REASONS FOR PERFORMING STUDY: Lameness has often been suggested to result in altered movement of the back, but there are no detailed studies describing such a relationship in quantitative terms. OBJECTIVES: To quantify the effect of induced subtle forelimb lameness on thoracolumbar kinematics in the horse. METHODS: Kinematics of 6 riding horses was measured at walk and at trot on a treadmill before and after the induction of reversible forelimb lameness grade 2 (AAEP scale 1-5). Ground reaction forces (GRF) for individual limbs were calculated from kinematics. RESULTS: The horses significantly unloaded the painful limb by 11.5% at trot, while unloading at walk was not significant. The overall flexion-extension range of back motion decreased on average by 0.2 degrees at walk and increased by 3.3 degrees at trot (P<0.05). Changes in angular motion patterns of vertebral joints were noted only at trot, with an increase in flexion of 0.9 degrees at T10 (i.e. angle between T6, T10 and T13) during the stance phase of the sound diagonal and an increase in extension of the thoracolumbar area during stance of the lame diagonal (0.7degrees at T13, 0.8 degres at T17, 0.5 degres at L1, 0.4 degrees at L3 and 0.3 degrees at L5) (P<0.05). Lameness further caused a lateral bending of the cranial thoracic vertebral column towards the lame side (1.3 degrees at T10 and 0.9 degrees at T13) (P<0.05) during stance of the lame diagonal. CONCLUSIONS: Both range of motion and vertebral angular motion patterns are affected by subtle forelimb lameness. At walk, the effect is minimal, at trot the horses increased the vertebral range of motion and changed the pattern of thoracolumbar motion in the sagittal and horizontal planes, presumably in an attempt to move the centre of gravity away from the lame side and reduce the force on the affected limb. POTENTIAL RELEVANCE: Subtle forelimb lameness affects thoracolumbar kinematics. Future studies should aim at elucidating whether the altered movement patterns lead to back and/or neck dysfunction in the case of chronic lameness.  相似文献

The effects of a seed tuber treatment with antagonistic isolates of fluorescentPseudomonas spp. were investigated on potato plants from 1981 to 1984. The experimental plots were located in fields in short and long rotations of potato. The short rotations are characterized by serious yield reductions which are caused by unknown microbial factors. The reductions varied from 30% in 1982 to only 3% in 1983 in the 3-year rotations. A statistically significant increase in yield (four to five months after planting) of ware potatoes varying from 9 to 20% was obtained in these plots through tuber bacterization, but only in 1981. In 1982 and 1983 initially significant improvements in shoot or tuber weight of seed potatoes were no longer detectable at ware potato harvest at the end of the growing period. Seed tuber bacterization had no effect on tuber yield in long rotations. Initial colonization of basal root parts by 53×104 colony forming units (cfu) of antibiotic-resistant mutants per gram of root (fresh) dropped significantly to 20×104 cfu per gram after three months. The bacterization effect on tuber yield depended on the development of harmful microbial activity and of introduced antagonists during the growing period. Seed tuber bacterization is more promising for seed potatoes than for ware potatoes in short rotations, the latter being harvested two months later.Samenvatting De invloed van pootgoedbehandeling met antagonistische isolaten van fluorescerendePseudomonas-soorten op de aardappelteelt, werd onderzocht in de periode van 1981 tot en met 1984. De proefvelden maakten deel uit van zowel ruime als nauwe rotaties met aardappelen. Kenmerkend voor de nauwe rotatie is, dat de opbrengst aanzienllijk gereduceerd wordt als gevolg van de aanwezigheid van nog onbekende microbiële factoren. Deze opbrengstverlaging varieerde van 30% in 1982 tot slechts 3% in 1983 in de 3-jarige rotaties. Pootgoedbacterisatie had in deze proefvelden een significante toename van de eindopbrengst (vier tot vijf maanden na pootdatum) van consumptieaardappelen tot gevolg, die varieerde van 9 tot 20%, echter allen in 1981. In 1982 en 1983 werd het effect van bacterisatie ook in de loop van de groeiperiode onderzocht. Aanvankelijk significante toenames van zowel spruit-als knolgewicht waren aan het einde van het groeiseizoen niet meer aantoonbaar. Pootgoedbacteristie bleek geen effect te hebben op aardappel in ruimte rotaties. Aanvankelijk werden de basale wortelgedeelten gekoloniseerd door antibioticum-resistente mutanten met 53×104 kolonievormende eenheden (kve) per gram wortel(vers); dit aantal liep (drie maanden na pootdatum) echter significant terug tot 20×104 kve per gram. Het effect van bacterisatie op de eindopbrengst werd bepaald door de ontwikkeling van de schadelijke microbiële activiteit en de ontwikkeling van de geïntroduceerde antagonisten tijdens het groeiseizoen. Pootgoedbacterisatie in nauwe rotaties biedt meer mogelijkheden voor de teelt van pootaardappelen dan die van consumptieaardappelen, die geruime tijd later geoogst worden.  相似文献

Fields infested with Synchytrium endobioticum can be descheduled when the soil is found free from sporangia of S. endobioticum . For direct examination, EPPO Standard PM 3/59 describes a soil extraction technique based on the use of a sieve shaker with six sieves. We compared recovery of sporangia between this (modified) method and an extraction method employed by the Dutch Plant Protection Service (PPS method). Recovery was determined using an inoculum dilution series: 125, 25, 5, 1, 0.2 or 0.04 sporangia per g soil. Extraction reagents used were chloroform and calcium chloride in the method described by EPPO, calcium chloride and zinc sulphate in the PPS method. At 125 sporangia per g soil, the mean density determined for the modified EPPO method was 228 sporangia per g soil when chloroform was used. Using calcium chloride, recovery percentage was higher for the modified EPPO method than for the PPS method (286, 136%, n.s. P  < 0.05). The advantage of the modified EPPO method was the larger soil volume to be processed; its disadvantages were use of complex equipment and noxious reagents (chloroform). Both extraction methods showed high variation in recovery between samples, making accurate estimation of sporangial densities in soil awkward.  相似文献

The mycoflora of culm bases and roots of rye was assessed in field trials, where benomyl was applied at dose rates ranging from 0.24 to 4.80 kg ha?1. Samples of culm bases were taken three times during the growing season, those of roots only at the harvest date. On culms with various symptoms from untreated plots,Pseudocercosporella herpotrichoides, Alternaria spp. andGerlachia nivalis were prevalent and on those from benomyl-treated plotsAlternaria spp. andFusarium culmorum. In later stages of growth,G. nivalis sharply declined andAlternaria spp. andF. culmorum increased. At the end of the season,Periconia macrospinosa andTyphula incarnata appeared in treated plots. In samples of roots taken more than two months after the last spray, porosporous dematiaceous species (Alternaria, Ulocladium andDendryphion), Mortierella spp. and other resistant fungi were prevalent in plots treated with 1.20 kg ha?1 or more, but not in those that received 0.24 kg ha?1, which is recommended for disease control. Lower counts were recorded only for species that are highly sensitive in vitro, e.g.Microdochium bolleyi andTrichoderma spp. Some fusaria were either not affected or tended to be slightly stimulated by the treatment. An attempt was made to attribute the incidence of these moderately sensitive fungi to the effect of the fungicide on non-pathogens.  相似文献

Tree and windbreak species considered for the Southern Sahelian Zone (SSZ) of West Africa have to be evaluated following multiple criteria such as fast establishment, shelter efficiency, production of feed for livestock, and firewood. An on-station experiment was conducted on a sandy soil in southwest Niger between 1988 and 1993 to assess the establishment, growth, nutritive, and calorific values of seven species considered for windbreaks:Andropogon gayanus, Bauhinia rufescens, Acacia holosericea, Acacia nilotica, Acacia senegal, Faidherbia albida, andAzadirachta indica. Mortality and stem diameter were monitored twice a year. From 20 months after planting (MAP) onwards, species were annually trimmed to 2 m height and to 1 m sidewards of the main stem. Fresh and dry weight of leaves, twigs, wood, and fruits were recorded. Leaves were analyzed for crude protein (CP) and metabolizable energy (ME) content. Wood was analyzed for its gross calorific value and its ash content. At 56 MAP, all species had less than 5% morality, except forA. holosericea, which had a mortality rate of 15.9%.A. indica andA. holosericea showed the highest stem diameter among species with 12.2 and 11.8 cm, respectively, at 56 MAP.A. senegal had the highest dry matter production with 22.3 t ha–1 at 56 MAP, followed byA. indica (12.0 t ha–1),A. holosericea (11.7 t ha–1) andB. rufescens (11.2 t ha–1), butA. holosericea was most productive at earlier harvests. OnlyA. senegal (6.4 t ha–1) andA. indica (5.1 t ha–1) had a significant wood production, whereasA. holosericea (5.7 t ha–1) had the highest phyllodes production. Leaves ofA. senegal andA. indica had highest CP contents of 258 and 214 g kg–1, respectively. The leaves of all species exceptA. gayanus had a higher CP:ME ratio than natural pasture in the region. The calorific values of firewood did not differ significantly among the species.A. nilotica, the species with one of the lowest firewood production, had the highest Fuelwood Value Index of 6.6. The choice of species for planting trees and windbreaks in the SSZ must be oriented along these criteria. Further research should be directed towards cost-benefit analyses, land tenure and property rights in combination with surveys on local knowledge of rural people.  相似文献

Oocyte maturation depends on the metabolic activity of cumulus–oocyte complex (COC) that performs nutritive and regulatory functions during this process. In this work, the enzymes [phosphofructokinase (PFK) and malate dehydrogenase (MDH)] were tested to elucidate the metabolic profile of porcine COCs during the in vitro maturation (IVM). Enzymatic activity was expressed in U/COC and U/mg protein (specific activity) as mean ± SEM. In vitro maturation was performed with 2‐oxoglutarate (5, 10 and 20 mm ) or hydroxymalonate (30, 60 and 100 mm ) inhibitors of PFK and MDH, respectively. The PFK and MDH activities (U) remained constant during maturation. For PFK, the U were (2.48 ± 0.23) 10?5 and (2.54 ± 0.32) 10?5, and for MDH, the U were (4.72 ± 0.42) 10?5 and (4.38 ± 0.25) 10?5 for immature and in vitro matured COCs, respectively. The specific activities were significantly lower after IVM, for PFK (4.29 ± 0.48) 10?3 and (0.94 ± 0.12) 10?3, and for MDH (9.08 ± 0.93) 10?3 and (1.89 ± 0.10) 10?3 for immature and in vitro matured COCs, respectively. In vitro maturation percentages and enzymatic activity diminished with 20 mm 2‐oxoglutarate or 60 mm hydroxymalonate (p < 0.05). Viability was not affected by any concentration of the inhibitors evaluated. The U remained unchanged during IVM; however, the increase in the total protein content per COC provoked a decrease in the specific activity of both enzymes. Phosphofructokinase and MDH necessary for oocyte IVM would be already present in the immature oocyte. The presence of inhibitors of these enzymes impairs the meiotic maturation. Therefore, the participation of these enzymes in the energy metabolism of the porcine oocyte during IVM is confirmed in this study.  相似文献

Carbadox is known to induce toxic effects on the adrenal cortex, resulting in hypoaldosteronism. To study the involvement of carbadox on the renin-angiotensin system, weaned piglets of five weeks old received feed supplemented with 0 (control group), 50, 100, 150 or 200 ppm carbadox. After four weeks the 100 and 150 ppm groups had significantly higher plasma renin activity levels than the control group and after nine weeks plasma renin activity levels of all treated groups were significantly higher than the control group. Five and 10 weeks after carbadox administration, three and two pigs, respectively, of all groups were necropsied and the kidneys were screened for immunohistochemically demonstrated renin. All dosed pigs demonstrated an increase of immunoreactive renin, which was dose- and time-related. From these results it is concluded that carbadox induces activation of the renin-angiotensin system, secondary to the suppressing effect on mineralocorticoid secretion and that these changes may be responsible for part of the clinical picture.  相似文献

The degree of piliation of 29 haemolytic and 4 non-haemolytic Australian strains of Moraxella bovis representing 7 different pilus antigen groups was determined. The infectivity and virulence for the eye was measured in steroid-treated mice and in cattle. Non-piliated strains failed to infect the murine eye. Most moderately or heavily piliated strains reproducibly produced the highest infectivity and virulence scores in mice when compared with lightly or very lightly piliated strains (p less than 0.05). Non-haemolytic, piliated strains were infective and in one instance virulent for mice. Almost similar levels of infectivity and virulence were observed for 7 representative haemolytic strains tested in both cattle and mice. The relative molecular weight of pilin sub-units was compared using sodium dodecyl-sulphate polyacrylamide gel electrophoresis. Three classes of pili, alpha, beta and gamma of ascending sub-unit size were identified among the 7 pilus antigen serogroups. Pilin sub-unit size bore no relationship to the degree of piliation but most strains that were highly virulent in mice and cattle expressed alpha and gamma sub-units. Some strains appeared capable of switching from alpha to beta or form beta to gamma sub-unit production.  相似文献

The purpose of this study was to estimate the expression and relative amounts of estrogen (ER) and progesterone receptors (PR) and their isoforms as well as heat shock protein 70 (HSP70) in ovaries of rats with induced cystic ovarian disease (COD). Primary, secondary, tertiary, atretic and cystic follicles were evaluated by immunohistochemistry and total ovarian proteins were analyzed by Western blot. In the granulosa layer, growing and cystic follicles in the treated group have a higher expression of ERα than growing follicles of control individuals. In the theca interna layer, tertiary follicles presented a significantly higher expression of ERα in the treated group. An increase in total ERα protein was detected in the treated group. Granulosa cells of all growing, atretic and cystic follicles show a lower expression of ERβ in animals with COD, and the total protein expression of ERβ was lower in this group. The expression of PR was lower in the granulosa cell layer of tertiary and cystic follicles in treated animals, and theca interna layer had less intense immunostaining in this group. Although there were no differences in the expression of PR-B by Western blotting, the expression of PR-A was higher and the expression of PR-C was smaller in the treated group. An intense HSP70 immunostaining was observed in the cells of cystic follicles. By Western blotting, higher protein expression of HSP70 was detected in the ovarian samples of the control group than those of the treated ones. Ovaries of animals with COD exhibited an altered steroid receptor expression and subtype balance as compared with control animals, and an increase in HSP70 immunoexpression.  相似文献

Allometric equations are routinely used in the estimation of biomass stocks for carbon accounting within forest ecosystems. However, generic equations may not reflect the growth trajectories of afforestation species that are introduced to degraded farmland characterized by water and nutrient limitations. Using sequential measurements of the height, basal diameter, and above- and belowground biomass of juvenile trees, we developed allometric equations for five woody species (Moringa oleifera Lam., Leucaena leucocephala Lam., Jatropha curcas L., Anacardium occidentale L. and Parkia biglobosa Jacq.) subjected to a gradient of water and nutrient availability in an afforestation trial on degraded cropland in the semi-arid zone of Benin, West Africa. For three of the species studied, the allometric relationships between basal diameter and biomass components (i.e. leaves, stems and roots) were described best by a simple power-law model (R2 > 0.93). The incorporation of species-specific height–diameter relationships and total height as additional predictors in the power-law function also produced reasonable estimates of biomass. Fifteen months after planting, roots accounted for 10–58% of the total biomass while the root-to-shoot ratio ranged between 0.16 and 0.73. The total biomass of the saplings ranged between 1.4 and 10.3 Mg ha?1, yielding 0.6–4.3 Mg C ha?1, far exceeding the biomass in the traditional fallow system. The rate of stem carbon sequestration measured ca. 0.62 Mg C ha?1 year?1. Overall, the allometric equations developed in this study are generally useful for assessing the standing shoot and root biomass of the five afforestation species during the juvenile growth stage and can help in reporting and verifying carbon stocks in young forests.  相似文献
