In order to optimize the production of embryos under tropical conditions and to test a possible seasonal effect on embryo quality, 40 Zebu cows were superovulated during the dry season (April to May) and during the rainy season (July to August). A total of 116 (average 2.7/cow) and 83 embryos (3.5 average/cow) were obtained during the respective seasons. After classification as good, fair or poor quality, embryos were tested based on their ultrastructural differences (n = 53 dry season 16 good, 20 fair and 17 poor and n = 61 rainy season 21 good, 20 fair and 20 poor) and their degree of apoptosis using the TUNEL technique (n = 30 during the dry season and n = 55 in the rainy season). Structural characteristics determining embryo quality varied between good and fair quality embryos. No difference, however, was observed between good, fair and poor quality embryos from the two seasons. The number of TUNEL-positive cells was different among embryos (p < 0.001), being lower in labelled cells of good quality embryos regardless of the season. Fewer apoptotic cells were observed in embryos assigned in all three quality levels during the rainy season (p < 0.001). Ultrastructural evaluations confirmed the results obtained by TUNEL. Cryopreserved embryos of good (n = 25 in each season) and fair quality (n = 11 dry season; n = 17 rainy season) showed a significant decrease of TUNEL-positive cells during the rainy season (p < 0.05). Results suggest that embryos collected in the dry season have more cellular damage in contrast; embryos cryopreserved in the rainy season appeared morphologically better equipped to result in a pregnancy following transfer.  相似文献

Background: Splenic venous thrombosis (SVT) is usually considered an incidental finding on abdominal ultrasound examination but can indicate the presence of underlying disease. Concurrent disease processes and conditions in dogs with SVT have not been identified previously. Objectives: To identify concurrent diseases and conditions in dogs with SVT. Animals: Eighty dogs with SVT. Methods: Retrospective review. Medical records from 1994 through 2008 were searched for dogs with SVT identified by ultrasound examination. These records were then reviewed for signalment, medical history, clinicopathologic testing, diagnostic imaging, and clinical diagnosis. Results: The most common concurrent conditions were neoplasia (54%), exogenous corticosteroid administration (43%), systemic inflammatory response syndrome (26%), disseminated intravascular coagulation (20%), pancreatitis (18%), and immune‐mediated disease (16%). The most common neoplastic disease was lymphoma, and the most common immune‐mediated disease was immune‐mediated hemolytic anemia. Protein‐losing nephropathy and naturally occurring hyperadrenocorticism were identified in <10% of the dogs. Concurrent splenic infarcts were identified in 33% of dogs, and concurrent portal vein thrombi were found in 18% of dogs. Conclusions: SVT is a sonographic finding of clinical importance, and dogs with SVT can have 1 or more coexisting diseases.  相似文献

A mathematical model was developed to investigate the impact of level of Teladorsagia circumcincta larval pasture contamination and anthelmintic treatment on genetic parameter estimates for performance and resistance to parasites in sheep. Currently great variability is seen for published correlations between performance and resistance, with estimates appearing to vary with production environment. The model accounted for host genotype and parasitism in a population of lambs, incorporating heritable between-lamb variation in host-parasite interactions, with genetic independence of input growth and immunological variables. An epidemiological module was linked to the host-parasite interaction module via food intake (FI) to create a grazing scenario. The model was run for a population of lambs growing from 2 mo of age, grazing on pasture initially contaminated with 0, 1,000, 3,000, or 5,000 larvae/kg DM, and given either no anthelmintic treatment or drenched at 30-d intervals. The mean population values for FI and empty BW (EBW) decreased with increasing levels of initial larval contamination (IL(0)), with non-drenched lambs having a greater reduction than drenched ones. For non-drenched lambs the maximum mean population values for worm burden (WB) and fecal egg count (FEC) increased and occurred earlier for increasing IL(0), with values being similar for all IL(0) at the end of the simulation. Drenching was predicted to suppress WB and FEC, and cause reduced pasture contamination. The heritability of EBW for non-drenched lambs was predicted to be initially high (0.55) and decreased over time with increasing IL(0), whereas drenched lambs remained high throughout. The heritability of WB and FEC for all lambs was initially low (~0.05) and increased with time to ~0.25, with increasing IL(0) leading to this value being reached at faster rates. The genetic correlation between EBW and FEC was initially ~-0.3. As time progressed the correlation tended towards 0, before becoming negative by the end of the simulation for non-drenched lambs, with increasing IL(0) leading to increasingly negative correlations. For drenched lambs, the correlation remained close to 0. This study highlights the impact of IL(0) and anthelmintic treatment on genetic parameters for resistance. Along with factors affecting performance penalties due to parasitism and time of reporting, the results give plausible causes for variation in genetic parameter estimates previously reported.  相似文献

Predicting the impacts of selection for decreased faecal egg count (FEC) (i.e. host resistance) in grazing ruminants is difficult, due to complex interactions between parasite epidemiology, management and host responses. A mathematical model including heritable between lamb variation in host-parasite interactions, Teladorsagia circumcincta epidemiology and anthelmintic drenching, was developed and used to (i) address such interactions and their impact on outcomes including FEC, live weight (LW, kg) and pasture larval contamination (PC, larvae/kg DM), and (ii) investigate how grazing management strategies, aimed at reducing host exposure to infective larvae via pasture moves at 40 day intervals, affect these outcomes. A population of 10,000 lambs was simulated and resultant FEC predictions used to assign the 1,000 lambs with the highest and lowest predicted FEC to 'susceptible' (S) and 'resistant', (R) groups, respectively. The predicted average FEC of the S group was ~8.5-fold higher than the R group across a grazing season. The R and S groups were then simulated to graze separate pastures (R(sep) and S(sep)); and repeated for 3 grazing seasons to allow predictions to diverge and stabilize. Further, different grazing strategies were superimposed on all groups. PC and average FEC were affected by whether lambs of different resistance genotype grazed together or separately, with differences increasing across grazing seasons. By the third grazing season the average PC of the R(sep) group was reduced by ~83%, and the S(sep) group was increased by ~240%, in comparison to the whole population average. Average FEC of the R(sep) group was reduced by ~40%, and the S(sep) group increased by ~46% in comparison to the R and S groups, respectively, whilst drenching had little impact on the proportional differences in FEC between groups. Predicted LW was similar for the R and R(sep) groups irrespective of anthelmintic treatment, whilst LW of the S(sep) group was reduced by ~14% compared to the S group for un-drenched lambs, and by ~4% for drenched lambs. The differing grazing strategies were predicted to have little impact on FEC or LW, with the exception of the S(sep) group which was predicted to have a 2kg increase in LW when drenched and moved to a clean pasture. Together, these results suggest that host genotype has a substantial impact on parasite epidemiology, however the benefits of anthelmintic treatment and grazing management should only be expected for susceptible animals. This supports the use of targeted selective treatment, focussing on susceptible animals.  相似文献

The objective of the present study was to evaluate the quality of bovine embryos cryopreserved in different years in Chiapas, Mexico. The embryos were obtained from a government institution (FIMEGEN) dedicated to promoting embryo transfer among dual-purpose cattle farmers. Forty-three embryos frozen in 1988, 1989, 2000 and 2002 were analysed with the Tunel technique to detect programmed cell death (apoptosis). Eleven fresh embryos were used as controls. Analysis of variance was used in embryos stored in the different years with averages tested using Tukey's test. Student's t-test was employed to compare fresh and frozen cells. Embryos with shorter storage time presented a lower number (p < 0.001) of Tunel-positive cells compared with embryos stored for longer time. On the contrary, when comparing the number of apoptotic cells between frozen and fresh embryos a higher number of positive cells (p < 0.05) were found in the former. The present results suggest that the cryopreservation per se caused damage that compromises the viability of the embryo. Another explanation for the lower pregnancy rate found in the tropics could be irreversible damage caused by poor storage technique in these large operations.  相似文献

Severe treading damage to soils often occurs when cattle and sheep graze standing forage crops during winter. Soil recovery is a long process that may take several months if not years. Noninversion tillage can speed up the recovery process by improving drainage and air diffusion. This research assessed the ongoing benefit of noninversion tillage for improving soil structure relative to non‐tillage. This assessment was made following a land‐use transition from winter forage cropping to re‐establishment of seasonal pasture that was rotationally grazed by cattle or sheep. Prior to commencement of this study, the research site had poor soil structure due to four consecutive years of cattle and sheep grazing of winter forage crops [macroporosity (0–100 mm) <0.075 and 0.113 m3/m3 under cattle and sheep, respectively]. Tillage was effective in increasing soil macroporosity to ca. 0.175 m3/m3 under both grazing classes, which was significantly higher than nontilled soils (ca. 0.140 m3/m3, 0–100 mm depth). Improvements gained from tillage generally did not persist longer than 18 months. Average annual pasture production in tilled plots was 22.1 and 20.9 tons of dry matter per hectare (t DM/ha) for respective cattle‐ and sheep‐grazed plots, while in the nontilled plots, it was 19.1 and 18.6 t DM/ha, respectively. Results indicate noninversion tillage can provide an immediate increase in the porosity of compacted soils and improve pasture growth. However, processes involved in the formation of resilient soil aggregates are curtailed if subsequent grazing events coincide with high moisture content that causes recompaction.  相似文献

Infectious diseases pose an important extinction risk for a number of endangered carnivore populations. Wild canids are particularly susceptible to generalist pathogens transmitted from domestic dogs - particularly rabies virus and canine distemper virus (CDV). Here we present a review of rabies as a threat to Ethiopian wolves, an endangered canid restricted to a few isolated Afro-alpine ranges in the Ethiopian highlands, and the impact of rabies on the largest population in the Bale Mountains. In the Bale Mountains, two confirmed rabies outbreaks among Ethiopian wolves in 1991-1992 and 2003-2004 were each responsible for over 70% mortality among focal packs and a third rabies outbreak was suspected in 1990. This review also presents an integrated disease management strategy for Ethiopian wolves combining long-term population monitoring, disease surveillance, conventional and emergency vaccination programs, and advanced modelling techniques.  相似文献

Automated handlining (jig fishing) has recently been trialled in waters around Shetland, UK. Although different types and colours of artificial lures are commercially available there is a paucity of information available on the effects of either lure design or lure colour on resulting catches. We compared the catch rates of (1) five colours of rubber eel tube lures; and (2) five different designs of artificial lure commonly used in handline fisheries. Lure colour did not significantly affect catches of pollack (Pollachius pollachius), saithe (Pollachius virens), cod (Gadus morhua), ling (Molva molva) or tusk (Brosme brosme). Cod showed a preference for lower hooks on the six hooks per reel set-up when lures were black and pollack showed a preference for lower hooks when the lures were blue. There were no significant differences in catches of saithe, cod, ling or tusk between the five lure types. However, catches of pollack on a novel ‘sonar’ lure were lower than on other lures. No evidence of a preference for any hook position on the reels was found with any of the lure types.  相似文献
