and water buffaloes of the same geographic origin, suggesting an association of these isolates with their host species. The trypanosomes from water buffalo reported here are the first T. theileri-like isolates from the Asiatic buffalo (Bubalus bubalis) to be continuously cultured and compared with cattle isolates using biological and molecular methods.  相似文献

Around the world, agricultural landscapes are increasingly seen as “multi-functional” spaces, expected to deliver food supplies while improving rural livelihoods and protecting and restoring healthy ecosystems. To support this array of functions and benefits, governments and civil society in many regions are now promoting integrated farm- and landscape-scale management strategies, in lieu of fragmented management strategies. While rural producers are fundamental to achieving multi-functional landscapes, they are frequently viewed as targets of, or barriers to, landscape-oriented initiatives, rather than as leading agents of change. In reality, however, rural producers in many areas have embraced elements of multi-functional land management. In this paper, we explore the role and recent evolution of producer movements in influencing multi-functional farm and landscape management. We explore these roles through six case studies, including a land reform movement in Brazil, indigenous territorial development in Bolivia, conservation agriculture associations in Canada, environmental cooperatives in the Netherlands, indigenous and biocultural heritage associations in Peru, and Landcare groups in the Philippines. These experiences suggest that producer movements are playing pivotal roles in supporting landscape multi-functionality, not only through agroecological farming practices but also through off-farm efforts to conserve ecosystems and support multi-stakeholder landscape planning. On the other hand, interests of producer movements are not always fully aligned with multi-functional landscape management approaches. The contribution of producer movements to multi-functional landscapes depends on these movements including farm and landscape stewardship in their values and goals, and having the political support and capacity to engage meaningfully in multi-stakeholder processes.  相似文献

The objective of this study was to determine the number, morphology and ultrastructure of preantral ovarian follicles of buffalo (Bubalus bubalis) foetuses at different ages. Quantification revealed number of primordial, primary and secondary follicles of 48 857 ± 17 506, 26 000 ± 20 452, 18 428 ± 10 875 and 18 375 ± 19 690, 225 ± 349, 326 ± 288 at 12–34 cm and 35–60 cm crown rump length (CRL), respectively. Follicular diameter values were 28.9 (±3.4), 34.7 (±5.9) and 59.4 (±12.6) μm; oocyte diameters were 21.7 (±2.8), 24.3 (±3.4) and 33.0 (±7.7) μm, and the numbers of follicular cells in the follicle equatorial section were 7.1 (±1.4), 12.0 (±2.4) and 13.8 (±2.4) for primordial, primary and secondary follicles, respectively. The primordial follicle consisted of an oocyte surrounded by a layer of flattened follicular cells with a normally eccentric oocyte nucleus. Dispersed Golgi complex, smooth endoplasmic reticulum, rounded mitochondria and several lipid vesicles were observed in the cytoplasm and cell junctions between the follicle cell membranes and the oocyte. This work describes the number, morphometry and ultrastructure of preantral follicles of buffalo foetuses, concluding that folliculogenesis is established between 8 and 34 cm CRL and that follicle number varies individually and according to age and that further studies are needed in this species.  相似文献

The objective of this experiment was to assess the features and extent of follicular apoptosis in the water buffalo (Bubalus bubalis) ovary using classical histology and nick end labelling technique. Ovaries (n = 40) procured from the slaughterhouse were used for the study. The sections (5 μm) were used for detection of terminal deoxynucleotidyl transferase‐mediated dUTP‐biotin nick end labelling (TUNEL) and classical histology (H&E). Those follicles showing ≥ 5% TUNEL positivity (TUNEL assay) and pyknotic nuclei (histology) in granulosa cells were classified as atretic. Based on histology, the atretic primary and secondary follicles (%) were 93.82 and 95.62 respectively. The histology study reveals that the rates (%) of atresia in <1, 1–3, 3–5 mm and >5 mm were 36.90, 40.50, 62.84 and 74.5 respectively. Further the atretic tertiary follicles (%) were significantly lower than the primary and secondary classes of follicles. TUNEL assay reveals that the atretic rate (%) of tertiary follicles in <1, 1–3, 3–5 and ≥ 5 mm class follicles were 50.88, 53.84, 81.81 and 36.36 respectively. The percentage of atresia in >5 mm diameter follicles were significantly lower in TUNEL than histology. Percentages of granulosa and thecal cells positive for atresia by TUNEL were 30.7 ± 0.53 and 13.82 ± 0.18 respectively per follicle. The initial structural changes in atretic follicles were seen primarily in the granulosa cells. In severely atretic follicles TUNEL positive granulosa cells along with theca cells have to be considered in assessing the rate and extent of atresia.  相似文献
