The objective was to study the use of ultrasound as a complementary test in the breeding soundness evaluation in male pigs and study the pattern of echogenicity of the testicular parenchyma in boars of different racial groups. Twenty‐six adult boars from four different racial groups were used, 10 from the Piau breed (group 1), four from the commercial and finishing group (group 2), six Pietrain breed (group 3) and six from the Duroc breed (group 4). All animals were evaluated for breeding soundness evaluation and the ultrasound examination of the testicles. The groups of animals that were evaluated showed no difference in the main semen parameters that were evaluated, except for the sperm volume, concentration of the ejaculated sperm and the supravital staining; the lowest figures were for the animals from the Piau breed (group 1). In relation to the testicular biometrics, Duroc animals (group 4) had a greater scrotal width compared to the other groups. But when we assessed the intensity of pixels of the testicles, there was a difference between groups. The groups 2 (finishing animals), 3 (Pietrain) and 4 had no difference between themselves. Group 3 had greater pixel intensity in relation to group 1. Of the 26 animals studied, five showed an abnormality during ultrasound evaluation, like hydrocele, hyperechoic mass in the testicular parenchyma, cyst in the head of the epididymis and the presence of fluid in the head and tail of the epididymis. The various animal groups studied did not differ in the principal reproductive parameters evaluated, showing that despite the great variability of reproductive traits between breeds and within the same breed, the breeding soundness evaluation, the more complete it is, is essential for the selection of breeders and the ultrasonography of the reproductive system becomes an important addition in this examination.  相似文献

This study aimed to assess the effects of different cooling curves and centrifugation regimes used in cryopreservation protocols on the post‐thaw viability of Piau‐breed wild boar (Sus scrofa) sperm using in vitro assessment tests. Two centrifugations (800  g for 10 min and 2400  g for 3 min) and two cooling curves (conventional cooling using nitrogen vapour – freezing 1 and automated cooling using a programmed freezing machine – freezing 2) were tested. Therefore, the treatments were divided into M3 – centrifugation at 2400  g for 3 min and freezing 2; M10 – centrifugation at 800  g for 10 min and freezing 2; R3 – centrifugation at 2400  g for 3 min and freezing 1; and R10 – centrifugation at 800  g for 10 min and freezing 1. No significant differences (p > 0.05) between treatments occurred post‐thawing regarding the total sperm motility means recorded. The mean values of the different treatments were not different from each other regarding the supravital staining (SV), hypo‐osmotic test (HO), sperm–egg binding assay or sperm morphology. This study showed that both the cooling curve and the centrifugation regime affected the quality of post‐thaw sperm, and centrifugation for shorter times and cooling curves using automated cooling are the most suitable for minimizing sperm injury.  相似文献

The relationship between grain protein concentration and grain yield in different cultivars of winter wheat was examined in a series of field experiments carried out over three years, in which 13, 12 and 8 cultivars were studied in each year, respectively. The plants were grown at sites located in Shropshire, west-central England, in years 1 and 2, and at three other locations in eastern England in year 3. Above ground plant samples were collected at an thesis and again at maturity, when they were separated into grain and straw, and analysed for dry matter and N content. Analysis of residuals from regression of grain protein concentration on grain yield (grain protein deviation, GPD) showed that some cultivars had a higher grain protein concentration than was predicted from grain yield alone. It was deduced that the capacity to accumulate a higher grain protein concentration than predicted from grain yield is under genetic control and thus may be improved through breeding. Other factors (weight of N accumulated in the biomass at anthesis, weight of N accumulated in the biomass between anthesis and maturity and the concentration of N remaining in the straw at maturity) were added step-wise into the regression to enable statistical analysis of their relative contributions to grain protein. High GPD may be achieved through increased N accumulation after anthesis, combined with efficient re-translocation of vegetative N reserves. The use of GPD provides a selection criteria in wheat breeding programs to screen for increased grain protein concentration without a concurrent grain yield reduction. Dormancy of wheat grains, the property conferring sprouting resistance, is affected by environmental conditions experienced during grain development. We investigated the hypothesis that short dormancy duration in U.K. wheat grain (thus a high risk of post-maturity sprouting) is related to weather conditions, i.e. high temperatures during grain development. Four wheat varieties were grown at four sites ranging from the far south to the far north of the country in the years 1995–1997,ensuring different temperature and rainfall conditions during grain development. This paper focuses on one variety, Hornet, which has a high sprouting resistance rating. Other varieties gave similar results. Serial laboratory germination tests (seven days, 20°C) at 100°C-dayintervals were used to measure dormancy duration, which was assessed from logistic curves fitted to the data. During the experiment the mean temperatures during grain development differed by over4°C, due to the site × year effect. Significant effects (p>0.05) of site and year (i.e. weather) on dormancy were found, when definitions of dormancy duration of DA (number of days from anthesis to 50% germination in seven days at 20°C) or DP (number of days from physiological maturity at 45% grain moisture to 50% germination) were used. Dormancy was markedly shorter in the hot, dry year 1995 compared to the cooler, wetter years 1996 and 1997. A relationship, as postulated by Belderok, between accumulated temperature during the dough stage of grain filling and dormancy duration was not found. However, a relationship of dormancy duration to the mean temperature during grain development was found, with short dormancy periods occurring after high mean temperatures were experienced. A total of 417 pigs with an estimated average liveweight of 91 kg were transported to slaughter in three journeys, carried out in late June, each consisting of a three-hour drive followed by a one-hour holding period at the slaughter plant before they were unloaded. The pigs were accommodated on the lower two decks of a three-deck articulated lorry, each deck having five equal-sized pens; half were kept in naturally ventilated pens and half in pens with fan-assisted ventilation. The pigs' body temperature was recorded as the temperature of the blood lost at exsanguination, and as the temperature of the inner surface of the ear measured by thermal imaging. The pigs carried on the upper deck were hotter than those carried on the lower deck, and the pigs carried at the front of the vehicle were hotter than those at the back. There were small differences between the temperatures of the pigs in the different pens, but overall the temperatures of the pigs kept in pens with fan-assisted ventilation were no lower than those of the pigs kept in pens with natural ventilation. The higher temperature of the pigs transported in the front of the vehicle was associated with significantly higher serum concentrations of cortisol and activities of creatine kinase, and a tendency to higher albumin concentrations and osmolality, suggesting that they may have been exposed to more physical and psychological stress, and were possibly slightly more dehydrated, than the pigs in the rearmost pens. The type of ventilation had no effect on the blood composition of the pigs.  相似文献

To study the role of abscisic acid (ABA) and gibberellin (GA) sensitivity in regulating pre-maturity α-amylase (PMA) in wheat grains, plants were grown in a glasshouse under cold-shock and ambient conditions. α-amylase activity in response to applied ABA and GA was measured in detached-grains with the embryo removed (in vitro) and in intact-grains attached to the plant (in situ). The in vitro experiment was conducted using Spark (low PMA-susceptible genotype) and Rialto (highly PMA-susceptible genotype), with the aim of defining the time point for GA-sensitivity. The results showed an increase in GA-sensitivity at about 640 degree days after anthesis (DAA) in Rialto. There was no evidence for a change in ABA-sensitivity in either variety. The in situ experiments were conducted using genotypes from a Spark × Rialto doubled haploid population segregating for the Rht-D1a (tall) or Rht-D1b allele and for the presence or absence of 1BS/1RS. For Rht-D1a (tall) or Rht-D1b genotypes with or without 1BS/1RS, the cold-shock significantly increased GA-sensitivity, whereas there was no significant change in ABA-sensitivity. These results show PMA is related to an increase in GA-sensitivity that occurs in the aleurone at around 640 degree DAA, and can be enhanced by environmental factors (e.g. cold-shock).  相似文献

Timely onset of postpartum ovarian activity is vital for optimal reproductive performance of dairy cows. Much depends upon genetic constitution of an animal although several factors interplay to govern the onset of postpartum ovarian activity. South Asian zebu cattle have much longer service period when compared with other exotic or crossbred cattle reared in the same Asian environment, which suggests differences in their genetic makeup. However, the cows with same genetic configuration expressed better reproductive potential when reared under different environment, such as in Brazil and Mexico, which suggests the role of extrinsic factors such as management, nutrition, environment and disease conditions. Better management of animals (provision of proper shade, water and housing, efficient oestrous detection and timely insemination), good quality nutrition supplemented with appropriate minerals and vitamins, prevention of diseases (vaccination, deworming, suitable therapeutic interventions) and application of biotechnology have helped in improving postpartum ovarian activity and, therefore, reproductive performance of zebu cattle in Asia. No comprehensive study appears to have been carried out on the various aspects of reproduction in zebu cattle reared under South Asian socio-agro-climatic conditions. This paper is a modest effort to collect what ever information available and to critically review the postpartum ovarian activity in zebu cattle with special reference to the effect of the various managemental practices and pharmacological interventions.  相似文献

ObjectiveTo measure the extradural pressures in goats before and after extradural injection, and to investigate the occurrence of extradural pressure waves.Study designProspective experimental trial.AnimalsNine healthy adult goats weighing 59.4 ± 6.4 kg, scheduled for stifle arthroscopy.MethodsThe goats were pre–medicated with midazolam and anaesthesia was induced with propofol and maintained with sevoflurane. The goats were placed in lateral recumbency and extradural puncture was performed via the lumbosacral space. Correct placement of the needle was assessed by lack of resistance to the injection of saline. The needle was connected to an electronic pressure transducer to record extradural pressure. Measurements were taken before and after extradural injection of methadone (0.1 mg kg?1, diluted to a total volume of 0.2 mL kg?1) and 10 minutes later. Contrast medium was injected and correct extradural needle placement confirmed by radiography.ResultsLack of resistance to injection of saline occurred in all goats, but there were no pressure waves observed before injection in any animal. Radiography indicated incorrect needle placement in four animals and one had pressure waves synchronous with the arterial pulse after methadone injection. Correct needle placement was confirmed in the remaining five animals which exhibited pressure waves after extradural methadone injection. In the five goats with successful needle placement the baseline extradural pressure ranged from 0.4 to 2.5 kPa (3–19 mmHg), increasing to 4.4–39.9 kPa (33–300 mmHg) after injection. Ten minutes after injection, extradural pressure remained elevated and ranged from 2.5 to 17.3 kPa (19–130 mmHg).Conclusions and clinical relevanceExtradural pressure waves were not useful to confirm correct extradural needle placement in laterally recumbent goats. The presence of such waves after injection of 0.2 mL kg?1 may be indicative of correct placement but even here we saw one of nine animals with extradural pressure waves where we failed to confirm correct needle placement.
