The durability of Akoya pearls is closely related to the mechanical strength of the nacre. Therefore, obtaining information about the mechanical strength of the nacre is very important in the quality control of pearls. The nacre of Akoya pearls is composed of aragonite contained within an organic matrix. The presence of the organic matrix provides mechanical strength and fracture toughness much higher than that of monolithic aragonite. In this study, we investigated the hardness and delamination strength of Akoya cultured pearls classified into four levels by “Gray Value (GV)”, a score given by a non-destructive inspection method based on UV fluorescence intensity that provides an indication of the condition of the nacre’s organic components. Pearl hardness decreased significantly with decreasing GV. Furthermore, delamination strength of the pearl surface layer was very low when the GV was very low. These results suggest that the GV indirectly represents the mechanical strength of the nacre and could be used as a non-destructive quality-control method to maintain the quality of pearls used as jewelry.

A total of 568 normal feces from calves on a beef farm in Fukui Prefecture, Japan, in 2011–2012 were examined by RT-semi-nested PCR for rotavirus A (RVA) VP4 genes. Through partial sequencing and BLAST analyses of 84 VP4-positive specimens, we identified an avian-like RVA strain, N2342, which shares highest nucleotide identity (80.0%) with known avian-like bovine strain 993/83, in one specimen. Phylogenetic analysis also revealed a close genetic relationship between N2342 and avian RVAs, suggesting bird-to-cattle transmission. We observed frequent contact of wild birds with calves in the farm, suggesting that these birds were the source of the virus.  相似文献

We exposed Akoya cultured pearls separately to heat (60–120 °C) and artificial light to investigate changes to fluorescence in the visible range and yellowing. We found that for both heat-treated and light-treated pearls, the fluorescence peak shifted from 480 to 430 nm with an increase in fluorescence intensity. This change in intensity was more prominent in heat-treated pearls, with the initial speed of increase rising with treatment temperature; treatment at 100 °C caused the greatest increase in fluorescence intensity. However, aminoguanidine suppressed the heat-induced change in the fluorescence of an ethylenediaminetetraacetic acid–soluble nacreous layer matrix. These results suggest that the heated-induced changes in the fluorescence of Akoya cultured pearls were caused largely by a buildup of fluorescent advanced glycation end products through the Maillard reaction. Although heat treatment led to a large increase in fluorescence intensity of the peak at approximately 430 nm in a deoxygenized environment, hardly any change in fluorescence intensity was observed after light treatment in this environment. Moreover, a new shoulder peak appeared at about 460 nm after light treatment. These results suggest that the Maillard reaction was not a major factor in the light-induced changes in the fluorescence of Akoya cultured pearls.  相似文献

The aim of this research was to characterize immune-related antibacterial substances from pearl oyster Pinctada fucata induced by bacterial invasion. Bacteria inoculation was performed by injecting 0.1 ml of 1.0 × 1012 colony-forming units/ml Vibrio parahaemolyticus into adductor muscle. Acidic extracts were prepared with 0.1% trifluoroacetic acid from different tissues after 8 h of injection, and antibacterial activity against V. parahaemolyticus was determined via the microdilution broth method. The acidic extracts from gills of inoculated oysters (AEg) showed stronger antibacterial activity than those from non-inoculated ones. Based on this result, antibacterial proteins were purified from AEg via two-step gel filtration chromatography, followed by high-performance liquid chromatography using a TSkgel G3000 column. Protein components were analyzed by both sodium dodecyl sulfate and native polyacrylamide gel electrophoresis. As a result, two antibacterial proteins, APg-1 (with a molecular mass of approximately 210 kDa) and APg-2 (of approximately 30 kDa), were obtained from AEg. Matrix-assisted laser desorption/ionization time of flight mass spectrometry analysis and partial amino acid sequences revealed that these proteins might be novel antibacterial proteins. These results indicate that antibacterial proteins are potentially upregulated in the gill of pearl oysters or released therefrom for defense against bacterial invasion.  相似文献

Electrospun atactic polypropylene (PP) fibers are thicker than those obtained from isotactic PP, although the viscosity of molten PPs is almost same. Thus we focused on the effect of tacticity of PP on fiber diameters. The PP samples with various tacticity were prepared by changing the blend ratio of isotactic PP and atactic PP. Melt-electrospinning is performed by using blended samples, and then electrospun fibers were observed by scanning electron microscope to evaluate fiber diameter of obtained fibers. It is clear that the diameter of electrospun PP fibers decreases as high tacticity content of PP increases. This result suggests that tacticity of samples is an important factor to control the electrospun fiber diameter.  相似文献
