UVB radiation proves to be one of the most relevant environmental risks because of its hazardous effects, such as premature skin aging and especially skin photocarcinogenesis. Anthocyanins, water-soluble pigments present in plants, are known to be powerful antioxidants that help protect plants from UV damage. In this study, we aimed at investigating the protective effect of anthocyanins from black soybean [Glycine max (L.) Merr] seed coats on UVB-induced apoptosis, and furthermore, we investigated the molecular mechanism responsible for regulation of apoptosis in vitro and in vivo. Pretreatment with anthocyanins reduced UVB-induced reactive oxygen species levels and inhibited UVB-induced apoptotic cell death through the prevention of caspase-3 pathway activation and reduction of proapoptotic Bax protein levels. UVB irradiation induced apoptotic cell death, which was inhibited by topical application of anthocyanins in hairless mice. It is concluded that anthocyanins from the seed coat of black soybeans may be useful compounds to modulate UVB-induced photoaging.  相似文献

The Eurasian badger (Meles meles) is a wildlife reservoir for Mycobacterium bovis infection in Ireland and Great Britain and has been implicated in the transmission of tuberculosis to cattle. Vaccination of badgers is an option that could be used as part of a strategy to control the disease. In this study we used an endobronchial infection procedure to inoculate groups of badgers with three different doses (3x10(3), 2x10(2) and <10 Colony Forming Units (CFUs)) of M. bovis. After 17 weeks the disease status of each animal was determined by post-mortem pathology and culture for M. bovis. Each of the inoculum doses resulted in establishment of infection in the badgers. The cell-mediated immune (CMI) responses were measured by lymphocyte transformation assay (LTA) of peripheral blood mononuclear cells (PBMCs) cultured with bovine tuberculin (PPD-B). In each infected group the CMI responses increased with a kinetic profile corresponding to the delivered dose and the post-mortem pathology. The serological responses were measured by ELISA and a multi-antigen print immunoassay (MAPIA) in order to investigate any changes in the antigenic repertoire associated with different infective doses. In contrast to the CMI responses, the ELISA and MAPIA showed that the recognition of antigens by the badgers was intermittent and not strongly influenced by the dose of M. bovis.  相似文献

Myelo-computed tomography of a paraparetic 14-year-old dog revealed subarachnoid distension with an intradural filling defect above the T13–L1 disc space. T12–L1 hemilaminectomy followed by durotomy allowed removal of a large piece of degenerated disc material that compressed the spinal parenchyma. Full return to function was achieved 10 days post-surgery. The distension was likely secondary to the intradural herniation, and is a rare and distinct finding.  相似文献

The C isotopic composition (δ13C) of pedogenic carbonates reflects the photosynthetic pathway of the predominant local vegetation because pedogenic (secondary) CaCO3 is formed in isotopic equilibrium with soil CO2 released by root and rhizomicrobial respiration. Numerous studies show the importance of pedogenic carbonates as a tool for reconstructing paleoecological conditions in arid and semiarid regions. The methodological resolution of these studies strongly depends on the time scale of pedogenic carbonate formation, which remains unknown. The initial formation rate can be assessed by 14C labeling of plants grown on loess and subsequent incorporation of 14C from rhizosphere CO2 into newly formed carbonate by recrystallization of loess CaCO3. We tested the feasibility of 14C and 13C tracers for estimating CaCO3 recrystallization rates by simultaneous 14C and 13C labeling and comparison with literature data. 14C labeling was more efficient and precise in assessing recrystallization rates than 13C labeling. This is connected with higher sensitivity of 14C liquid scintillation counting when compared with δ13C measurement by IRMS. Further, assessment of very low amounts of incorporated tracer is more precise with low background signal (natural abundance), which is true for 14C, but is rather high for 13C. Together, we obtained better reproducibility, higher methodological precision, and better plausibility of recrystallization rates calculated based on 14C labeling. Periods for complete CaCO3 recrystallization, extrapolated from rates based on 14C labeling, ranged from 130 (125–140) to 240 (225–255) y, while it was ≈ 600 (365–1600) y based on the 13C approach. In terms of magnitude, data from late‐Holocene soil profiles of known age provide better fit with modeled recrystallization periods based on the 14C approach.  相似文献

According to the United Nations International Panel on Climate Change good practice guidance, an annual forest biomass carbon balance (AFCB) can be estimated by either the stock-difference (SD) or the gain–loss (GL) method. An AFCB should be accompanied by an analysis and estimation of uncertainty (EU). EUs are to be practicable and supported by sound statistical methods. Sampling and model errors both contribute to an EU. As sample size increases, the sampling error decreases but not the error due to errors in model parameters. Uncertainty in GL AFCB estimates is dominated by model-parameter errors. This study details the delta technique for obtaining an EU with the SD and the GL method applicable to the carbon in aboveground forest biomass. We employ a Brownian bridge process to annualize the uncertainty in SD AFCBs. A blend of actual and simulated data from three successive inventories are used to demonstrate the application of the delta technique to SD- and GL-derived AFCBs during the years covered by the three inventories (SD) and rescaled national wood volume harvest statistics (GL). Examples are limited to carbon in live trees with a stem diameter of 7 cm or greater. We confirm that a large contribution to the uncertainty in an AFCB comes from models used to estimate biomass. Application of the delta technique to summary statistics can significantly underestimate uncertainty as some sources of uncertainty cannot be quantified from the available information. We discuss limitations and problems with the Monte Carlo technique for quantifying uncertainty in an AFCB.  相似文献

Seedlings of Pistacia vera L. developed from seeds of two separate populations in Turkmenistan, Kepele and Agachli, were evaluated for their growth potential and genetic polymorphism. Plant growth rate as well as random amplified polymorphic DNA (RAPD) analysis showed distinct differences between the two populations. In plant height growth rate, 17 Agachli accessions were 1.3 times higher on average than that of 10 accessions of Kepele (significant at p = 0.046) and 1.2 times higher for trunk diameter growth rate (p = 0.062). Cluster analysis divided most accessions into two main genetic groups according to their geographic origin. The Agachli group was further divided into two subgroups. One Kepele accession (K4), was genetically different from the rest and clustered on a separate outgroup. Two Agachli accessions (A12 and A17) were outside the two main populations clusters. Accessions K9 and K10 from Kepele were exceptional and were clustered in each of the two Agachli's subgroups, indicating a close genetic relationship between the two populations. In addition, high similarity values (0.58–1.00) and small genetic distances reflect plausible gene flow between Kepele and Agachli, which are 100 Km apart. Mantel test revealed significant relationship between the RAPD and the morphological traits matrices, pointing to the genetic basis for the measured differences in the growth rate. Growing the accessions on the same plot, under similar conditions enabled the evaluation of genotypic differences. The combination of morphological traits and molecular markers will further assist in preservation of genetic variability and cultivation of useful genotypes of P. vera L.  相似文献

Soil macroinvertebrates were studied in a Mediterranean-type forest on brown-pebble forest soils in southern Russia. At the site, 144 intact soil cores (76 cm2 each) forming a grid of 24 × 6 units were taken in order to determine animal spatial distribution. Abundance of isopods was 166.3 ± 16.0 indiv. m–2 and they constituted about 12% of the total macrofaunal abundance. Biomass of isopods was 3.5 g m–2, or about 21% of the total biomass of macrofauna. Three woodlice genera (Armadillidium, Cylisticus, and Trachelipus) were found at the site. The two latter genera formed almost all (93%) of the isopod population. We found that spatial distribution of woodlice was heterogeneous: areas with 4–5 individuals per sample were neighboring those without animals. In order to study soil factors influencing isopod distribution in the brown-pebble forest soil, the size of a sample was artificially increased by combining adjacent sample units. Litter mass (r = 0.41) and loss on ignition (LOI) (r = –0.55) significantly influenced isopod distribution. Soil pH was near neutral (6.79), LOI was 8.39, and the water holding capacity was 70.9%. Pebbles comprised up to 84% of the sample's mass. Ca. 40 samples are recommended for estimation of isopod abundance in brown forest soil.  相似文献

Russia, Ukraine, Belarus, Kazakhstan and some other neighboring countries bear the heritage of several Soviet era nuclear disasters and the resulting severe radioactive pollution of vast territories. The most famous of them is the Chernobyl catastrophe on April 26, 1986 which resulted in a massive radioisotope fallout (0.185 MBq m−2 or higher) over about 25,000 km2 of the territory of the former USSR alone. Extensive radioecological research around Chernobyl demonstrated that despite high resistance of most of soil-dwelling organisms to ionizing radiation, some soil animals were very vulnerable to radioactive pollution due to low motility, direct contact with hot particles and radioisotope accumulation in soil. These are the reasons that soil organisms are very important organisms for long-term radioecological observations. In this review, we analyze published data on the response of different soil taxa to radioisotope contamination of soil near Chernobyl and other nuclear accident locations. Field results are compared with the available experimental data. Earthworms, millipedes, collembolans and oribatid mites were recognized as the most appropriate biomonitors of different radioactivity levels and types of radioactive pollution. Synthesis of this knowledge allowed us to propose a multilevel system of soil radioecological monitoring, which may be useful for studying the short- and long-term environmental consequences of the recent catastrophe at Fukushima-1 nuclear power plant in Japan, as well as other locations vulnerable to radioactive pollution.  相似文献
