Veterinary inspection in slaughterhouses allows for the detection of macroscopic lesions reminiscent of bovine tuberculosis, but the presence of Mycobacterium bovis must be confirmed by laboratory methods. This study aimed at comparing the performances of the standard diagnostic tools used to identify M. bovis in tissue specimens sampled from suspicious animals. During a two years period, 1390 cattle were inspected at the Machachi abattoir in the Mejia canton - Ecuador. A total of 33 animals with granulomatous lesions were detected, representing 2.33% (16/687) and 2.42% (17/703) animals examined in 2007 and 2008, respectively. Ninety-four tissue specimens were sampled and screened for the presence of mycobacteria. Acid-fast bacilli were identified in one third of the suspicious cattle (11/33) and suggestive microscopic lesions in 27.3% (9/33) of the samples examined by direct microscopy and histopathology, respectively. Culturing on Stonebrink medium and 16S-rRNA-based polymerase chain reaction (PCR) yielded 36.4% (12/33) and 27.3% (9/33) of positives, respectively. Compared to culture, other diagnostic procedures displayed a lower sensitivity, with 56.5% for PCR, and 43.5% for direct microscopy and histopathology; however, the specificity was higher (94.4% for PCR and microscopy, and 97.2% for histopathology). We conclude that reliable post-mortem laboratory testing either requires the combination of a set of available diagnostic tools or necessitates the development of improved new-generation tools with better sensitivity and specificity characteristics.  相似文献

Native beta-lactoglobulin binds and increases the activity of Kluyveromyces lactis beta-galactosidase. Construction of a three-dimensional (3D) model of beta-lactoglobulin showed that lysine residues 15, 47, 69, and 138 are the most exposed ones, thus the ones more likely to interact with beta-galactosidase. Molecular docking estimated the interaction energies of amino acid residues with either lactose or succinic anhydride, showing that Lys(138) is the most likely to react with both. Affinity chromatography demonstrated that succinylated beta-lactoglobulin diminished its ability to bind to the enzyme. Furthermore, when activity was measured in the presence of succinylated beta-lactoglobulin, its activating effect was lost. Since succinylation specifically blocks Lys epsilon-amino groups, their loss very likely causes the disappearance of the activating effect. Results show that the activating effect of beta-lactoglobulin on beta-galactosidase activity is due to the interaction between both proteins and that this interaction is very likely to occur through the Lys epsilon-amino groups of beta-lactoglobulin.  相似文献

In the present study, digestive dynamics, measured through the changes in the in situ pH along the digestive tract, was evaluated along with the glycogen concentration in the digestive gland (DG) and the proteolytic enzyme activity of the gastric juice (GJ) (both acid and alkaline proteases) to obtain useful information that will allow for the understanding of the DG's function and the role that it plays as an extracellular digestive enzyme source and for energy storage, providing new information on the digestive physiology of Octopus maya. The results showed that pH along the digestive tract changed according to the postprandial time following the food transit. At the beginning, the pH on crop (St) was 5, changing to 6 when the food arrived. Similar changes were observed on the caecum (Ce) and DG. Glycogen from DG is used as a source of energy during digestion and recovered 8 h after feeding. Maximum activities of digestive gland GJ enzymes were observed 6 h after feeding, indicating that chyme enzymes are still active when they arrive at the DG's lumen. The presence of food in the digestive tract modifies the pH, which, in combination with GJ, favours the activity of the released enzymes of O. maya.  相似文献

Although alien and invasive plant species have been researched extensively in the European part of Russia,the situation in Siberia is another matter.Hitherto,alien and invasive species in Siberia have not received much attention because this problem was not especially acute in Siberia.The lack of attention on alien and invasive species in Siberia is attributed to three major reasons:1) Low vegetative productivity and sparse human populations in the Siberian territory have limited botanical research interest in the area.2) Severe Siberian climate likely prevents many alien and invasive species from increasing their distribution into Siberia.3) Most Siberian plant communities have not been human-transformed and thus may be resistant to newcomers.Nevertheless,recent increased economic activities have resulted in increasing plant migration to Siberia,and this process should be monitored.Furthermore,global environmental changes may also have made Siberia more favorable for more alien and invasive species.Currently,research on alien and invasive species has begun in the Altai-Sayan region(Western Siberia) and the Magadan region(Northeastern Asia).  相似文献

The Heteroptera have holocentric chromosomes with kinetic activity restricted to the end of chromosomes. The first meiotic division is reductional for the autosomes and equational for the sexual. Only a few species of this suborder have been analyzed. In this study, we observed the morphologies of the testes of the Heteroptera species Belostoma anurum (Herrich-Schäffer, 1948), Belostoma micantulum (Stal, 1858), Gelastocoris angulatus (Melin, 1929), Gelastocoris flavus flavus (Guérin-Méneville, 1844), Rheumatobates crassifemur crassifemur (Esaki, 1926), Buenoa amnigenus (White, 1879), Buenoa unguis (Truxal, 1953), Martarega brasiliensis (Truxal, 1949), Martarega membranácea (White, 1879), Martarega uruguayensis (Berg, 1883), Rhagovelia tenuipes (Champion, 1898) and Rhagovelia zela (Drake, 1959). We found that the testes of these species can be round, round/spiral, or elongated/spiral. The size of the prophase I cells was found to vary, with the smallest ones being detected in B. micantulum and Rha. zela, the largest in G. f. flavus, and ones of intermediate size in R. c. crassifemur and M. brasiliensis. With respect to the chromosome complement, we verified the presence of 2n = 16 (14A+XY, B. micantulum and G. angulatus), 21 (20A+X0, R. c. crassifemur), 23 (22A+X0, Rha. zela and Rha. tenuipes), 25 (24A+X0, Bu. amnigenus and Bu. unguis; 22A+2m+X0, M. membranacea), 27 (24A+2m+X0, M. brasiliensis and M. uruguayensis), 29 (26A+X1X2Y, B. anurum), and 35 (30A+X1X2X3X4Y, G. f. flavus). We found that the features of spermatogenesis in these species are similar to those of other previously described Heteroptera species, differing only in testicular morphology, chromosome number, and sex chromosome system.  相似文献

Bovine tuberculosis (bTB) is a chronic granulomatous disease that primarily affects lung tissue and lymph nodes (LN) in cattle, with economic impact on their productivity. Furthermore, it is potential zoonoses that may cause public health hazard. In this study, we evaluated the presence of bTB in two abattoirs: Cayambe and Pelileo countries located in the Ecuadorian provinces of Pichincha and Tungurahua, respectively. In total, 578 cattle were sampled (Cayambe 271 and Pelileo 307): 1,156 LN and 578 lung tissue samples were collected to apply in vitro culture and nested-PCR, respectively. The results determined a total apparent prevalence of 4.33 %, with 4.06 % at Cayambe’s abattoir and 4.56 % at Pelileo’s abattoir. Additionally, the Bayesian analysis showed a total true prevalence of 2.51 %, with 89.7 % of sensitivity and 97.6 % of specificity. The risk factors were evaluated by the use of simple logistic regressions with and without the random effect of places of origin. Associations of the origin of cattle in the selected slaughterhouses were found. The results showed an efficient method for the detection of bTB, which could identify a large number of infected animals, and the usefulness of lung tissue samples for early diagnosis of the disease was demonstrated in this study.  相似文献

Two experiments were conducted to evaluate the initial stocking density in larvae of Bay snook, Petenia splendida, and Mexican mojarra, Cichlasoma urophthalmus, using a recirculation system. Five initial stocking densities (0.5, 1, 5, 10, and 20 larvae/L) were evaluated by triplicate for 45 d. Weight and total length (TL) were measured every 15 d, and fish production was calculated for each density. The larvae stocked at the lowest densities (0.5 and 1 larvae/L) presented the highest growth for both species: C. urophthalmus (0.78 g and 45‐mm TL, and 0.76 g and 45‐mm TL, respectively) and P. splendida (0.80 g and 52‐mm TL, and 0.79 g and 49‐mm TL, respectively). However, lowest fish production was recorded (35 and 69 fish per tank, respectively, for C. urophthalmus and 34 and 70 fish per tank, respectively, for P. splendida) compared with those at densities of 5, 10, and 20 larvae/L (336, 584, and 604 fish per tank, respectively, for C. urophthalmus and 341, 679, and 912 fish per tank, respectively, for P. splendida). The polynomial model for biomass production related to the stocking density shows that the optimum stocking densities for C. urophthalmus and P. splendida are 12 and 14 larvae/L, respectively.  相似文献

The presence of Giardia and Cryptosporidium was investigated in 274 faecal samples of alpacas (Vicugna pacos) from 12 herds from Peru by immunofluorescence microscopy and PCR amplification and sequencing of fragments of the ssu-rRNA and β-giardin genes from Giardia spp., as well as the ssu-rRNA gene from Cryptosporidium spp. A total of 137 samples (50.0%) were positive for Giardia spp., and 12 samples (4.4%) for Cryptosporidium spp. In ten samples (3.6%), co-infection by both pathogens was found. Herd prevalence was found to be 91.7% (11/12 herds) for Giardia and 58.3% (7/12 herds) for Cryptosporidium. Regarding the age of the animals, although Giardia was detected in animals as young as 1 week, the prevalence increased with age, reaching 80% by 8 weeks. Similarly, the highest percentage of Cryptosporidium detection (20%) was also found in the 8 week-old group. By PCR, 92 of the 274 analysed samples were positive for Giardia. Sequencing of the amplicons showed the existence of Giardia duodenalis assemblage A in 67 samples; G. duodenalis assemblage E in 24 samples; and inconsistent results between the two molecular markers used in a further sample. Cryptosporidium was only detected by PCR in 3 of the 274 samples; Cryptosporidium parvum was identified in two samples and Cryptosporidium ubiquitum in one sample. This study is the first performing molecular characterisation of both parasites in Peruvian alpacas, and the first report of C. ubiquitum in this host. The identification of G. duodenalis assemblage A, C. parvum and C. ubiquitum, suggests that zoonotic transmission of these enteropathogens between alpacas and humans is possible.  相似文献
