Transplantation of ovarian tissue has high potential for female gamete conservation. However, optimal timing of oocyte recovery for in vitro maturation and fertilization is still critical. Therefore the aim of the present study was to use high-resolution transcutaneous ultrasonography to monitor follicular development within xenografted ovarian tissue. Ovarian cortex fragments (n=44) from domestic cats were transplanted into athymic nude rats (n=12). Graft development in the animals was assessed weekly by high frequency ultrasound (10-22 MHz) under two different FSH regimes. Blood collection for serum estradiol determination and vaginal smears were performed simultaneously. The xenografts were removed at different time points according to the ultrasound findings. The survival rate of the transplants 4 weeks after surgery was 54.5% and antral follicular growth was observed within 10 grafts from 5 different hosts (8.6 +/- 6.43 follicles per graft). Early follicle antrums could be detected from 0.4 mm onwards. The growth rate of the antral cavity was calculated from weekly measurements (0.56 +/- 0.44 mm per week). Although vaginal cells and estradiol levels followed a cyclic pattern, no correlation was found between follicular diameter, estradiol and keratinized vaginal cells. We recovered 5, 1 and 4 cumulus oocyte complexes from three different individuals during weeks 19, 21, and 23 respectively. Extrusion of a polar body (1 oocyte) and germinal vesicle break down (7 oocytes) indicated progression of maturation after in vitro culture. We conclude that ultrasonography und provided a reliable method to examine xenograft survival and follicular development within the grafts. Furthermore, this technique is suitable for assessment of the efficiency of hormonal treatment and narrowing of the optimal time frame for oocyte retrieval. To our knowledge, this is the first report of the in vivo development of early antral follicles in mammalian species.  相似文献

The effect of nitrogen addition on the activity of rhizosphere bacteria was studied using barley seedlings. Three different nitrogen sources were added to the soil (nitrate, ammonium and ammonium+nitrate) at four different concentrations (0, 100, 300 and 500 mg N kg−1 soil) and the plants were allowed to grow for 6 weeks. The bacterial activity was estimated by measuring thymidine and leucine incorporation into bacteria extracted using homogenisation-centrifugation. Bulk soil bacterial activity was low compared with that of rhizosphere bacteria. Nitrogen addition did not affect the activity of the bulk soil bacteria, indicating that the activity was not nitrogen limited. The thymidine and leucine incorporation rates of rhizosphere bacteria decreased when ammonium or ammonium+nitrate was applied compared with the non-amended controls. No effect on bacterial activity was found following nitrate addition. There was a significant positive correlation between rhizosphere bacterial activity and rhizosphere pH. Shoot length following ammonium treatment was significant lower than in the non-amended control, while nitrate and ammonium+nitrate addition had no effect. This indicates that the varying effects due to nitrogen sources on rhizosphere bacterial activity were not due to effects on plant growth.  相似文献

Intraluminal vesicles of multivesicular endosomes are either sorted for cargo degradation into lysosomes or secreted as exosomes into the extracellular milieu. The mechanisms underlying the sorting of membrane into the different populations of intraluminal vesicles are unknown. Here, we find that cargo is segregated into distinct subdomains on the endosomal membrane and that the transfer of exosome-associated domains into the lumen of the endosome did not depend on the function of the ESCRT (endosomal sorting complex required for transport) machinery, but required the sphingolipid ceramide. Purified exosomes were enriched in ceramide, and the release of exosomes was reduced after the inhibition of neutral sphingomyelinases. These results establish a pathway in intraendosomal membrane transport and exosome formation.  相似文献

The efficiency of utilisation of food energy by female growing minks, from weaning to adult age, was studied. The food given, pelleted according to an original technology, has the following chemical composition on a DM basis: 87.0% organic matter, 37.1% crude protein, 11.7% crude fat, 2.6% crude fiber, 35.6% nitrogen-free extractives and 13.0 per cent ash. Young minks had a feed intake, in relation to body weight, warying from 11.6 g to 58.6 g DM/d. Maximum feed intake related to kg0.75 was recorded at 700 g body weight (approximately 98 g DM/kg0.65). Digestibility of the given food expressed in DE, averaged 87.7 +/- 1.2%, while metabolizability, 82.3 +/- 1.1%. Total heat production related to the intaked gross energy, was 48.0 +/- 3.0%, and the retained energy, 34.3 +/- 4.0 per cent. The net efficiency of the metabolizable energy used for maintenance and production could not be accurately determined. However, taking to account the calculated values required for maintenance, of 649 kJ/kg0.75 in 300 to 600 g young minks, and of 607 kJ/kg0.75 in 600 to 1100 g young minks and also the maintenance efficiency, Km = 0.75, the coefficient for ME utilisation in protein and fat synthesis, of 0.50 and 0.75, respectively, it was able to determine the average ME efficiency used as net energy for maintenance and production: 70%. The highest values of nictemeral metabolism were recorded in the evening, and the lowest ones, at noon; the difference between the maximal and the minimal value did not exceed 6 per cent.  相似文献

Background

The branched chain amino acid leucine is a potent stimulator of insulin secretion. Used in combination with glucose it can increase the insulin response and the post exercise re-synthesis of glycogen in man. Decreased plasma amino acid concentrations have been reported after intravenous or per oral administration of leucine in man as well as after a single per oral dose in horses. In man, a negative correlation between the insulin response and the concentrations of isoleucine, valine and methionine have been shown but results from horses are lacking. This study aims to determine the effect of repeated per oral administration with a mixture of glucose and leucine on the free amino acid profile and the insulin response in horses after glycogen-depleting exercise.

Methods

In a crossover design, after a glycogen depleting exercise, twelve Standardbred trotters received either repeated oral boluses of glucose, 1 g/kg body weight (BW) at 0, 2 and 4 h with addition of leucine 0.1 g/kg BW at 0 and 4 h (GLU+LEU), or repeated boluses of water at 0, 2 and 4 h (CON). Blood samples for analysis of glucose, insulin and amino acid concentrations were collected prior to exercise and over a 6 h post-exercise period. A mixed model approach was used for the statistical analyses.

Results

Plasma leucine, isoleucine, valine, tyrosine and phenylalanine concentrations increased after exercise. Post-exercise serum glucose and plasma insulin response were significantly higher in the GLU+LEU treatment compared to the CON treatment. Plasma leucine concentrations increased after supplementation. During the post-exercise period isoleucine, valine and methionine concentrations decreased in both treatments but were significantly lower in the GLU+LEU treatment. There was no correlation between the insulin response and the response in plasma leucine, isoleucine, valine and methionine.

Conclusions

Repeated post-exercise administration with a mixture of leucine and glucose caused a marked insulin response and altered the plasma amino acid profile in horses in a similar manner as described in man. However, the decreases seen in plasma amino acids in horses seem to be related more to an effect of leucine and not to the insulin response as seen in man.  相似文献

Branding is the traditional and well-established method used to mark horses, but recently microchip transponders for implantation have become available. In this study, behaviour, physiological stress variables and skin temperature in foals were determined in response to hot-iron branding (n=7) and microchip implantation (n=7). Salivary cortisol concentrations increased in response to branding (1.8 ± 0.2 ng/mL) and microchip implantation (1.4 ± 0.1ng/mL), but cortisol release over time did not differ. In response to both manipulations there was a transient increase in heart rate (P<0.001) and heart rate variability (P<0.01). Branding and microchip implantation induced a comparable aversive behaviour (branding, score 3.86 ± 0.85; microchip, score 4.00 ± 0.82). Both techniques thus caused similar physiological and behavioural changes indicative of stress. Acutely, implantation of a microchip was as stressful as branding in foals. Branding caused a necrotising skin burn lasting at least 7 days. Moreover branding, but not microchip implantation (P<0.001), was accompanied by a generalized increase in skin temperature which was comparable to low degree post-burn hypermetabolism in humans.  相似文献
