Tryptophan (Trp) catabolism appears to be an important mechanism for regulation of inflammatory responses, resulting in T-cell tolerance and survival of semi-allogeneic concepti during pregnancy. Trp catabolism can be induced by IFN-gamma, and is therefore an important host defence mechanism against intracellular pathogens. Chlamydophila abortus is a bacterial pathogen that can cause persistent infection in non-pregnant sheep, but invades the placenta and causes abortion in late pregnancy. IFN-gamma was found to control the growth of Chlamydophila abortus in ovine cells in a highly dose-dependent manner. Addition of 200U/ml IFN-gamma eradicated all traces of infection from the cultures, whereas concentrations less than 50U/ml failed to control the growth of the organism, resulting in cell lysis. However, concentrations in the range of 50-100U/ml were found to restrict growth to an extent that a persistent infection was established, allowing survival of the organism in tissue culture for several months. Removal of IFN-gamma resulted in the re-appearance of infectious organisms. Addition of exogenous Trp to the cells treated with 50-100U/ml IFN-gamma prevented the establishment of persistence. These effects in tissue culture are analogous to the persistent infection observed in pregnant sheep prior to abortion. These data suggest that control of C. abortus growth in the periphery is linked to the balance of pro-inflammatory cytokine production and availability of Trp during pregnancy.  相似文献

The influence of potato cultivar and soil type on effectiveness of plant growth-promoting rhizobacteria (PGPR) was examined. Rhizobacteria were isolated from potato roots and tubers obtained from fields with a history of high potato yields. Fluorescent pigment-producing rhizobacteria. identified as strains of Pseudomonas putida and P. fluorescens, were selected for their antibiosis against Erwinia carovotora ssp. carotovora and growth-promoting activity on potatoes. In greenhouse tests, treatments of potato seedpieces and stem cuttings increased shoot dry weight from 1.23- to 2.00-fold and root dry weight from 1.27- to 2.78-fold. Survival of PGPR in the rhizosphere was monitored using antibioticresistant strains. Populations of these strains decreased from 3.6 × 109 cgu g?1 dry root weight to 4.5 × 105 cfu g?1 dry root weight 4 weeks after treatment. In field trials, PGPR strains were applied to seedpieces of cultivars Kennebec, Pungo, Red Pontiac and Superior and planted in Cape Fear loam. Plymouth loamy sand or Delanco sandy loam. Significant yield increases of 1.17–1.37-fold over controls were observed in two of three field trials. Variability in plant growth-promoting activity was observed between greenhouse and field trials, and no given treatment combination of PGPR strain, potato cultivar and soil type was consistently better than another.  相似文献

In a variety of human genetic diseases, replacement of the absent or defective protein provides significant therapeutic benefits. As a model for a somatic cell gene therapy system, cultured murine fibroblasts were transfected with a human growth hormone (hGH) fusion gene and cells from one of the resulting clonal lines were subsequently implanted into various locations in mice. Such implants synthesized and secreted hGH, which was detectable in the serum. The function of the implants depended on their location and size, and on the histocompatibility of the donor cells with their recipients. The expression of hGH could be modified by addition of regulatory effectors, and, with appropriate immunosuppression, the implants survived for more than 3 months. This approach to gene therapy, here termed "transkaryotic implantation," is potentially applicable to many genetic diseases in that the transfected cell line can be extensively characterized prior to implantation, several anatomical sites are suitable for implantation, and regulated expression of the gene of therapeutic interest can be obtained.  相似文献

Signs of severe otitis media in 20% of dairy calves on one farm were associated with Mycoplasma bovis infection, based on isolation from the external ear canal and nares. Affected calves seroconverted to M. bovis and no other significant bacteria were isolated. Infection was considered likely to have originated from cows in the milking herd based on evidence of seroconversion and detection of infection in a milk sample. M. bovis infection should be considered when investigating otitis problems in calves.  相似文献
