An isolate of Trichoderma harzianum Rifai from an infected cacao pod produces and secretes nonanoic (pelargonic) acid into a liquid culture medium. Nonanoic acid (NA) was very inhibitory to spore germination and mycelial growth of two cacao pathogens, Crinipellis perniciosa Stahel and Moniliophthora roreri Cif. H.C. Evans. It was highly active causing 75% inhibition of spore germination in an in vitro assay at a rate as low as 0.09 μM for M. roreri and 0.92 μM for C. perniciosa. Mycelial growth was comparatively less sensitive to inhibition, but still there was a 75% reduction in growth with 0.62 μM in M. roreri and 151 μM NA in C. perniciosa. In contrast, NA did not affect Trichoderma mycelial growth or spore germination at concentrations that were inhibitory to the pathogens. 6-pentyl-α-pyrone was also produced and secreted into the medium by T. harzianum, however; it was not antagonistic to the cacao pathogens. Although a number of metabolites produced by Trichoderma spp. have been identified in the past, this is the first report of NA production and secretion by any Trichoderma. The results suggest that NA may play a role in the successful use of some Trichoderma spp. isolates in the biocontrol of fungal diseases of plants.  相似文献

Soil organic matter level, mineralizable C and N, microbial biomass C and dehydrogenase, urease and alkaline phosphatase activities were studied in soils from a field experiment under a pearl millet-wheat cropping sequence receiving inorganic fertilizers and a combination of inorganic fertilizers and organic amendments for the last 11 years. The amounts of soil organic matter and mineralizable C and N increased with the application of inorganic fertilizers. However, there were greater increases of these parameters when farmyard manure, wheat straw or Sesbania bispinosa green manure was applied along with inorganic fertilizers. Microbial biomass C increased from 147 mg kg–1 soil in unfertilized soil to 423 mg kg–1 soil in soil amended with wheat straw and inorganic fertilizers. The urease and alkaline phosphatase activities of soils increased significantly with a combination of inorganic fertilizers and organic amendments. The results indicate that soil organic matter level and soil microbial activities, vital for the nutrient turnover and long-term productivity of the soil, are enhanced by use of organic amendments along with inorganic fertilizers. Since 1997, avian pneumovirus (APV) has caused estimated annual losses of $15 million to the Minnesota turkey industry. In order to develop an attenuated live vaccine against APV, we serially passaged a Minnesota isolate of APV (APV/MN/turkey/1-a/97) in vitro in cell cultures for 41 passages. Laboratory experiments with this high-passage virus (P41) indicated that the attenuated virus provided immunogenic protection to turkeys against challenge with virulent APV, although some birds showed mild to moderate dinical signs after inoculation. To reduce the residual pathogenicity of P41, while maintaining its immunogenicity, we decided to vaccinate turkeys with P41 in the presence of an immunomodulator, S-28828 (1-n-butyl-2-ethoxymethyl-1H-imidazo[4,5-c]quinolin-4-amine-hydrochloride), which is a potent cytokine inducer. The combined inoculation of S-28828 (5 mg/kg body weight) and P41 resulted in a significant reduction in the incidence of virus-induced clinical signs in comparison with birds that received P41 without immunomodulator (P < 0.05). Only 17% of birds inoculated with S-28828 + APV P41 showed mild respiratory symptoms at 5 days postinoculation as compared with 46% of the vaccinated turkeys that did not receive S-28828. Vaccination with either P41 or with P41 + S-28828 protected turkeys against dinical signs and viral replication after challenge with virulent APV. These results indicate that immunomodulators, such as S-28828, may act as good vaccine adjuvants that can reduce the pathogenicity but maintain the immunogenicity of partially attenuated vaccines.  相似文献

The H3N2 subtype of influenza A viruses isolated from pigs in the United States and Canada has shown both genetic and antigenic diversity. The objective of this study was to determine the serologic and genetic characteristics of contemporary strains of these viruses. Genetic analysis of 18 reference strains and 8 selected strains demonstrated differences in 1% to 9% of the nucleotides of the hemagglutinin (HA) gene. Phylogenetic analysis of the HA gene revealed 3 genetic clusters, as well as divergence of cluster III viruses from a cluster III prototype virus (A/Swine/Illinois/21587/99). By means of 1-way cross-hemagglutination inhibition with antiserum against 5 field isolates and 3 vaccine viruses, most of 97 isolates tested could be placed in 1 of 3 serogroups. The several isolates that did not react with any antiserum were in genetic cluster III, which suggests that continuous antigenic drift in cluster III may have resulted in virus variants. The efficacy of commercial vaccines against these virus variants should be evaluated with vaccination and challenge studies.  相似文献

  1. India plays a significant role in dugong conservation by having the largest population within South Asia. The status of dugongs in India is largely unknown due to a paucity of reliable ecological data. This study generated mitochondrial control region sequences from ~10% of dugong individuals from existing populations within India. Furthermore, data generated in this study were compared with the global data to assess genetic lineages, population structure, and genetic diversity of Indian populations.
  2. Multiple analyses suggest that the Indian dugong populations are part of a single genetic cluster, comprising South Asia, North-west Indian Ocean, and South-west Indian Ocean populations. Despite small population size, they retain high genetic diversity with unique mitochondrial DNA haplotypes within South Asia. Within India, novel haplotypes are observed from all dugong habitats sampled, with overall high haplotype diversity (0.85 ± 0.04) but low nucleotide diversity (0.005 ± 0.001). Indian populations exhibit genetic differentiation with higher within-population variance (63.41%) than among populations (36.59%). Two of the haplotypes observed in India are shared with Sri Lanka, implying genetic connectivity between these populations.
  3. The genetic data from Indian dugong populations provide critical insights into the identification of dugong corridors and important dugong conservation zones in India. We suggest site-specific interventions, including the creation of new marine protected areas and boundary reorganization and expansion of other existing protected areas, to ensure population connectivity. In addition, simultaneous efforts towards seagrass meadow restoration, reduction of dugong mortalities, and community participation in dugong conservation are recommended for population recovery of this threatened marine herbivore.

The view that biodiversity-rich areas partially or largely managed by local residents, sometimes referred to as community-conserved areas (CCAs), can be effective in biological conservation has gained considerable ground over the past decade. In this paper, we review available scientific information on the conservation effectiveness of such areas globally. We compiled studies undertaken during the last 5 years (2004-2009) that use quantifiable ecological attributes to: (1) compare CCAs with strictly protected areas (SPAs); (2) compare CCAs with open-access ecosystems and (3) study trends in biological attributes of CCAs over time. We found that there were few consistent differences in diversity/species richness of flora or fauna protected under the two types of management or in deforestation rates. However, CCAs tend to harbour a species complement distinct from that of SPAs and show lowered abundances of monitored taxa that are of conservation importance. CCAs conserve biological values more effectively than open-access areas. Also, biological values tend to decline in CCAs over time. We conclude that CCAs could represent a significant improvement over open-access areas in terms of conservation effectiveness, yet fall short of the needs of comprehensive biological conservation. While extremely few studies have been undertaken in India, the trends seen largely concur with global ones. This review, based on a limited sample size, is only a beginning, and is expected to serve as an invitation for further research to address both the question of biological effectiveness of diverse forest governance regimes as well as the socio-economic, demographic and institutional reasons underlying these differences.  相似文献
