OBJECTIVE: To evaluate the potential of an implant of a GnRH-agonist (deslorelin) to create a progesterone free animal suitable for studying progesterone (P4) metabolism in intact cows by measuring blood P4 and faecal P4 metabolites. METHODS: Experiment 1: Eighteen non-lactating cycling Holstein-Friesian cows, 4 to 7 years old, were allocated to one of three groups to study plasma P4 concentrations preceding an intravaginal insert. These groups comprised: i) a deslorelin group (GnRH-agonist implanted); ii) a PGF group receiving two injections of prostaglandin (PGF2alpha) 12 days apart; and, iii) an ovariectomised (OVX) group. An intravaginal device (CIDR) was inserted into the vagina of each animal and left in place for 11 days. Plasma P4 concentrations were measured during the study period. Experiment 2: Twelve non-lactating cycling Holstein-Friesian cows, 4 to 7 years old, were allocated to two groups: i) a deslorelin group (GnRH-agonist implanted); and ii) an ovariectomised group. Plasma P4 and faecal P4 metabolites (20-oxo-pregnanes, 20alpha-OH and 20beta-OH) were monitored for a period of 5 weeks. RESULTS: Experiment 1: Average plasma P4 concentration did not differ between the three groups (1.28, 1.43 and 1.55 ng/mL for deslorelin, OVX and PGF cows, respectively, P = 0.8) during the period of supplementation. Experiment 2: There was no difference in plasma P4 (mean plasma P4 < 0.02 ng/mL, P = 0.9) and faecal P4 metabolites between deslorelin and OVX cows 2 weeks after the implantation (P = 0.7). CONCLUSIONS: These data showed that a GnRH-agonist (deslorelin) implant may be used as an alternative to ovariectomy to create a progesterone free animal suitable for studying the metabolism of administered P4.  相似文献

Evaluation of Tissue Adhesive to Contain Axonal Regeneration in Horses   总被引:1,自引:1,他引:0  
Bilateral palmar and plantar digital neurectomies were completed in 10 horses (a total of 80 neurectomies) using one of three methods: (1) simple transection (guillotine method); (2) epineural capping; (3) n-butyl cyanoacrylate injected into the epineural sheath to act as a nerve sealant. Horses were regularly evaluated clinically for tenderness in and around the surgical site, as well as skin sensation at the coronary band in the heel region, during the 12-week course of the study. None of the surgical sites exhibited any signs of drainage or infection. Horses were then euthanatized, the nerve stumps were dissected from surrounding tissues, and the length and width of the tissue mass that had formed on the end of the nerve was recorded. Longitudinal and transverse sections of the nerve endings were examined histologically for numbers of proliferating axon sprouts (neuroma formation); whether the axons had penetrated the epineurium; degree of Schwann cell proliferation; degree of chronic inflammation; extent of foreign body reaction; extent of retrograde degeneration of the nerve bundles; and amount of fibro vascular proliferation. The proportion of legs exhibiting tenderness or heel sensation did not differ significantly between the three different treatments at any of the six different times they were examined. There was no difference between the three treatments in the length or width of the fibrous tissue scar on the ends of the nerves or in the number of sprouting axons from the ends of the nerves. Of 80 nerves examined, only two nerves were not confined to the epineurium. Both these nerves had been treated by simple transection. Statistically there was more chronic inflammation and foreign body reaction in the acrylic treated nerves, but no difference in Schwann cell proliferation or retrograde degeneration between the three treatments. There was slightly less fibrovascular proliferation in the transected nerves than in those subjected to epineural capping or acrylic, but the difference was not statistically significant. The use of the tissue adhesive n-butyl cyanoacrylate to prevent the continuous growth of axons after digital neurectomy seems to offer little advantage over more traditional methods of neurectomy.  相似文献

Hepatic Lobe Torsion as a Cause of Colic in a Horse   总被引:2,自引:0,他引:2  
A 14-year-old Arabian gelding was examined for colic. An exploratory celiotomy was subsequently performed and the left lobe of the liver was found to be twisted. The lobe was resected using a TA-90 surgical stapling instrument. Histologic examination of the resected liver indicated portal vein and sinusoid dilation and congestion with blood. There were focal areas of necrosis and bacterial cocci and rods throughout the section. The histologic findings were consistent with hepatic lobe torsion. After surgery, the horse was treated with broad spectrum antibiotics, anti-inflammatory drugs, heparin, and intravenous fluids. The horse recovered without complications, although serum liver enzymes remained elevated for more than 1 week after surgery. Seven months after surgery the horse showed no adverse affects from the disease.  相似文献

To assess respiratory function in dogs with tick paralysis, respiratory measurements were recorded on 14 dogs experimentally infested with Ixodes holocyclus. There was a progressive fall in respiratory rate with no change in tidal volume, which resulted in a significant fall in minute respiratory volume in the latter stages of the disease. The fall in respiratory rate was possibly central in origin and was accompanied by an increased alveolar-arterial oxygen tension difference, probably caused by pulmonary congestion and oedema. The "grunting" respiration seen in tick paralysis was due to closure of the vocal cords during expiration and could represent an attempt to re-expand collapsed parts of the lung.  相似文献

Summary. Three box experiments examining the effects of shoot removal on rhizome carbohydrate reserves are reported.
During summer in England the optimum frequency of hand defoliation for maximum loss of carbohydrate was 10–14 days. As in previous experiments, high levels of soil nitrogen sometimes increased the rate of carbohydrate loss from 9-in. rhizome pieces. This result was obtained when nitrogen was supplied as a nitrate, an ammonium salt, or urea.
Repeated applications of 2 or 4 oz/ac paraquat over a period of about 60 days reduced carbohydrate reserves to about 50% of the level obtained with the same number of hand clippings. It is suggested that paraquat may have been translocated to the rhizomes, the carbohydrate content of which may have been reduced by an effect of the paraquat on the respiration rate.
When plants which had been subjected to different methods and frequencies of defoliation were uprooted and buried beneath 2 in. of soil, there was a connection between the rhizome carbohydrate content at the time of burial, the ability of the plants to regenerate, and the rate at which new carbohydrate reserves built up during the following two months.  相似文献

Rotaviruses (RV) have a high prevalence in piggeries worldwide and are one of the major pathogens causing severe diarrhoea in young pigs. RV species A, B, and C have been linked to piglet diarrhoea in Australian pig herds, but their genetic diversity has not been studied in detail. Based on sequencing of the structural viral protein 7 (VP7) RVA G genotypes G3, G4 and G5, and RVC types G1, G3, G5, and G6 have been identified in Australian piggeries in previous studies. Although occurrence of RVB was reported in Australia in 1988, no further genetic analysis has been conducted. To improve health management decisions in Australian pig herds, more information on RV prevalence and genetic diversity is needed. Here, 243 enteric samples collected from 20 pig farms within Eastern Australia were analysed for the presence of RV in different age groups using a novel PCR-based multiplex assay (Pork MultiPath™ enteric panel). RVA, RVB, and RVC were detected in 10, 14, and 14 farms, respectively. Further sequencing of VP7 in selected RV-positive samples revealed G genotypes G2, G5, G9 (RVA), G6, G8, G14, G16, G20 (RVB), and G1, G3, G5, G6 (RVC) present. RVA was only detected in young (<10 weeks old) pigs whereas RVB and RVC were also detected in older animals (>11 weeks old). Interestingly, RVB and RVC G-type occurrence differed between age groups. In conclusion, this study provides new insights on the prevalence and diversity of different RV species in pig herds of Eastern Australia whilst demonstrating the ability of the Pork MultiPath™ technology to accurately differentiate between these RV species.  相似文献

The variation in nitrogen (N) uptake by rice has been widely studied but differences in rice root morphology that may contribute to this variation are not completely understood. Field and greenhouse experiments were carried out to study N accumulation, root dry weights, total root lengths, root surface areas, and root bleeding rates of two rice cultivars, Elio with low N-use efficiency and Nanguang with high N-use efficiency. Low (1 mmol N L-1) and high (5 mmol N L-1) N applications were established in the greenhouse experiment, and the N rates were 0, 120, and 240 kg ha-1 in the field experiments at Jiangning and Jiangpu farms, Nanjing, China. The results showed that the N accumulation, root dry weight, total root length, and root surface area increased with an increase in N application. At the heading stage, N accumulation in the shoots and roots of Nanguang was greater than that of Elio in the field experiments and that of Elio at 5 mmol N L-1 in the greenhouse experiment. After the heading stage, N accumulation was higher for Nanguang at both 1 and 5 mmol N L-1 in the greenhouse experiment. The total root length and root surface area were significantly different between the two cultivars. Over the range of the fertilizer application rates, the root lengths of Nanguang at Jiangning Farm were 49%-61% greater at booting and 26%-39% greater at heading than those of Elio, and at Jiangpu Farm they were 22%-42% and 26%-38% greater, respectively. Nanguang had a greater root bleeding rate than Elio. It was concluded that the N-use efficiency of the two rice cultivars studied depended to a great extent on the root morphological parameters and root physiological characteristics at different growth stages.  相似文献
