Ghrelin and growth hormone secretagogues receptor (GHS‐R or ghrelin receptor) have been reported as being one of the factors of adipogenesis in adipocytes. To investigate the involvement of ghrelin and GHS‐R in adipocytes, the effect of the GHS‐R antagonist, [D‐Lys‐3]‐GHRP‐6 (His‐D‐Trp‐D‐Lys‐Trp‐D‐Phe‐Lys‐NH2), on the process of adipogenesis in ovine and rat adipocytes was evaluated. [D‐Lys‐3]‐GHRP‐6 (10?7 mol/L) significantly inhibited adipogenic differentiation of ovine and rat preadipocytes prepared from adipose tissues. The level of peroxisome proliferator activated receptor (PPAR)‐γ2 mRNA, an adipogenic marker, was decreased during the differentiation of adipocytes treated with [D‐Lys‐3]‐GHRP‐6 for 10 days. Ghrelin stimulated adipogenesis, also causing an increment of glycerol‐3‐phosphate dehydrogenase and upregulation of PPAR‐γ2. Furthermore, the antilipolytic effect of ghrelin was attenuated by treatment with [D‐Lys‐3]‐GHRP‐6 in both types of isolated adipocytes. Overall, the results of the present study highlight that GHS‐R in adipogenesis can be blocked by treatment with [D‐Lys‐3]‐GHRP‐6.  相似文献

Point-of-care (POC) devices that veterinary practitioners can use to easily and rapidly measure blood ionized calcium (iCa) levels in cows immediately after withdrawing a blood sample on the dairy farm are needed. Aims of present studies was to compare the commercially available ion-selective electrode handheld iCa meter (bovine blood iCa checker) with the benchtop blood gas analyzer GEM premier 3500 and handheld analyzer i-STAT 1. Sixty-two paired-point whole blood samples were obtained from three cows with hypocalcemia experimentally induced by Na2-EDTA infusion. Whole blood samples were also obtained from the 36 cows kept on a farm in field conditions. The results using the bovine blood iCa checker correlated with those using the GEM premier 3500 and i-STAT 1. Bovine blood iCa checker was “compatible” with the GEM premier 3500 and i-STAT 1 because the frequency of differences between the measurements within ± 20% of the mean were 100% (65/65, >75%) and 90.8% (59/65, >75%), respectively. In the field trial, the blood iCa concentration measured by the bovine blood Ca checker was significantly positively correlated with that measured by the i-STAT 1 portable analyzer. Bovine blood iCa checker was “compatible” with the i-STAT 1 because the frequency of differences between the measurements within ± 20% of the mean was 100% (36/36, >75%). Results from these findings, the bovine blood iCa checker may be applied as a simplified system to measure the iCa concentration in bovine whole blood.  相似文献

Mixed cropping is a cultivation method widely practiced in tropical regions. The newly developed close mixed planting technique mitigates the flood stress of drought-adapted upland cereal species by co-growing rice (Oryza sativa) plants under field flood conditions. We tested the hypothesis that O2 was transferred from rice to upland crops using the model system of hydroponic culture. To confirm the hypothesis, the phenomena of O2 absorption and release by plants were evaluated in a water culture condition without soil. Experiments were conducted in a climate chamber to estimate the amount of O2 released from the roots of rice and pearl millet (Pennisetum glaucum) under both O2-rich (20.0 ± .0% conc. in phase I) and O2-free dark (.8 ± .0% conc. in phase II) conditions. The total O2 change (between the two phases) in a single planting of rice and pearl millet was significantly higher than that of the mixed planting of rice and pearl millet, which indicated that O2 was transferred from rice to pearl millet under a water culture condition. The result indicated that approximately 7 μM O2 g fresh root weight?1 h?1 was transferred between the two plant species. O2 transfer was confirmed between the two plant species in a mix cultured in water, implying its contribution to the phenomenon that improved the physiological status of drought-adapted upland crops under field flood conditions.  相似文献

Most anesthetics have an immuno-suppressive effect on cellular and neurohumoral immunity, and research shows that total intravenous anesthesia (TIVA) with propofol has a greater immuno-protective effect than inhalational anesthesia in human medicine. However, in veterinary clinics, these effects remain ambiguous. To clarify the details, we focused on propofol and isoflurane, investigating clinical blood hematology and immunological profiles drawn from healthy dogs under and after two anesthesia techniques. Twelve healthy adult beagles were included in this study, randomly assigned to the propofol anesthesia group (group P: n=6) or the isoflurane anesthesia group (group I: n=6). In both groups, the number of lymphocytes in peripheral blood decreased after 2 hr of anesthesia (2 hr), but group P showed significantly less decrease than group I. For T-lymphocyte subsets examined by flowcytometry, the ratio of CD3+, CD4+ and CD8+ lymphocytes in the peripheral blood mononuclear cell (PBMC) of group P at 2 hr also exhibited a high level compared to group I. Moreover, for mRNA expression of cytokines measured by real-time PCR, the IL2 (pro-inflammatory cytokine) of group P showed no decrease like group I. The IL10 (anti-inflammatory cytokine) of group P also showed no increase like group I, while both cytokines maintained nearly the same level until 2 hr. These results suggest that, compared to propofol, isoflurane had more strongly immuno-suppression caused by anesthesia, and propofol itself might have some immuno-protective effects. Thus, TIVA with propofol might benefit immunological support in the perioperative period of dogs.  相似文献

To elucidate the effects of hydrographic conditions on the migration and distribution of diamond squid ( Thysanoteuthis rhombus ) in the Sea of Japan, two numerical experiments were conducted using a Lagrangian particle-tracking model. First, Lagrangian simulations with different horizontal swimming speeds were conducted in the area off the San-in coast, the southwest Sea of Japan. The comparison results of simulations and experimental tagging records from 2003 suggest that horizontal movement of diamond squid over a few days is influenced by individual swimming, but long-term migration over more than about 10 days is dependent on the ambient current field. Secondly, further Lagrangian simulations with biological processes, forced by different hydrographic conditions in 2003–2005, were conducted. Temporal variations in distributions and sizes calculated by the model corresponded approximately to those of catch data. A large number of particles simulating squid were distributed over the nearshore region from the San-in coast to the Noto Peninsula during September–November every year. Differences in the migration route and distribution of particles among years were closely related to differences in hydrographic conditions such as the position of the main stream of the Tsushima Warm Current. Prediction of the catch off the San-in coast might be improved by considering hydrographic conditions in the southwest Sea of Japan.  相似文献
