Midday leaf water potential (Ψmd) was monitored for 3 years at a commercial vineyard (cv. Pinot Noir) under four irrigation strategies. Three treatments were established based on irrigating vines with 4–6 mm/day, when daily measured Ψmd was more negative than the pre-defined threshold. After the first experimental year, thresholds were adjusted for each treatment as: (1) Control (C), irrigated when Ψmd was less than −0.6 MPa at the beginning of the season and gradually fell to −0.8 MPa at about mid-June, after which the threshold was maintained at −0.8 MPa until harvest. (2) Control–Deficit (CD), irrigated as C from bud-break to mid-June (around the middle of Stage II of fruit growth), and from then until harvest when Ψmd decreased below −1.2 MPa. (3) Deficit–Deficit (DD), irrigated when Ψmd was less than −1.0 from bud break to mid-May (about the middle of fruit growth Stage I), and after that time the Ψmd threshold became −1.2 MPa until harvest. A fourth treatment was applied following a soil water budget approach (WB). All treatments were replicated five times but irrigation in the Ψmd-based treatments were independently applied to each of the replicate plots, whereas irrigation for WB was applied equally to all replications. The more site-specific information obtained from Ψmd thresholds in C provided substantial advantages for yield homogeneity and repeatability of results with respect to WB, thus demonstrating the method’s greater ability to account for spatial variability. Average applied water for the 3 years in C, CD, and DD was 374, 250, and 178 mm, respectively, while the yields were 11.8, 9.2, and 6.1 kg/vine, respectively. The CD treatment produced better juice quality than C, and was superior in other quality parameters to both C and DD. However, over the study period, an important carryover effect was observed in the yields and the grape size of CD, which tended to diminish from year to year relative to C.  相似文献

Genetic diversity was evaluated by RAPD markers and morpho-agronomic characters for a total of 42 accessions of Barberton daisy (Gerbera jamesonii) consisting of 29 commercial and 13 wild accessions. A total of 74 polymorphic bands were obtained employing a set of 12 primer pairs. The average genetic similarity coefficient for the 42 accessions, evaluated by Jaccard index was 0.55 ranging from 0.28 to 1.00. The genetic structure found among Barberton daisy accessions was evaluated by hierarchic classification analyses and UPGMA modeling, revealing six clusters of genotypes where two of them include the wild accessions and the remaining four including commercial material, except for wild genotype number 9. Shannon (H′) index was calculated using the molecular markers to investigate the genetic variation among the Gerbera accessions and showed higher values for the commercial cluster in comparison to the values obtained for the individuals from the non-commercial cluster, namely 0.34 versus 0.27, respectively. Therefore, both calculated indices (Jaccard and Shannon) indicated the presence of higher genetic variation among commercial accessions in comparison to the cluster representing non-commercial accessions, suggesting that genetic breeding programs may focus on commercial accessions to recombine interesting genotypes with commercially important and marketing-desired characteristics.  相似文献

The antioxidant status of the red blood cells of buffaloes (n = 20) suffering from post-parturient haemoglobinuria (PPH) was assessed by comparing their tocopherol (vitamin E) and reduced glutathione contents with those of red blood cells from apparently healthy buffaloes (n = 20). The red cell tocopherol content of the diseased buffaloes (1.76±0.11 g/ml) was significantly (p<0.01) lower than that of healthy buffaloes (2.45±0.14 g/ml). This may be the first report comparing the concentration of tocopherol in the red blood cells of buffaloes suffering from PPH and apparently healthy buffaloes.There was a drastic reduction in the reduced glutathione content in the red cells of haemoglobinuric buffaloes (23.74±2.86 mg%) compared to the healthy control buffaloes (73.71±3.87 mg%). The diseased buffaloes also exhibited severe hypophosphataemia. These findings suggest that an impaired or insufficient antioxidant potential of the red blood cells in this disease in buffaloes is associated with the phosphorus deficiency.  相似文献

Geranium niveum S. Watson (Geraniaceae) is a medicinal herb widely used by the Tarahumara Indians of Mexico. This species is rich in proanthocyanidins and other phenolics. Previous in vitro assays have demonstrated that proanthocyanidins exhibited antiinflammatory, antiviral, antibacterial, enzyme-inhibiting, antioxidant, and radical-scavenging properties. In view of its medicinal use and chemical composition, the aim of the present study was to determine the in vitro antioxidant activity of the extracts and two proanthocyanidins (geranins A and D) from the roots of G. niveum by using seven different assay systems, namely, 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS), 1,1-diphenyl-2-picrylhydrazyl (DPPH), superoxide anion (O2*-), hydrogen peroxide (H2O2), hydroxyl radical (OH*), hypochlorous acid (HOCl), and singlet oxygen ((1)O2). Two known antioxidants, resveratrol and ascorbic acid, were used as positive controls. The results showed that geranins A and D and the extracts were able to scavenge ABTS, DPPH, O2*-, OH*, and HOCl. The scavenging ability of geranins A and D was similar to that of resveratrol and ascorbic acid in the following assays: ABTS, O2*-, and HOCl. The scavenging capacity of ascorbic acid for DPPH was higher than that of both geranins and resveratrol. On the other hand, the OH* scavenging action of both geranins and resveratrol was similar. The methanol-CHCl3 (1:1) extract had a higher ability to scavenge ABTS, DPPH, and O2*- radicals than the chloroform extract. In turn, the latter was more potent than the methanol-CHCl3 (1:1) extract as OH* or HOCl scavenger agent. Neither geranins A and D nor the extracts were able to scavenge H2O2 and (1)O2. In conclusion, G. niveum roots have proanthocyanidins with powerful radical scavenging in vitro activity. This property may partially explain the wide use of this plant in the Tarahumara indigenous system of medicine for the treatment of gastrointestinal illnesses (other than spasms), pain, and fevers associated with oxidative stress.  相似文献

Improved approaches for irrigation scheduling require specific protocols for adaptation to different growing conditions. We assessed crop intercepted radiation as the main factor for decision on irrigation scheduling. Over two growing seasons (2007-2008), apple trees growing in a large weighing lysimeter were used to measure daily canopy transpiration (Td). Seasonal patterns of daily canopy intercepted photosynthetically active radiation (IPARd) and midday stem water potential were also measured. In 2007, irrigation was withheld in two different times to study Td responses to midday stem water potential. Before harvest, under full irrigation, Td increased linearly with IPARd (R2 = 0.81 in 2007 and 0.84 in 2008). With the two year data combined, R2 increased from 0.74 to 0.80 when VPD was considered as a second variable. When irrigation was withheld in 2007 the ratio between Td and IPARd, which is defined here as transpiratory radiation use efficiency (TRUE), decreased linearly (R2 = 0.49) as midday stem water potential decreased. Due to the highly significant effect of IPARd and VPD on Td, TRUE showed potential applications in estimating the amount of irrigation water.  相似文献

We report here the self-assembly of macroscopic sacs and membranes at the interface between two aqueous solutions, one containing a megadalton polymer and the other, small self-assembling molecules bearing opposite charge. The resulting structures have a highly ordered architecture in which nanofiber bundles align and reorient by nearly 90 degrees as the membrane grows. The formation of a diffusion barrier upon contact between the two liquids prevents their chaotic mixing. We hypothesize that growth of the membrane is then driven by a dynamic synergy between osmotic pressure of ions and static self-assembly. These robust, self-sealing macroscopic structures offer opportunities in many areas, including the formation of privileged environments for cells, immune barriers, new biological assays, and self-assembly of ordered thick membranes for diverse applications.  相似文献

This study aimed to find a way for increasing the water content after rehydration of microdiet particles for fish larvae and to assess whether such increase contributes to a better disaggregation and digestion of particles within the gut of gilthead seabream larvae (4–15 days after hatching, dph). Four microdiets were prepared with increasing amounts of carboxymethylcellulose (CMC): 0, 20, 40 and 80 g kg?1. The inclusion of CMC resulted in an effective increase of the hydration capacity of particles. Water content after immersion ranged from 800 g kg?1 in the microdiet without CMC to 900 g kg?1 with 80 g kg?1 CMC. Larvae of different ages were fed during 10 h with these microdiets. The microparticles containing 80 g kg?1 CMC were more degraded in the gut of the younger larvae. No clear differences were observed from 10 dph onwards. Differences in enzymatic activities were not evident except for α‐amylase in 6 dph larvae. Gene expression of the enzyme precursors did not show any relation to the water content of microdiets. Overall, the results indicate that increasing the water content up to 900 g kg?1 may enhance the disintegration of food particles within the gut of younger larvae and to a lesser extent affect the response of digestive enzyme activities.  相似文献

We studied the relief of water stress associated with fruit thinning in pear (Pyrus communis L.) trees during drought to determine what mechanisms, other than stomatal adjustment, were involved. Combinations of control irrigation (equal to crop water use less effective rainfall) and deficit irrigation (equal to 20% of control irrigation), fruit load (unthinned and thinned to 40 fruits per tree) and root pruning (pruned and unpruned) treatments were applied to pear (cv. 'Conference') trees during Stage II of fruit development. Daily patterns of midday stem water potential (Psi(stem)) and leaf conductance to water vapor (g(l)) of deficit-irrigated trees differed after fruit thinning. In response to fruit thinning, gl progressively declined with water stress until 30 days after fruit thinning and then leveled off, whereas the effects of decreased fruit load on Psi(stem) peaked 30-40 days after fruit thinning and then tended to decline. Soil water depletion was significantly correlated with fruit load during drought. Our results indicate that stomatal adjustment and the resulting soil water conservation were the factors determining the Psi(stem) response to fruit thinning. However, these factors could not explain differences in daily patterns between g(l) and Psi(stem) after fruit thinning. In all cases, effects of root pruning treatments on Psi(stem) in deficit-irrigated trees were transitory (Psi(stem) recovered from root pruning in less than 30 days), but the recovery of Psi(stem) after root pruning was faster in trees with low fruit loads. This behavior is compatible with the concept that the water balance (reflected by Psi(stem) values) was better in trees with low fruit loads compared with unthinned trees, perhaps because more carbon was available for root growth. Thus, a root growth component is hypothesized as a mechanism to explain the bimodal Psi(stem) response to fruit thinning during drought.  相似文献

The ability of Mycobacterium paratuberculosis to survive the commercial pasteurization process of raw milk remains controversial. In a study undertaken in Venezuela to culture M. paratuberculosis from commercially pasteurized cows' milk, 83-200 ml containers of milk were processed and cultured on Herrold's egg yolk slants. No M. paratuberculosis was cultured but a total of six colonies of Mycobacterium bovis were isolated from one container each from two different milk providers. Because laboratory cross-contamination was suspected, the laboratory records were reviewed and spoligotyping was carried out on the isolated individual colonies. On the day before these milk specimens were processed, the biological safety cabinet had been used for the isolation of M. bovis from lymph nodes from infected cattle. Spoligotyping showed that that the colonies isolated from the milk all had the same pattern as the strains isolated from the lymph nodes that were processed the previous day. As far as we know, this is the first report of cross-contamination in a veterinary mycobacterial laboratory. False-positive cultures in the mycobacterial laboratory are not rare. In this setting M. bovis was isolated because it is the most common manipulated organism in this laboratory. We believe that reports on the isolation of M. paratuberculosis from commercially pasteurized milk should exclude cross-contamination before reporting, especially when this organism is routinely isolated from animal material in the same lab.  相似文献
