Quantifying the effects of watershed improvement efforts is critical to agencies responsible for protecting water resources of the semiarid western United States. A complex water quality data set collected from 1994 to 2004 of upper Muddy Creek Basin was subjected to cluster analysis, discriminant analysis, and canonical correlation analysis to improve understanding of basin fluvial processes and to investigate whether livestock grazing best-management practices (BMPs) improved the water quality of the watershed. Hierarchical agglomerative cluster analysis grouped nine sampling sites into two clusters based on similarity of biological indices, separating the clusters into aquatic communities more and less tolerant of degraded stream conditions. Discriminant analysis yielded strong spatial and temporal distinctions, providing important data reduction by rendering seven key parameters (total dissolved solids [TDS], temperature, elevation, slope, 10-dominant taxa, percent collector-gatherers, and percent Plecoptera) for the spatial variation and four parameters (TDS, dissolved oxygen, total taxa, and community tolerance quotient) for the temporal variation. Canonical correlation analysis identified strong negative relationships among Plecoptera taxa and total taxa with TDS and turbidity in addition to strong positive associations with elevation, slope, and channel substrate weighted embeddedness value. Despite the onset of severe drought midway through the study period, overall reductions of 13% for TDS and a 30% increase in macroinvertebrate total taxa occurred across years, strongly suggesting that improvements in water quality were correlated to BMPs that stabilized stream channels and improved the condition of riparian areas. 相似文献
Apoptosis is necessary for both initiation and control of spermatogenesis; however, an increase in apoptosis can lead to subfertility/infertility in stallions, causing substantial financial loss in the equine industry. The ability of stem cell factor (SCF), leukemia-inhibiting factor (LIF), granulocyte–macrophage colony-stimulating factor (GM-CSF), and estradiol (E2), alone or in combination, to prevent apoptosis of germ cells in short-term equine testicular cultures was examined. Testicular tissue was sectioned into approximately 2-mm cubes and placed in media-filled culture chambers. Concentrations of SCF (100 ng/mL), LIF (10 ng/mL), GM-CSF (5 ng/mL), and E2 (10−9 mol/l) were added alone or in combination to each well. After 6 hours in culture, the tissue was fixed and immunohistochemically (terminal deoxynucleotidyl transferase-mediated nick-end labeling; TUNEL) stained for apoptosis detection. Apoptotic cells per 100 Sertoli cell nuclei within seminiferous tubules were counted until the 500th Sertoli cell nuclei was reached. This counting procedure was used for each slide. An analysis of variance (ANOVA) with a Tukey's test was used to compare apoptotic rates. In comparison with the control, GM-CSF alone lowered apoptosis by 34.77%. GM-CSF–treated tissue combined with SCF and LIF as well as GM-CSF combined with SCF, LIF, and E2 reduced apoptosis when compared with the control (37.45% and 44.40%, respectively) or other treatment combinations. GM-CSF alone reduced apoptosis; results suggest possible synergy for the combinations of SCF and LIF with GM-CSF and for E2 with SCF, LIF, and GM-CSF. 相似文献
Infectious salmon anemia (ISA) is a viral disease occurring in farmed Atlantic salmon (Salmo salar) that is characterized by lethargy, anorexia, anemia and death. To control the disease in New Brunswick, Canada, 7.5 million fish from outbreak cages have been destroyed since 1997. Despite changes made by farmers, 2002 was the worst year ever for ISA losses in the region.
We evaluated the associations between potential risk factors and ISA outbreaks in the Atlantic-salmon sites in New Brunswick. This was a multilevel study in which the site-level design was a retrospective cohort study while the cage-level design was a modified case-cohort study. The questionnaire was divided into site-level questions, cage-level questions and hatchery information.
The important factors identified by this study can be categorized as environmental, farmer controlled or industry controlled according to the capacity to change or eliminate them. Environmental risk factors such as increasing the depth of the net (if nets were ≤9 m, odds ratio (OR) = 3.34) and decreasing the depth of water underneath the net (if depth of water underneath the net >3 m, OR = 3.34) are for the most part dictated by site location. Wild pollock (Pollachius virens) in the cage reflects the number of wild pollock that live in the site location. If there were ≥1000 pollock in the cage, the odds of disease in the cage increased 4.43-fold. Risk factors that are under farm control include increasing the number of times that the salmon are treated for sea lice (OR = 3.31 if lice treatments are ≤2 times), transferring small smolts into seawater (OR = 2.40 if smolts weighed >99 g) and improving on the adaptation of smolts to seawater to reduce post-transfer mortalities (OR = 4.52 if there was at least one cage with post-transfer mortalities >5%). The industry-controlled factors need to be addressed by the industry as a whole. Organizing boat travel to minimize the time and frequency of boats travelling to or by sites currently is being reviewed. This will be extremely important because the OR = 9.43 if processing boats travel within 1 km of the site and the OR = 4.03 if the site has dry feed delivered by the feed company. Because the hazard ratio increased stepwise from 1 if the nearest neighbor with ISA was ≥5 km up to 5.5 if the nearest site with ISA was within 0.5 km, increasing the distance between sites might be necessary for effective control. 相似文献
The use of overhead trellis systems for the production of dry-on-vine (DOV) raisins and table grapes in California is expanding. Studies were conducted from 2006 to 2009 using Thompson Seedless grapevines grown in a weighing lysimeter trained to an overhead arbor trellis and farmed as DOV raisins for the first two years and for use as table grapes thereafter. Maximum canopy coverage for the two lysimeter vines across years was in excess of 80 %. Seasonal (15 March–31 October) evapotranspiration for the lysimeter vines (ETLys) was 952 mm in 2007 (farmed as DOV raisins) and 943 and 952 mm (when farmed as table grapes). The maximum crop coefficient (KcLys) across all 4 years ranged from 1.3 to 1.4. These maximum values were similar to those estimated using the relationship where Kc is a function of the amount of shaded area measured beneath the canopy at solar noon (Kc = 0.017 × percent shaded area). Covering the lysimeter’s soil surface with plastic (and then removing it) numerous times during the 2009 growing season (1 June–14 September) reduced ETLys from an average of 6.4 to 5.6 mm day?1 and the Kc from 1.07 to 0.93. A seasonal basal Kc (Kcb) was calculated for grapevines using an overhead trellis system with a 13 % reduction in the KcLys across the growing season. 相似文献
The expansion of permanent trickle irrigation systems in Sao Paulo (Brazil) citrus has changed the focus of irrigation scheduling
from determining irrigation timing to quantifying irrigation amounts. The water requirements of citrus orchards are difficult
to estimate, since they are influenced by heterogeneous factors such as age, planting density and irrigation system. In this
study, we estimated the water requirements of young ‘Tahiti’ lime orchards, considering the independent contributions from
soil evaporation and crop transpiration by splitting the crop coefficient (Kc = ETc/ETo) into two separate coefficients; Ke,
a soil evaporation coefficient and Kcb, a crop transpiration coefficient. Hence, the water requirement in young ‘Tahiti’ lime
(ETy) is ETy= (Ke + Kcb) · ETo, where ETo is the reference crop evapotranspiration. Mature tree water requirement (ETm) is ETm = Kcb · ETo, assuming no soil water evaporation. Two lysimeters were used; one was 1.6 m in diameter and 0.7 m deep, and
the other was 2.7 m in diameter and 0.8-m deep. The first one was used to calculate evaporation and the second one was used
for transpiration. ETo was estimated by the Penman–Monteith method (FAO-56). The measurements were conducted during a period
between August 2002 and April 2005 in Piracicaba, Sao Paulo state, Brazil. The lysimeters were installed at the center of
a 1.0-ha plot planted with ‘Tahiti’ lime trees grafted on ‘Swingle’ citrumelo rootstock. The trees were 1-year old at planting,
spaced 7 × 4 m, and were irrigated by a drip irrigation system. During the study period, Kc varied between 0.6 and 1.22, and
Kcb varied between 0.4 and 1.0. The results suggested that for young lime trees, the volume of water per tree calculated by
Ke + Kcb is about 80% higher than the volume calculated using Kc. For mature trees, the volume of water per tree calculated
using just Kcb can be 10% less than using Kc. The independent influence of soil evaporation and transpiration is important to better understand the water consumption of
young lime trees during growth compared to mature lime trees. 相似文献
In vegetated filter strips (VFS) the presence of perennial vegetation, rhizodeposition of labile organic substrates and the accumulation of an organic residue thatch layer may enhance microbial numbers and activity, thereby increasing the potential for mineralisation of herbicides and herbicide metabolites retained during run-off events. The objective of this laboratory experiment was to compare the mineralisation of atrazine and metolachlor with that of their respective metabolites in VFS and cultivated soil. With the exception of total bacteria, propagule density of the microbial groups, endogenous soil enzymes and microbial diversity were higher in the VFS soil. This correlated with increased mineralisation of metolachlor and its metabolites in the VFS soil and indicates potential for VFS to curtail the subsequent transport of these compounds. In contrast, the mineralisation of atrazine and the majority of its metabolites was substantially reduced in VFS soil relative to cultivated soil. Consequently, the potential for subsequent transport of atrazine and many of its metabolites may be greater in VFS soil than in cultivated soil if reduced mineralisation is not offset by increased sorption in the VFS. 相似文献
Microspectrophotometry was performed on intact, pigmented vesicle-like inclusions within living sorghum cells that were accumulating phytoalexins as a response to attempted fungal infection. The results indicate that the deoxyanthocyanidin phytoalexins are present in inclusions. Moreover, the phytoalexin concentration within a single inclusion, based on luteolinidin, was calculated to be 0·15 m. The amounts of luteolinidin and apigeninidin in cells involved in the phytoalexin response at individual infection sites were also determined. The data showed that luteolinidin accumulated to levels of 0·48–1·20 ng/cell whereas apigeninidin accumulated to levels of 0·24–0·91 ng/cell. The results of both analyses confirmed that at the infection site the deoxyanthocyanidins accumulate to levels in substantial excess of those required for inhibition of the fungus Colletotrichum graminicola. 相似文献
